The Difference in Calcium Levels in Aspergillus nidulans Grown on Glucose or Pectin

Understanding the growth regulatory mechanisms in filamentous fungi is very important for the production of medicines for antifungal therapies. It is well established that Ca2+ gradient is essential for hyphal growth and that one mechanism responsible for the Ca2+ cellular concentration starts with the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) by receptor-regulated forms of phosphoinositide-specific phospholipase C (PI-PLC). In the present study the levels of calcium in Aspergillus nidulans wild type (A26) and plcA-deficient mutant (AP27) growing in a carbon source readily assimilated, as glucose or pectin a non-readily assimilated carbon source was investigated. Intracellular calcium levels in A26 were higher in the presence of glucose than in pectin, but lower in AP27 independently of the carbon source and in AP27 the vesicular calcium distribution occurred mainly at the apex of the hyphae. Delay in nuclear division was also observed if A26 and AP27 were grown in pectin presence when compared with growth in glucose. For the first time, it is demonstrated that the levels of intracellular Ca2+ were higher when A. nidulans was growing in glucose than in a non readily assimilated carbon source as pectin. Further, it also showed that the plcA gene, although not essential, may be responsible for high-molecular weight carbon source recongnation, for the intracellular Ca2+ levels maintenance and consequently by the nuclear division in A. nidulans.

Patient with Churg Strauss Syndrome and Myocarditis Treated with Cyclophosphamide

Cardiac involvement in Churg Strauss Syndrome is common and a poor prognostic indicator. Myocarditis in Churg Strauss Syndrome can present in different ways. It has been shown that basic cardiac investigations including echocardiography can be normal even in symptomatic patients. More recently cardiac magnetic resonance imaging (MRI) has been shown to be more sensitive in its diagnosis. Our case report describes a 45 year old male who presented with palpitations and breathlessness. Echocardiography was normal but cardiac MRI demonstrated abnormalities consistent with Myocarditis. He was treated with Cyclophosphamide and follow up MRI imaging demonstrated complete resolution of these abnormalities which was accompanied by resolution of symptoms. This case therefore supports the use of cardiac MRI in Churg Strauss Syndrome as a sensitive diagnostic tool in and as a means of monitoring response to therapy. It also supports the therapeutic effectiveness of Cyclophosphamide therapy in Churg Strauss related Myocarditis, something that has yet to be assessed on a large scale.

Long-Term and Immediate Impacts of Health Emergency and Disaster Risk Management(Health-EDRM) Education Interventions in a Rural Chinese Earthquake-Prone Transitional Village

Ma'an Qiao Village, a Dai and Yi ethnic minority-based community in Sichuan Province, China sustained complete infrastructure devastation during the 2008 Panzhihua earthquake. Health emergency and disaster risk management(Health-EDRM) education intervention programs were implemented in 2010 and 2011. This serial cross-sectional survey study aimed to examine the immediate and long-term impacts of the Health-EDRM interventions in this remote rural community. The findings demonstrate knowledge improvement in areas of water and sanitation, food and nutrition, and disaster preparedness immediately after the Health-EDRM education interventions. Temporal stability of knowledge retention was observed in household hygiene and waste management and smoking beliefs in 2018, 7 years after the *** important findings include knowledge uptake pattern differences of oral rehydration solution(ORS) between earthquake-prone and flood-prone communities. Usage of Internet and mobile technology for accessing disaster-related information was found to be independent of gender and income. Overall, this study demonstrated the knowledge improvement through Health-EDRM education interventions in a remote rural community. Promoting behavioral changes through interventions to raise awareness has the potential to reduce health risks in transitional post-disaster settings. Future programs should aim to identify evidence-based practices and explore how technology can support Health-EDRM education among vulnerable subgroups.

近年来,藓类植物作为一类模式植物,在分子生物学、抗性生理学研究中受到重视,因此,开展逆境条件下藓类植物渗透调节物质和膜保护系统相关指标变化的研究,将为了解藓类植物对逆境条件适应的生理机制提供基础资料。多蒴灰藓(Hypnum fertile Senden.)分布广泛,生境多样,本文研究了低温(5℃)和高温(40℃和60℃)在不同胁迫时间(0、1、2、4、6、8 h)下多蒴灰藓体内可溶性糖、游离脯氨酸、丙二醛(MDA)含量和过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性的变化,以期了解多蒴灰藓对不同温度条件的适应能力和响应的生理机制。结果表明:在5℃低温处理下,随处理时间延长,多蒴灰藓的可溶性糖含量逐渐增加,在高温处理下,呈现先升高后降低的趋势;游离脯氨酸、MDA在低温和高温胁迫下随处理时间延长都逐渐升高。CAT在5℃低温处理下活性逐渐升高,在40℃高温处理下先升高后降低,在60℃高温处理下则迅速降低,6 h后酶完全丧失活性。SOD的活性在5℃低温处理下随处理时间延长逐渐降低,在高温处理下则表现出先升高后降低趋势,40℃高温胁迫下6 h达到最大值,60℃胁迫下0.5 h达到最大值。POD在5℃和40℃处理下活性随处理时间延长逐渐升高,在60℃高温处理下则先升高后降低。这些结果表明,多蒴灰藓适生于20℃左右的温度条件下,60℃高温对多蒴灰藓的伤害明显,与高温相比,多蒴灰藓更适合生长于低温环境;多蒴灰藓通过增加游离脯氨酸以提高对温度逆境的适应;POD和CAT在多蒴灰藓抵御低温胁迫时可能起着主要作用,而SOD则在其抵御高温胁迫时起一定的作用。

BACKGROUND The coronavirus disease 2019(COVID-19)pandemic has made it more challenging for patients to undergo yttrium-90(Y-90)radioembolization(RE).Same day Y-90 RE provides an opportunity to minimize logistical challenges and infection risk associated with COVID-19,thus improving patient *** To describe the use of same day Y-90 RE with routine single photon emission computed tomography/computed tomography(SPECT/CT)in order to optimize *** All patients were selected for Y-90 RE through a multidisciplinary tumor board,and were screened and tested for COVID-19 infection per institutional protocol.A same day procedure was developed,consisting of angiography,imaging,and Y-90 resin particle *** SPECT/CT after technetium-99m macroaggregated albumin(Tc-99m MAA)administration was performed for assessment of arterial supply,personalized dosimetry,and extrahepatic ***-treatment Y-90 bremsstrahlung SPECT/CT was performed for confirmation of particle delivery,by utilization of energy windowing to limit signal from previously administered Tc-99m MAA *** A total of 14 patients underwent same day Y-90 RE between March and June *** lung shunt fraction was 6.13%(range 3.5%-13.1%).Y-90 RE was performed for a single lesion in 7 patients,while the remaining 7 patients had treatment of multifocal *** largest lesion measured 8.3 *** patients tolerated the procedure well and were discharged the same *** Same day Y-90 RE with resin-based microspheres is feasible,and provides an opportunity to mitigate infection risk and logistical challenges associated with the COVID-19 pandemic and *** recommend consideration of SPECT/CT,especially among patients with complex malignancies,for the potential to improve outcomes and eligibility of patients to undergo same day Y-90 RE.
