Reconstruction of Rabbit Corneal Layer Composed of Corneal Fibroblasts and Corneal Epithelium on the Lyophilized Amniotic Membrane

Many researchers have employed the cryopreserved amniotic membrane（CAM） and corneal epithelial cells in the treatment of a severely damaged burned cornea, with corneal epithelial cells cultured on an amniotic membrane （AM）. The lyophilized amniotic membrane（LAM） has a higher graft take and a longer shelf life; it is easier to store and safer because of gamma irradiation. Two Teflon rings（ Ahn＇s supporter） were made for culturing the cells on the LAM, and were then used to support the LAM. To reconstruct a corneal layer composed of corneal fibroblasts and epithelium, the corneal fibroblasts were first cultivated on the stromal side of LAM for five days, foUowed by epithelial cells culture on the epithelial side, by using the air-liquid interface culture. The reconstructed corneal layer composed of corneal fibroblasts and corneal epithelial cells has a much healthier basal layer of corneal epithelium than the reconstructed corneal epithelium, which was got by using only corneal epithelial cells, and resembles the epithelium of normal corneas, without the horny layer. Thus, the reconstruction of the corneal layer by using a LAM is considered to be a good in vitro model, not only for its application in toxicological test kits, but also for transplantation in patients with a severely damaged cornea.

Rational design of ultrafine FeSe_(2) nanocrystals emb e dde d within hollow mesoporous carbon bowls for potassium-ion batteries with long-term cycling stability and high volumetric capacity

Hollow structures are commonly used to alleviate the mechanical stress on electrode materials and to provide more active sites in potassium-ion batteries(KIBs).Nevertheless,the excessive internal voids within these structures significantly reduce the packing density of particles,resulting in a relatively low volumetric energy density of the fabricated electrodes,which is undesirable for practical *** designed a hollow mesoporous carbon bowl embedded with ultrafine bis(selanylidene)iron(FeSe 2)nanocrystals(FeSe 2@HMCB)via a controllable impregnation method and subsequent selenization pro-cess for high-performance *** as-obtained FeSe 2@HMCB can inherit the advantages of conventional hollow carbon-based composites,such as alleviation of volume variation in active materials,abundant ion storage sites,and high electrical ***,the bowl structure has a higher pack-ing density than the conventional hollow structure,resulting in a significant increase in the volumet-ric energy density of the fabricated *** of these advantages,the FeSe 2@HMCB exhibits a high,stable reversible capacity of 326 mA h g^(-1) even after 1000 cycles at 0.5 A g^(-1),and excellent rate capacities(182 mA h g^(-1) at 3.0 A g^(-1)).Compared with the hollow structured counterpart,the vol-umetric capacity(mA h cm−3)of FeSe 2@HMCB increased by 60%.Furthermore,a full cell consisting of FeSe 2@HMCB//Prussian blue(PB)exhibits excellent electrochemical performance(99 mA h g^(-1) after 100 cycles at 0.1 A g^(-1)).

Most microfluidic separation techniques rely largely on object size as a separation *** ability to separate micro-objects based on their shape is crucial in various biomedical and chemical ***,we develop an ondemand,label-free acoustofluidic method to separate prolate ellipsoids from spherical microparticles based on traveling surface acoustic wave-induced acoustic radiation force and *** freely rotating non-spherical microobjects were aligned under the progressive acoustic field by the counterrotating radiation torque,and the major axis of the prolate ellipsoids was parallel to the progressive wave *** specific alignment of the ellipsoidal particles resulted in a reduction in the cross-sectional area perpendicular to the wave *** a consequence,the acoustic backscattering decreased,resulting in a decreased magnitude of the radiation *** the variation in radiation force,which depended on the micro-object morphology enabled the acoustofluidic shape-based *** conducted numerical simulations for the wave scattering of spherical and prolate objects to elucidate the working mechanism underlying the proposed method.A series of experiments with polystyrene microspheres,prolate ellipsoids,and peanut-shaped microparticles were performed for *** quantitative analysis of the separation efficiency,we confirmed the high purity and high recovery rate of the proposed acoustofluidic shapebased separation of *** a bioparticle,we utilize Thalassiosira eccentrica to perform shape-based separation,as the species has a variety of potential applications in drug delivery,biosensing,nanofabrication,bioencapsulation and immunoisolation.

N-terminal pro-B-type natriuretic peptide as prognostic marker for patients of non ST-segment elevation myocardial infarction

In this work, we analyzed only the patients of the NSTEMI (non ST-segment elevation myocardial infarction) who arrived at the hospital within 12 h after symptoms started. Using NSTEMI follow-up data within, the characteristics of the clinical data, the risk factor, and the blood tested in the hospital visit were analyzed for MACE (major adverse cardiac events) patients. MACE includes cardiac death, MI (myocardial infarction), Re-PCI, and CABG (coronary artery bypass graft). As a result, from the NSTEMI patients which can be followed up for over 12 m, NT-ProBNP (p=0.014) and age (p=0.045) are found to be the independent risk factors related to MACE. Accordingly, they can be useful for the diagnosis and prognosis for NSTEMI patients as a biomarker.

Structural and electrochemical stabilization enabling high-energy P3-type Cr-based layered oxide cathode for K-ion batteries

Layered-type transition metal(TM)oxides are considered as one of the most promising cathodes for K-ion batteries because of the large theoretical gravimetric capacity by low molar ***,they suffer from severe structural change by de/intercalation and diffusion of K^(+)ions with large ionic size,which results in not only much lower reversible capacity than the theoretical capacity but also poor power ***,it is important to enhance the structural stability of the layered-type TM oxides for outstanding electrochemical behaviors under the K-ion battery ***,it is investigated that the substitution of the appropriate Ti^(4+)contents enables a highly enlarged reversible capacity of P3-type KxCrO_(2) using combined studies of first-principles calculation and various *** the pristine P3-type KxCrO_(2) just exhibits the reversible capacity of∼120 mAh g^(−1) in the voltage range of 1.5-4.0 V(vs.K^(+)/K),the∼0.61 mol K^(+)corresponding to∼150 mAh g^(−1) can be reversible de/intercalated at the structure of P3-type K0.71[Cr_(0.75)Ti_(0.25)]O_(2) under the same ***,even at the high current density of 788 mA g^(−1),the specific capacity of P3-type K0.71[Cr_(0.75)Ti_(0.25)]O_(2) is∼120 mAh g^(−1),which is∼81 times larger than that of the pristine P3-type KxCrO_(2).It is believed that this research can provide an effective strategy to improve the electrochemical performances of the cathode materials suffered by severe structural change that occurred during charge/discharge under not only K-ion battery system but also other rechargeable battery systems.

Inhibition of CD82 improves colitis by increasing NLRP3 deubiquitination by BRCC3

CD82 is a transmembrane protein that is involved in cancer suppression and activates immune cells;however, information on the NLRP3 inflammasome is limited. Herein, we show that although CD82 suppressed the activation of the NLRP3 inflammasome in vivo and in vitro, CD82 deficiency decreased the severity of colitis in mice. Furthermore, two binding partners of CD82, NLRP3 and BRCC3, were identified. CD82 binding to these partners increased the degradation of NLRP3 by blocking BRCC3-dependent K63-specific deubiquitination. Previous studies have shown that CD82-specific bacteria in the colon microbiota called Bacteroides vulgatus (B. vulgatus) regulated the expression of CD82 and promoted the activation of the NLRP3 inflammasome. Accordingly, we observed that B. vulgatus administration increased mouse survival by mediating CD82 expression and activating NLRP3 in mice with colitis. Overall, this study showed that CD82 suppression reduced the pathogenesis of colitis by elevating the activation of the NLRP3 inflammasome through BRCC3-dependent K63 deubiquitination. Based on our findings, we propose that B. vulgatus is a novel therapeutic candidate for colitis.

Microstructure and compressive deformation of hypereutectic Al-Si-Fe based P/M alloys fabricated by spark plasma sintering

Al-Si-Fe based alloys are attractive light-weight structural materials for automotive engine components because of their high wear resistance, low density and low thermal expansion. Al-17Si 5Fe-2Cu-lMg-lNi-lZr alloys were produced in compact form by a spark plasma sintering （SPS） technique using gas atomized powders. The mean grain size of the compact was 530 nm, and fine equiaxed grains and uniformly distributed precipitates were observed in the compact. The compressive deformation behavior of the SPSed materials was examined at various temperatures and strain rates. All the true stress-true strain curves showed steady state flow after reaching peak stress. The peak stress decreased with increasing test temperature and decreasing strain rate. In the deformed specimens, the equiaxed grain morphology and the dislocation microstructure within the equiaxed grains were observed. These facts strongly indicated the occurrence of dynamic recrystallization during high temperature deformation of the present alloy.

Occult hepatitis B virus infection is not associated with disease progression of chronic hepatitis C virus infection

AIM To clarify the prevalence of occult hepatitis B virus(HBV) infection(OBI) and the association between OBI and liver disease progression, defined as development of liver cirrhosis or hepatocellular carcinoma(HCC), worsening of Child-Pugh class, or mortality in cases of chronic hepatitis C virus(HCV) infection. METHODS This prospective cohort study enrolled 174 patients with chronic HCV infection(chronic hepatitis, n = 83; cirrhosis, n = 47; HCC, n = 44), and evaluated disease progression during a mean follow-up of 38.7 mo. OBI was defined as HBV DNA positivity in 2 or moredifferent viral genomic regions by nested polymerase chain reaction using 4 sets of primers in the S, C, P and X open reading frame of the HBV genome. RESULTS The overall OBI prevalence in chronic HCV patients at enrollment was 18.4%, with 16.9%, 25.5% and 13.6% in the chronic hepatitis C, liver cirrhosis and HCC groups, respectively(P = 0.845). During follow-up, 52 patients showed disease progression, which was independently associated with aspartate aminotransferase > 40 IU/L, Child-Pugh score and sustained virologic response(SVR), but not with OBI positivity. In 136 patients who were not in the SVR state during the study period, OBI positivity was associated with neither disease progression, nor HCC development. CONCLUSION The prevalence of OBI in chronic HCV patients was 18.4%, and OBI was not associated with disease progression in South Koreans.

为了研究大林姬鼠两亚种(韩国的指名亚种及中国东北和内蒙古地区的东北亚种)线粒体DNA的变异程度并确定朝鲜亚种的分类地位,我们分别将来自韩国和中国东北长白山地区的两亚种的线粒体DNA的细胞色素b基因和控制区进行了测序分析。我们将测序所得到细胞色素b基因序列与来自基因库的大林姬鼠5个亚种的相应的单倍型进行了分析,结果显示大林姬鼠可分为4个类群[类群1:韩国大林姬鼠指名亚种;类群2:中国长白山和内蒙古地区的东北亚种、俄罗斯外加贝尔的majuculus亚种;类群3:中国长春的东北亚种、俄罗斯Primorye(俄罗斯远东地区)rufulus亚种、俄罗斯库页岛(俄罗斯远东地区)和日本北海道地区的giliacus亚种;类群4:中国黑龙江海林地区的东北亚种]。线粒体的控制区序列分析显示韩国指名亚种也不同于中国东北地区的东北亚种。本研究的类群1,2和3与Serizawaetal.(2002)的研究的K、S和R的分支相对应。这表明韩国指名亚种(类群1和分支K)的线粒体DNA与其他类群不同。另外,我们还发现在细胞色素b基因构建的系统树中,东北亚种可以与类群2(分支S)及类群3(分支R的不同亚种聚合在一起。我们认为线粒体DNA的母性遗传与两个相邻亚种的个体之间的种内杂交造成了基于细胞色素b序列对东北亚种的聚类分析结果与基于形态学特征的分类结果的不一致。因此,我们提出对这些显示出核苷酸序列多样性的东北亚种不能只用细胞色素b的数据进行亚种分类,还应该结合形态学和核DNA特征进行进一步分析。最后,我们还发现韩国的指名亚种的细胞色素b序列在平均距离16.93%的基础上不同于来自基因库的***。Jones and Johnson(1965)指出了韩国的大林姬鼠在形态上的区别,所以我们认为韩国的大林姬鼠指名亚种***是一种具有形态和遗传特异性的地方亚种。
