Evaluation of three sets of advanced backcrosses of eggplant with wild relatives from different gene pools under low N fertilization conditions

The development of new cultivars with improved nitrogen use efficiency(NUE)is key for implementing sustainable agriculture *** wild relatives(CWRs)provide valuable genetic resources for breeding programs aimed at achieving this *** this study,three eggplant(Solanum melongena)accessions together with their advanced backcrosses(ABs;BC3 to BC5 generations)were evaluated for 22 morpho-agronomic,physiological,and NUE traits under low nitrogen(LN)fertilization *** ABs were developed with introgressions from the wild relatives Solanum insanum,Solanum dasyphyllum,and Solanum *** AB population comprised a total of 25,59,and 59 genotypes,respectively,with overall donor wild relative genome coverage percentages of 58.8%,46.3%,and 99.2%.The three *** recurrent parents were also evaluated under control(normal)N *** of N fertilization in the parents resulted in decreased chlorophyll content-related traits,aerial biomass,stem diameter,and yield and increased NUE,nitrogen uptake efficiency(NUpE),and nitrogen utilization efficiency(NUtE).However,the decrease in yield was moderate,ranging between 62.6%and 72.6%.A high phenotypic variation was observed within each of the three sets of ABs under LN conditions,with some individuals displaying improved transgressive characteristics over the recurrent *** the single primer enrichment technology 5 k probes platform for high-throughput genotyping,we observed a variable but high degree of recurrent parent genome recovery in the ABs attributable to the lines recombination,allowing the successful identification of 16 quantitative trait loci(QTL).Different allelic effects were observed for the introgressed QTL *** candidate genes were identified in the QTL regions associated with plant growth,yield,fruit size,and NUE-related *** results show that eggplant materials with introgressions from CWRs can result in a dramatic impact in eggplant breedi

Molecular and in vitro Characterization of Field Isolates of Bovine Herpesvirus-1

Bovine Herpesvirus-1 (BoHV-1) is distributed worldwide and is a major pathogen in cattle, being the causal agent of a variety of clinical syndromes. The aim of this study was to isolate and to characterize (molecular and biological characterization) BoHV-1 from 29 immunosuppressed animals. It was possible to obtain 18 isolates, each from a different animal, such as from the respiratory and reproductive tracts. In some cases the cytopathic effect was visible 12 hours post-inoculation, and became characteristic after 36-48 hours. Biological characteristics were evaluated and compared with Iowa and Colorado-1 reference strains, and differences were found in plaque size, virus titer measured by TCID50 and PFU/mL, and one step virus curves. These results showed that some isolates had a highly virulent-like behavior in vitro, compared to the reference strains, with shorter eclipse periods, faster release of virus into the supernatants, and higher burst size and viral titer. There were no differences in glycoprotein expression of BoHV-1 isolates, measured by Western blot on monolayers. Moreover, using restriction endonucleases analysis, most of the viruses were confirmed as BoHV-1.1 and just one of them was confirmed as BoHV-1.2a subtype. These findings suggest that some wild-type BoHV-1 isolates could be useful as seeds to develop new monovalent vaccines.

Factors affecting somatic embryogenesis in conifers

This review seeks to examine the extreme response of isolated somatic plant cells of apical meristematic tissues of mature conifer trees towards specific stress conditions in vitro resulting in somatic embryo- genesis. Signal molecules regulating embryo development have been described in angiosperms, but very little is known about somatic rejuve- nation in conifers. Recent studies on cloning of mature conifers provide new perspectives on signal molecules on cellular dedifferentiation into the embryogenic pathway. Our recent studies show that signal molecules such as butenolide, calcium ions, salicylic acid, antioxidants, amino acids triacontanol and 24-epibrassinolide all play an important role in the con- version of somatic cells into an embryogenic pathway in many recalcitrant pines. This constitutes a major breakthrough in forest biotechnology with many practical applications in clonal forestry.

The ability of sperm selection techniques to remove single- or double-strand DNA damage

A wide variety of techniques for the preparation of sperm are currently available, of which the most commonly employed are densitygradient centrifugation （DGC） and swim-up （SUP）. To date, these methods appear to be effective in selecting functional sperm for assisted reproduction techniques （ART）, but they may have negative effects on sperm DNA. In this study, the ability of these semen processing techniques to eliminate spermatozoa containing single- and double-strand DNA damage was assessed by the two-tailed comet assay and the sperm chromatin dispersion test in 157 semen samples from patients seeking assisted reproduction treatment. Our results indicated that SUP and DGC are equally efficient in eliminating spermatozoa containing double-strand DNA damage and sperm with highly damaged （degraded） DNA, as characterized by the presence of both single- and double-strand DNA breaks. However, DGC is more efficient than SUP in selecting spermatozoa that are free from single-strand DNA damage. Future studies should characterise the importance of the various types of DNA damage and examine the sperm processing protocols used in each laboratory to determine their ability to eliminate DNA damage and hence, prevent the potential transmission of genetic mutations via ART.

Pterocarpus santalinus L. f.(Fabaceae;red sanders) is prized for its wood whose colour and fragrance is due to the presence of santalins that have pharmaceutical and industrial uses. Red sanders is listed as an endangered plant species on the IUCN red data list as a result of the exploitation of its wood and essential oil. This review emphasizes the pollination biology, seed germination, vegetative propagation and micropropagation of P. santalinus. Excessive use of P. santalinus has also caused the emergence of various adulterants, so accurate identification is essential.

Leaf Expansion and Transpiration Responses of Millet Species to Soil Water Deficit

Plant processes, such as leaf expansion, stomatal conductance and transpiration, are affected by soil water, particularly in waterstressed environments. Quantifying the effects of soil water on plant processes, especially leaf expansion and transpiration, could be useful for crop modeling. In order to quantify the leaf expansion and transpiration in response to soil water deficit in three millet species, common(Panicum miliaceum L.), pearl(Pennisetum glaucum L.) and foxtail(Setaria italica L.) millets, a pot experiment was performed at the Gorgan University of Agricultural Sciences and Natural Resources, Gorgan, Iran. The soil water status was characterized by the fraction of transpirable soil water(FTSW). Leaf area and transpiration were measured daily. Relative leaf area expansion(RL) and relative transpiration(RT) data were plotted against FTSW. Finally the FTSW thresholds for RL and RT were calculated using linear-plateau and logistic models. The results showed that the thresholds for RL and RT were 0.68 and 0.62,respectively, based on all measured data of the three millet species using the linear-plateau model, indicating that RL and RT were constant when FTSW decreased from 1 to the threshold point. Thereafter, until FTSW = 0, RL and RT declined linearly with a slope of 1.48 and 1.43, respectively. Although millet is cultivated as a resistant crop in arid, semiarid and marginal lands, it showed an early response to soil water deficit at high FTSW thresholds. As leaf expansion and transpiration can be considered morphological and physiological variables, respectively, the results in this study indicate that millet has strong morphological flexibility when faced with soil water deficit.

Applicability of the PRECISE-DAPT score in elderly patients with myocardial infarction

Background Elderly patients with acute coronary syndromes(ACS)are at higher risk both for ischemic and bleeding *** guidelines recommend the PRECISE-DAPT score for bleeding risk stratification in this setting,but no study assessed its applicability in elderly *** study aimed to assess the performance of the PRECISE-DAPT score in a series of non-selected elderly patients with ACS from routine clinical *** The IFFANIAM registry included prospectively patients aged>75 years with ST segment elevation myocardial infarction(STEMI).Main outcome measured was the incidence of relevant bleeding after discharge(bleeding leading to hospital readmission,need for transfusion,intervention,stop of antithrombotic drugs or death).Bleeding risk was classified:(A)according to PRECISE-DAPT values above or not the recommended cut-off point(>25);and(B)according to the quartiles of PRECISE-DAPT values observed in the IFFANIAM series(QI:45).Results A total of 208 patients were *** age was 81.9±4.5 *** patients(92.6%)had a PRECISE-DAPT value>25.A total of 25 patients(12.0%)had bleeding events and 49 patients(23.6%)*** significant differences regarding the incidence of bleeding were observed according to the recommended cutt of point>***,a progressive increase in the incidence of bleeding was observed across PRECISE-DAPT quartiles observed in this series(P=0.038).Conclusions The vast majority of elderly patients have PRECISE-DAPT values above the recommended cut-off point for bleeding *** different cut-off points could be a more rational approach for predicting bleeding risk in these complex patients.
