A New Technique for Use in Culturing Prokaryotes Comprising the Mouse Intestinal Microbiome

The microbiome has a profound impact on host fitness. pH, oxygen, nutrients, or other factors such as food or pharmaceuticals, subject the microbiome to variations in the gastrointestinal tract. This variation is a cause for concern given dysbiosis of the microbiome is correlated with various disease states. Currently, much research relies on model organisms to study microbial communities since intact microbiomes are challenging to utilize. The objective of this study is to culture an explanted colon microbiome of 4 Balb/c mice to develop an in vitro tool for future microbiome studies. We cultured homogenates of the distal colons of 4 mice in trans-well culture dishes. These dishes were incubated for 24 hours in two different oxygen concentration levels and the pH was compared before and after incubation of the cultures. To analyze the integrity of the microbiome, we utilized massively paralleled DNA sequencing with 16S metagenomics to characterize fecal and colon samples to speculate whether future studies may utilize feces in constructing an in vitro microbial community to spare animal lives. We found that pH and familial relationships had a profound impact on community structure while oxygen did not have a significant influence. The feces and the colon were similar in community profiles, which lends credence to utilizing feces in future studies. The gut microbiome is of great interest and great importance for studies in a variety of different diseases. Many laboratories do not have access to germ-free mice, which is one optimal way to study mammalian microbiomes, but this technique allowed for the in vitro culturing of a majority of the prokaryotes isolated from the colons of mice. This may allow an alternative to study the interactions of this very diverse population of microorganisms without the need for germ-free conditions.

众所周知,干旱曾经造成过几次重大的饥荒。对亚洲雨水灌溉地区的水稻产量具有重大的破坏性影响。有研究发现野生作物在恶劣环境下仍可正常的生长。野生稻在干旱、低温、盐碱等胁迫下表现出比栽培稻在生长状况方面显著的优势,有研究表明胁迫下野生稻内部及根际有些优势菌群辅助了植物抵抗逆境。鉴于此,我们在干旱胁迫下对东乡野生稻根际微生物和内生微生物进行了分离和筛选,制备成菌剂,作用于栽培稻东稻4,期望能有助于栽培稻在干旱条件下的生长和生存。研究结果如下:1.我们种植了东乡野生稻,并给予干旱胁迫,然后筛选分离抗旱的内生菌和根际菌。利用PEG 6000模拟干旱胁迫,浓度分别为30.3%(-1 MPa)和43.8%(-2 MPa),筛选出多株具有研究潜力的菌株。测定各种抗逆指标后,选择3株优良菌种,分别命名为P1、N1g和N2。通过形态学、生理生化和分子生物学鉴定手段,P1鉴定为卡贝肠杆菌Enterobacter kobei,N1g鉴定为丰盈芽孢杆菌Bacillus toyonensis,N2鉴定为阿斯氏肠杆菌Enterobacter asburiae。经过测试,发现这些菌株在促进植物生长方面也具有一定的研究潜力,如具有ACC脱氨酶活性、溶解磷酸盐(P)和钾盐(K)、产氨(NH)、产铁载体和氢氰酸(HCN)等。2.以东稻4号栽培稻为研究材料,自制菌剂采用叶面喷施,研究其对水稻抗旱性和促生长的影响。盆栽实验设置为常规栽培组CK(+)、干旱对照组CK(-)、N1g+N2处理组、P1处理组。研究结果表明,菌剂处理组与干旱对照组CK(-)相比,植株的形态、生理生化指标均有显著提高,表现在地上部分高度、地上部分和根的鲜重、叶片相对含水量、叶面积指数、光合色素、抗氧化等指标上,同时发现MDA含量显著降低。在干旱胁迫下的两组实验中,接种菌株P1和N1g+N2的处理与常规栽培组CK(+)之间的一些指标没有显著差异,表明菌剂的抗旱能力非常强。与干旱对照组CK(-)相比,P1组叶片相对含水量增加297.8%。与常规栽培组CK(+)相比栽培稻根系鲜重和干重也有显著增加,接种N1g+N2的水稻根系鲜重增加了42.9%;干重增加了34.1%。结论,N1g+N2菌株组合和P1菌株单独叶面喷施在干旱条件下都能够很好地帮助栽培稻抗逆和促生。进而表明野生稻根际微生物和内生微生物在干旱条件下用于促进植物生长发育很有前景、很有意义。

Acoustic engineers are faced with the challenge of minimising reverberation time in their designs so as to contribute to the health and well-being of those traveling by train and those on the *** the problem is easy to identify,it is not as simple to *** acoustical environment of a train tunnel is complex,with a variety of noise contributing factors such as train announcements,speech of commuters,ventilation systems,electrical equipment and wheel and rail *** a result,there is some difficulty in modeling the complete acoustic environment with computational or acoustic first *** this study,an experimental rig was constructed to model the acoustic behavior within a *** modal properties for the 300 Hz to 1500 Hz range,including resonances and mode shapes were identified and were shown to successfully correspond to theoretical results and a computational model created in COMSOL using Finite Element Analysis.

Two experiments were conducted to determine the nitrogen-corrected apparent metabolizable energy(AMEn) of differently processed meals from Juncea(Brassica juncea), yellow and black seeded canola(Brassica napus), with or without supplementation of multi-carbohydrase enzymes(Enz) in diets for broiler chickens. The first experiment was a 3×2×2 factorial arrangement with the main factors being seed type(yellow [Yellow] or black [B1] canola seeds and Juncea seeds), processed at two temperatures(high temperature desolventized-toasted [HTDT] at 95℃ or low temperature desolventized-toasted[LTDT] at 57℃), with or without Enz. In Exp. 1, a total of 384 one-day-old male broiler chicks were randomly assigned to 64 battery cages, with 6 birds/cage. The second experiment was a 2×2×2 factorial arrangement with the main factors being seed type(Yellow or black [B2]), seed source(Scott,Saskatchewan or Truro, Nova Scotia) and Enz(with or without) supplementation. A total of 264 one-dayold male broiler chicks were randomly assigned to 44 battery cages, with 6 birds per cage. In Exp, 1 and 2,all birds were fed a common starter diet from 1 to 14 days of age. From d 15 to 21, the birds were fed one of the test treatments, a basal grower diet or the basal grower diet replaced with 30% test ingredient with celite(0.8%) added as an inert marker. Excreta was collected on d 20 and 21. In Exp. 1, there were no interactions(P > 0.05) among seed type, processing temperature and Enz. Processing temperature and dietary Enz did not affect(P > 0.05) AMEn of different canola meals. The AMEn of prepress solvent extracted canola and juncea meals(PSEM) from Yellow(11.2 MJ/kg) was higher(P 0.05) among seed color,location and Enz. For decades,African economies have embarked on financial sector ***,the empirical implications of these reforms have been *** paper investigates the impact of financial development on Economic growth using time series data in *** investigation was carried out using three common indicators of financial development(broad money,deposit/GDP and domestic credit to private sector).Using the Auto Regressive Distributive Lag(ARDL)technique of estimation,it was discovered that there exist a short-run positive relationship between monetary mass(M2),government expenditure and economic growth,a short run negative relationship between bank deposits,private investment and economic growth equally *** in the long run,all indicators of financial development show a positive and significant impact on economic *** paper thus confirms the existence of a positive and long-term impact of all the indicators of financial development on economic growth through bound *** is therefore proposed that the financial reforms in Cameroon should be pushed forward in order to boost the development of the financial sector thus an increase in its role on economic growth.

Objective:Electro-acoustic stimulation (EAS) is an effective method to enhance cochlear-implant performance in individuals who have residual low-frequency acoustic *** help the majority of cochlear implant users who do not have any functional residual acoustic hearing,electro-tactile stimulation (ETS) may be used because tactile sensation has a frequency range and perceptual capabilities similar to that produced by acoustic stimulation in the EAS ***:Following up the first ETS study showing enhanced English sentence recognition in noise,1 the present study evaluated the effect of ETS on Mandarin tone recognition in noise in two groups of adult Mandarin-speaking *** first group included 11 normalhearing individuals who listened to a 4-channel,noise-vocoded,cochlear-implant *** second group included 1 unilateral cochlear-implant user and 2 bilateral users with each of their devices being tested *** groups participated in a 4-alternative,forced-choice task,in which they had to identify a tone that was presented in noise at a 0-dB signal-to-noise ratio via electric stimulation (actual or simulated cochlear implants),tactile stimulation or the combined ***:While electric or tactile stimulation alone produced similar tone recognition (～40％ correct),the ETS enhanced the cochlear-implant tone recognition by 17-18 percentage *** size of the present ETS enhancement effect was similar to that of the previously reported EAS effect on Mandarin tone *** analysis on tactile sensation showed an important role of frequency discrimination in the ETS ***:Tactile stimulation can potentially enhance Mandarin tone recognition in cochlear-implant users who do not have usable residual acoustic *** optimize this potential,high fundamental frequencies need to be transposed to a 100-200 Hz range.

The potential for dengue fever epidemic due to climate change remains uncertain in tropical *** study aims to assess the impact of climate change on dengue fever transmission in four South and Southeast Asian *** collected weekly data of dengue fever incidence,daily mean temperature and rainfall from 2012 to 2020 in Singapore,Colombo,Selangor,and Chiang *** for temperature and rainfall were drawn for three Shared Socioeconomic Pathways(SSP126,SSP245,and SSP585)*** a disease transmission model,we projected the dengue fever epidemics until 2090s and determined the changes in annual peak incidence,peak time,epidemic size,and outbreak duration.A total of 684,639 dengue fever cases were reported in the four locations between 2012 and *** projected change in dengue fever transmission would be most significant under the SSP585 *** comparison to the 2030s,the peak incidence would rise by 1.29 times in Singapore,2.25 times in Colombo,1.36 times in Selangor,and>10 times in Chiang Mai in the 2090s under ***,the peak time was projected to be earlier in Singapore,Colombo,and Selangor,but be later in Chiang Mai under the SSP585 *** in a milder emission scenario of SSP126,the epidemic size was projected to increase by 5.94%,10.81%,12.95%,and 69.60%from the 2030se2090s in Singapore,Colombo,Selangor,and Chiang Mai,*** outbreak durations in the four settings were projected to be prolonged over this century under SSP126 and SSP245,while a slight decrease is expected in 2090s under *** results indicate that climate change is expected to increase the risk of dengue fever transmission in tropical areas of South and Southeast *** greenhouse gas emissions could be crucial in reducing the transmission of dengue fever in the future.
