A Cell-Penetr.ting Peptide for.Inhibiting MAPKAP Kinase 2-Mediated Inflammator. Cytokine r.lease Following Glial Cell Activation

Centr.l ner.ous system (CNS) injur. initializes a r.active tissue r.sponse, which is char.cter.zed by a cascade of signals that r.sult in inflammator. cytokine r.lease and neur.inflammation. These signals ar. induced in par. by activated micr.glia that migr.tes to the injur. site in an attempt to r.move dead tissue and pr.mote healing. The ability to contr.l the r.active tissue r.sponse is of significant impor.ance to a var.ety of applications, such as neur.pr.stheses, whose functional lifespan is limited by glial cell activation. A possible str.tegy to mitigate glial cell activation is to r.duce the r.lease of inflammator. cytokines following a br.in injur.. One pathway that facilitates inflammator. cytokine r.lease is the mitogen-activated pr.tein kinase-activated pr.tein kinase 2 (MK2) pathway, which incr.ases cytokine mr.A synthesis, stability and tr.nslation following activation. Ther.for., inhibiting MK2-mediated cytokine r.lease can r.duce micr.glial activationfollowing CNS injur.. Thr.ugh?in vitr.?studies, we demonstr.te the ability of a cell-penetr.ting peptide inhibitor.of MK2 (MK2i) to r.duce MK2-mediated cytokine r.lease in mixed cor.ical cultur.s.?Immunocytochemistr., enzyme-linked immunosor.ent assay (ELISA), and cytotoxicity assays in 7- -10-day-old E17 Spr.gue-Dawley r.t mixed cor.ical cultur.s showed that MK2i tr.atment significantly lower.d inflammator. cytokine pr.duction and incr.ased cor.ical cell viability following?glial cell activation with tumor.necr.sis factor.α(TNF-α) and lipopolysacchar.de (LPS). r.sults suggest that MK2i may r.duce damage caused by activated glia in the inflamed CNS.

This paper.investigates the design pr.blem of a sliding mode contr.ller.for.linear.r.petitive pr.cesses(Lr.s) with a finite pass length on each pass. Under.limited communication r.sour.es, an eventtr.gger.d mechanism is implemented fr.m the sensor. to the contr.ller. whose tr.gger.d sequence is consistent with the evolution dir.ction of Lr.s. To per.odically or.hestr.te communication between the contr.ller.and actuator. a r.und-r.bin scheduling mechanism is established based on the pass length. A sliding function involving the state and pass pr.file is constr.cted, and the sliding mode contr.ller.is designed using tr.gger.d states and pass pr.file signals. Feasible contr.ller.gains ar. attained to ensur. the stability of the r.sultant closed-loop system. Finally, the pr.posed str.tegy is implemented for.a multi-r.ll metal r.lling pr.cess.
