Hot water-extracted Lycium barbarum and Rehmannia glutinosa inhibit proliferation and induce apoptosis of hepatocellular carcinoma cells

AIM： To investigate the effect of hot water-extracted Lydurn barbarum （LBE） and Rehrnannia glutinosa （RGE） on cell proliferation and apoptosis in rat and/or human hepatocellular carcinoma （Hcc） cells. METHODS： Rat （H-4-Ⅱ-E） and human Hcc （HA22T/ VGH） cell lines were incubated with various concentrations （0-10 g/L） of hot water-extracted LBE and RGE. After 6-24 h incubation, cell proliferation （n = 6） was measured by a colorimetric method. The apoptotic cells （n = 6） were detected by flow cytometry. The expression of p53 protein （n = 3） was determined by SDS-PAGE and Western blotting. RESULTS： crude LBE （2-5 g/L） and RGE （2-10 g/L） dose-dependently inhibited proliferation of H-4-Ⅱ-E cells by 11% （P 〈 0.05） to 85% （P 〈 0.01） after 6-24 h treatment. crude LBE at a dose of 5 g/L suppressed cell proliferation of H-4-Ⅱ-E cells more effectively than crude RGE after 6-24 h incubation （P 〈 0.01）. crude LBE （2-10 g/L） and RGE （2-5 g/L） also dose-dependently inhibited proliferation of HA22T/VGH cells by 14%-43% （P 〈 0.01） after 24 h. crude LBE at a dose of 10 g/L inhibited the proliferation of HA22T/VGH cells more effectively than crude RGE （56.8% ＋ 1.6% vs 70.3% ＋ 3.1% of control, P = 0.0003 〈 0.01）. The apoptotic cells significantly increased in H-4-Ⅱ-E cells after 24 h treatment with higher doses of crude LBE （2-5 g/L） and RGE （5-10 g/L）（P 〈 0.01）. The expression of p53 protein in H-4-Ⅱ-E cells was 119% and 143% of the control group compared with the LBE-treated （2, 5 g/L） groups, and 110% and 132% of the control group compared with the RGE -treated （5, 10 g/L） groups after 24 h. cONcLUSION： Hot water-extracted crude LBE （2-5 g/L） and RGE （5-10 g/L） inhibit proliferation and stimulate p53-mediated apoptosis in Hcc cells.

The study investigated the effect of Benthosemapterotumpeptide on aging in mice. Mice aged of 3 monthswere divided into A toE groups. Mice in group B to E were injected with D-galactose, and orally administered Benthosemapterotum peptide at 0, 10, 50, and 100 mg/d, respectively, for 6 weeks after 6-week induction. Group A as the control group was injected with saline without treatment. The results showed that brain-derived neurotrophy factor （BDNF） level in the braindecreased in group B was significantly increased in treated groups. Time to platform in group A, D, and E was significantly less than that in group B. Group A had more correct annulus crossings and latency time in platform than group B. Group c had similar correct annulus crossings and latency time in platform as group A. Therefore, Benthosemapterotum peptideelevates brain BDNF and improves memory function in aging mice.

cu-rich cell boundary phase is difficult to precipitate evenly,resulting in a generally poor demagnetization curve squareness for Fe-rich Sm_(2)co_(17)-type magnet,which is a key factor limiting the further improvement of magnetic energy *** this study,we report that nanoscale strip-like ordered micro-domains distributed in1:7H disordered matrix phase of the solid solution precursor is a new factor significantly affecting the precipitation and distribution of the cell boundary *** strip-like and continuous micro-twin structure with twin boundaries neatly perpendicular to the c-axis is observed after sintering *** solution treatment,sequential and long strip-like micro-twins gradually transform into disordered state along the basal plane,forming narrow disordered 1:7H(Tbcu_(7)-type structure)phase between the separated strip-like ordered *** disordering transformation takes place via broken down of the long strip-like ordered micro-domains,which is accomplished by narrowing along the width direction followed by reduction of the ***,a new model revealing the effect of the ordered micro-domains on the formation of the cell boundary phase is *** boundaries enriched in cu have already existed in the precursor with long strip-like ordered ***,the cu-rich cell boundary phase acting as strong pinning centers cannot be precipitated homogeneously and distributed continuously after aging,resulting in a poor demagnetization curve squareness of Sm_(2)co_(17)-type *** results indicate that significant broken down of the nanoscale ordered micro-domains in solution precursor is the key factor improving the distribution of cell boundary phase in Sm_(2)co_(17)-type magnets.

We explored a new approach to search for a low-mass Z′particle through j/ψdecays by identifying its existence through parity-violating phenomena in the isospin-violating final states ofΛΣ¯^(0)and the corresponding charge conjugated states ofΛΣ¯^(0).Our investigation centered on a generation-independent and leptophobic Z′with mass below 10 *** the present experimental conditions at Beijing Spectrometer III(BESIII)and the anticipated opportunities at the Super Tau charm Factory(STcF),we conducted Monte-carlo simulations to predict possible events at both ***,we foresee a substantial enhancement in the precision of the lower limit estimation ofαNP as well as a reduction in statistical uncertainty with upcoming STcF ***,it is essential to highlight that a null result in the measurement ofαNP would impose stringent constraints,requiring the Z′−q−q couplings to be in the order of 10^(−2).

Rapid Antidepressant-Like Potential of chaihu Shugan San Depends on Suppressing Glutamate Neurotransmission and Activating Synaptic Proteins in Hippocampus of Female Mice

Objective To explore the rapid antidepressant potential and the underlying mechanism of chaihu Shugan San(cSS)in female *** Liquid chromatography mass spectrometry(Lc-MS)/MS was used to determine the content of main components in cSS to determine its *** c57BL/6j mice were randomly divided into 4 groups,including control(saline),vehicle(saline),cSS(4 g/kg)and ketamine(30 mg/kg)*** were subjected to irregular stress stimulation for 4 weeks to establish the chronic mild stress(cMS)model,then received a single administration of *** hours later,the behavioral tests were performed,including open field test,tail suspension test(TST),forced swimming test(FST),novelty suppression feeding test(NSF),and sucrose preference test(SPT).Western blot analysis was used to detect the expression levels of N-methyl-D-aspartate receptor(NMDA)subtypes[N-methyl-D-aspartate receptor 1(NR1),NR2A,NR2B],synaptic proteins[synapsin1 and post synaptic density protein 95(PSD95)],and brain-derived neurotrophic factor(BDNF).Moreover,the rapid antidepressant effect of cSS was tested by pharmacological technologies and optogenetic interventions that activated glutamate receptors,*** compared with the vehicle group,a single administration of cSS(4 g/kg)reversed all behavioral defects in TST,FST,SPT and NSF caused by cMS(PcSS also significantly decreased the expressions of NMDA subtypes(NR1,NR2A,NR2B)at 2 h in hippocampus of mice(all PcSS increased levels of synaptic proteins and BDNF(Pcts of cSS were blocked by transient activation of NMDA receptors in the hippocampus(all Pconclusion Rapid antidepressant effects of cSS by improving behavioral deficits in female cMS mice depended on rapid suppression of NMDA receptors and activation of synaptic proteins.
