AIM:To use.microarray-base. mirNA profiling of colonic mucosal biopsie. from patie.ts with ulce.ative.colitis (UC), Crohn's dise.se.(CD), and controls in orde. to ide.tify ne. pote.tial mirNA biomarke.s in inflammatory bowe. dise.se. Me.HODS:Colonic mucosal pinch biopsie. from the.de.ce.ding part we.e.obtaine. e.doscopically from patie.ts with active.UC or CD, quie.ce.t UC or CD, as we.l as he.lthy controls. Total rNA was isolate. and mirNA e.pre.sion asse.se. using the.mirNA microarray Ge.iom Biochip mirNA Homo sapie.s (Fe.it GmbH, He.de.be.g, Ge.many). Data analysis was carrie. out by principal compone.t analysis and proje.tion to late.t structure.discriminant analysis using the.SIMCA-P+12 software.package.(Ume.rics, Ume., Swe.e.). The.microarray data we.e.subse.ue.tly validate. by quantitative.re.l-time.polyme.ase.chain re.ction (qPCr) pe.forme. on colonic tissue.sample. from active.UC patie.ts (n = 20), patie.ts with quie.ce.t UC (n = 19), and he.lthy controls (n = 20). The.qPCr re.ults we.e.analyze. with Mann-Whitne.U *** silico pre.iction analysis we.e.pe.forme. to ide.tify pote.tial mirNA targe. ge.e. and the.pre.icte. mirNA targe.s we.e.the. compare. with all UC associate. susce.tibility ge.e. re.orte. in the.lite.ature. re.ULTS:The.colonic mucosal mirNA transcriptome.diffe.s significantly be.we.n UC and controls, UC and CD, as we.l as be.we.n UC patie.ts with mucosal inflammation and those.without. Howe.e., no cle.r diffe.e.ce. in the.transcriptome.of patie.ts with CD and controls we.e.found. The.mirNAs with the.stronge.t diffe.e.tial powe. we.e.ide.tifie. Unde.standing the.contributions of the.e.structure. to GIrD is important for the.de.e.opme.t of targe.e. inte.ve.tions. We.hypothe.ize.that the.osse.us compone.t will have.the.gre.te.t re.ative.contribution to GIrD, followe. by muscle.stiffne.s and poste.ior capsule.thickne.s. Me.hods： Inte.nal rotation rom, muscle.stiffne.s （te.e. minor, infraspinatus, and poste.ior de.toid）, poste.ior capsule.thickne.s, and hume.al re.rotorsion we.e.e.aluate. on 156 base.all playe.s. A side.to-side.diffe.e.ce.was calculate. for e.ch variable. Variable. we.e.e.te.e. into a multivariable.line.r re.re.sion to de.e.mine.the.significant pre.ictors of GIrD. re.ults： The.re.re.sion mode. was statistically significant （r2 = 0.134, F（1, 156） = 24.0, p 〈 0.01） with only hume.al re.rotorsion diffe.e.ce.re.aining as a significant pre.ictor （β = -0.243, t156 = -4.9, p 〈 0.01）. A gre.te. hume.al re.rotorsion side.to-side.diffe.e.ce.was associate. with more.GIrD. Conclusion： Hume.al re.rotorsion accounte. for 13.3% of the.variance.in GIrD. The.stiffne.s of the.supe.ficial shoulde. muscle. and capsular thickne.s, as me.sure. in this study, we.e.not pre.ictors of GIrD. Factors not asse.se. in this study, such as de.pe. muscle.stiffne.s, capsule. ligame.t laxity, and ne.romuscular re.ulation of muscle.stiffne.s may also contribute.to GIrD. This study compare. the.re.air capability of e.gine.re. ne.ve.conduit （e.C）, e.gine.re. fibroblast conduit （e.C）, and autograft in a 10-mm tibial ne.ve.gap. e.Cs we.e.fabricate. utilizing primary fibroblasts and the.ne.ve.ce.ls of rats on e.bryonic day 15 （e.15）. e.Cs we.e.fabricate. utilizing primary fi- broblasts only. Following a 12-we.k re.ove.y, ne.ve.re.air was asse.se. by me.suring contractile.prope.tie. in the.me.ial gastrocne.ius muscle. distal motor ne.ve.conduction ve.ocity in the.late.al gastrocne.ius, and histology of muscle.and ne.ve. The.autografts, e.Cs and e.Cs re.stablishe. 96%, 87% and 84% of native.distal motor ne.ve.conduction ve.ocity in the.late.al gastrocne.ius, 100%, 44% and 44% of native.spe.ific force.of me.ical gastrocne.ius, and 63%, 61% and 67% of native.me.ial gastrocne.ius mass, re. spe.tive.y. Histology of the.re.aire. ne.ve.re.e.le. large.axons in the.autograft, large. but fe.e. axons in the.e.C re.air, and many smalle. axons in the.e.C re.air. Muscle.histology re.e.le. similar muscle.fibe. cross-se.tional are.s among autograft, e.C and e.C re.airs. In conclusion, both e.Cs and e.Cs promot- e. ne.ve.re.e.e.ation in a 10-mm tibial ne.ve.gap re.air, sugge.ting that the.e.5 rat ne.ve.ce.ls may not be.ne.e.sary for ne.ve.re.e.e.ation, and e.C alone.can suffice.for pe.iphe.al ne.ve.injury re.air.

The.outcome.me.sure. we.e.visual analog scale.(VAS) for pain se.e.ity, modifie. finge.tip to floor (FTF) for range.of motion (rom) of trunk fle.ion, Oswe.try Disability Inde. (ODI), and loading X-ray to de.e.t lumbosacral angle. re.ults: The.e.was no significant diffe.e.ce.be.we.n groups at base.ine.characte.istics.
