Objective: As isolates of Cryptococcus are frequently kept collection stocks in institutions, sometimes without proper characterization, we sought to determine the genotype profiles, protease and phospholipase activities “in vitro” and the susceptibility testing for azoles and amphotericin B. Methodology: 84 isolates from several regions of Brazil (40 samples from clinical origin and 44 isolates from environmental origin) were maintained at the microorganism’s bank of the Biomedical Science Institute (ICB-USP) of the São Paulo University, in São Paulo, Brazil. This isolates was submitted fungal strains determination, DNA extraction and purification, determination of genotype by URA5 gene RFLP of CGB-positive isolates, protease and phospholipase activity and susceptibility to antifungals. Results: Of six CGB positive isolates tested by RFLP-PCR, only four presented a genomic profile consistent C. gattii species (VGII), while two other were C. neoformans (VNI and VNIII), indicating the existence of canavanine-resistante C. neoformans isolates in the culture collections. The clinical isolates secreted higher levels of phospholipase and environmental isolates but no differences were observed for the protease levels. Almost all isolates were sensible to azoles and amphotericin B. Conclusion: We point out in this research the existence of C. neoformans strains resistant to canavanine and intrinsic characteristic of C. gatti. These results demonstrate the importance to perform a detailed characterization of isolates kept in culture collections.

Validating cone-beam computed tomography for peri-implant bone morphometric analysis

Cone-beam computed tomography （CBCT） has been recently used to analyse trabecular bone structure around dental implants. To validate the use of CBCT for three-dimensional （3D） peri-implant trabecular bone morphometry by comparing it to two-dimensional （2D） histology, 36 alveolar bone samples （with implants n=27 vs. without implants n=9） from six mongrel dogs, were scanned ex vivo using a high-resolution （80 ~m） CBCT. After scanning, all samples were decalcified and then sectioned into thin histological sections （-6 jim） to obtain high contrast 2D images. By using CTAn imaging software, bone morphometric parameters including trabecular number （Tb.N）, thickness （***）, separation （***） and bone volume fraction （BV/TV） were examined on both CBCT and corresponding histological images. Higher *** and ***, lower BV/TV and Tb.N were found on CBCT images （P〈0.001）. Both measurements on the peri-implant trabecular bone structure showed moderate to high correlation （r=0.65-0.85）. The Bland-Altman plots showed strongest agreement for *** followed by ***, Tb.N and BV/TV, regardless of the presence of implants. The current findings support the assumption that peri-implant trabecular bone structures based on high-resolution CBCT measurements are representative for the underlying histological bone characteristics, indicating a potential clinical diagnostic use of CBCT-based peri-implant bone morphometric characterisation.

Dietary Knowledge, Behaviours and Attitudes of Students of Public School in a Northeastern City in Brazil

This quasi-experimental intervention was conducted with 10- to 14-year-old students enrolled in public schools in Teresina, Brazil, to evaluate the effects of a nutritional education program on the students’ knowledge, behaviour and attitudes towards nutrition. The dietary attitudes were evaluated in terms of food tastes/preferences and food behaviours. The participants in the study included 126 students from four schools. Differences between before and after the intervention were found regarding knowledge about the “food pyramid”, “healthy eating” and “nutrients”

Agate geodes are treated and stained for the production of handmade artifacts and jewelry, which greatly increases their aggregate value. Unit operations involve storage, sorting, cutting, crushing, dyeing, polishing, washing, and finishing. Substantial amounts of waste materials are produced, which include agate pieces, powders (with or without oils), and wastewaters from organic and inorganic staining procedures. The aim of this work was to quantify the amounts of these waste materials, their characteristics, and possibilities for reuse or recycling in other productivity sectors in the context of the second decade of the 21st century. A review of the theme was carried out and possibilities were analyzed in the light of classic and new technologies.

Design Optimization and Weight Reduction of 500 mL CSD PET Bottle through FEM Simulations

Simulation description of a poly(ethylene terephthalate) bottle, blowing through the injection stretch blow molding process, using computer aided engineering softwares and analysis by finite elements method in order to better fit the bottle project using this material and weight reduction. The work was conducted from existing preforms and focused on the design process of the bottles, approach in which the available literature is scarce or nonexistent. The design of a bottle was executed, applied and validated through parametric computer aided design software and finite elements method analysis, allowing the simulation of the blowing process from data input to the process variables listed in the available literature. Using this approach, we simulate the blowing process and the mechanical demands related to the application with the aim of assessing the efficiency of packaging design and enable an efficient and correct sizing of the bottles. The data obtained in simulations were verified by the fabrication of prototypes using the same material, preform and process data considered in the virtual environment. The results indicate the feasibility of producing 500 mL bottles for carbonated soft drinks from 18 g preforms, with a reduction of up to 21% on weight of the packaging, keeping the standard bottle finish PCO-1810-28 mm.

Secondary Metabolites of the Genus Trichilia:Contribution to the Chemistry of Meliaceae Family

According to the literature data on the chemical composition of the Trichilia genus performed in this work, it can be concluded that 334 different compounds were isolated and identified, distributed in monoterpenes, sesquiterpenes, diterpenes, triterpenes, steroids, limonoids, coumarins, flavonoids, lignans, phenolic acids, amino acids and lactones. Together with the structures of this compound, data from botanical classification and pharmaco-logical results from extracts and pure compounds on the Trichilia genus were also described. The compounds derived from terpenes pathway were more significant, corresponding to about 87.7% of isolated and identified compounds from various Trichilia species. Among the different terpenoid skeletons of this kind, limonoids were meanly reported, appearing a total of 33.9% of compounds isolated from several Trichilia species.

Use of Coal Waste as Fine Aggregates in Concrete Paving Blocks

The aim of this work was to study the use of coal waste to produce concrete paving blocks. The methodology considered the following steps: sampling of a coal mining waste;gravity separation of the fraction with specific gravity between 2.4 and 2.8;comminution of the material and particle size analysis;technological characterization of the material and the production of concrete paving blocks. The results showed that the coal waste considered in this work can be used to replace conventional sand as a fine aggregate for concrete paving blocks. This practice can collaborate in a cleaner coal production.

A method for adsorption involving banana peel (BP) was studied to remove the pollutant acetylsalicylic acid (ASA) in aqueous medium. The results show that bioadsorbent has satisfactory maximum adsorption capacity (2.29 mg/g) for removing this analgesic and anti-inflammatory drug in aqueous solution (pH 7.0) using the Langmuir mathematical model. The tested concentrations of this pollutant were higher than the levels commonly found in the aquatic environment. This and other results suggest the BP as an alternative to ASA removal in water contaminated with pharmaceuticals pollutants.

Weather events put human lives at risk mostly when people might occupy areas susceptible to natural *** Professional Weather Stations(PWS)in vulnerable areas is key for monitoring weather with reliable ***,such professional instrumentation is notably expensive while remote sensing from a number of stations is *** imposes challenges on the large-scale weather station deployment for broad monitoring from large observation networks such as in Cemaden—The Brazilian National Center for Monitoring and Early Warning of Natural *** this context,in this paper,we propose a Low-Cost Automatic Weather Station(LCAWS)system developed from Commercial Off-The-Shelf(COTS)and open-source Internet of Things(IoT)technologies,which provides measurements as reliable as a reference PWS for natural disaster *** being automatic,LCAWS is a stand-alone photovoltaic system connected wirelessly to the Internet in order to provide real-time reliable end-to-end weather *** achieve data reliability,we propose an intelligent sensor calibration method to correct *** a 30-day uninterrupted observation with sampling in minute resolution,we show that the calibrated LCAWS sensors have no statistically significant differences from the PWS *** such,LCAWS has opened opportunities for reducing maintenance costs in Cemaden's observational network.

The assessment of detrital zircon age records is a key method in basin analysis,but it is prone to several biases that may compromise accurate sedimentary provenance *** to ultrahigh temperature(HT-UHT)metamorphism(especially if T>850℃)is herein presented as a natural cause of bias in provenance studies based on U-Pb detrital zircon ages,since zircon from rocks submitted to these extreme and often prolonged conditions frequently yield protracted,apparently concordant,geochronological *** age spreading can result from disturbance of the primary U-Pb zircon system,likewise from(re)crystallization processes during multiple and/or prolonged metamorphic *** this contribution,available geochronological data on Archean,Neoproterozoic and Palaeozoic HT-UHT metamorphic rocks,acquired by different techniques(SIMS and LA-ICP-MS)and showing distinct compositions,are reassessed to demonstrate HT-UHT metamorphism may result in modes and age distributions of unclear geological *** a consequence,it may induce misinterpretations on UPb detrital zircon provenance analyses,particularly in sedimentary rocks metamorphosed under such extreme temperature *** evaluate the presence of HT-UHT metamorphism-related bias in the detrital zircon record,we suggest a workflow for data acquisition and interpretation,combining a multi-proxy approach with:(i)in situ U-Pb dating coupled with Hf analyses to retrieve the isotopic composition of the sources,and(ii)the integration of a petrochronological investigation to typify fingerprints of the HT-UHT metamorphic *** proposed workflow is validated in the investigation of one theoretical and one natural example allowing a better characterization of the sedimentary sources,maximum depositional ages,and the tectonic setting of the *** workflow allows to the appraisal of biases imposed by HT-UHT metamorphism and resulting disturbances in the U-Pb detrital zircon record,particularly for sedimentary
