The joint European Space Agency and Chinese Academy of Sciences Solar wind Magnetosphere Ionosphere Link Explorer(SMILE)mission will explore global dynamics of the magnetosphere under varying solar wind and interplanetary magnetic field conditions,and simultaneously monitor the auroral response of the Northern Hemisphere ionosphere.Combining these large-scale responses with medium and fine-scale measurements at a variety of cadences by additional ground-based and space-based instruments will enable a much greater scientific impact beyond the original goals of the SMILE mission.Here,we describe current community efforts to prepare for SMILE,and the benefits and context various experiments that have explicitly expressed support for SMILE can offer.A dedicated group of international scientists representing many different experiment types and geographical locations,the Ground-based and Additional Science Working Group,is facilitating these efforts.Preparations include constructing an online SMILE Data Fusion Facility,the discussion of particular or special modes for experiments such as coherent and incoherent scatter radar,and the consideration of particular observing strategies and spacecraft conjunctions.We anticipate growing interest and community engagement with the SMILE mission,and we welcome novel ideas and insights from the solar-terrestrial community.

Observability analysis(OA)is vital to obtaining the available input measurements of state estimation(se)in an integrated electricity and heating system(IEHS).Considering the thermal quasi-dynamics in pipelines,the measurement equations in heating systems are dependent on the estimated results,leading to an interdependency between OA and se.Conventional OA methods require measurement equations be known exactly before se is performed,and they are not applicable to IEHSs.To bridge this gap,a scenario-based OA scheme for IEHSs is devised that yields reliable analysis results for a predefined set of time-delay scenarios to cope with this interdependency.As its core procedure,the observable state identification and observability restoration are formulated in terms of integer linear programming.Numerical tests are conducted to demonstrate the validity and superiority of the proposed formulation.

Background:Adenosine triphosphatase inhibitory factor 1(IF1)is a key protein involved in energy metabolism.IF1 has been linked to various agerelated diseases,although its relationship with physical activity(PA)remains unclear.Additionally,the apolipoprotein A-I(apoA-I),a PA-modulated lipoprotein,could play a role in this relationship because it shares a binding site with IF1 on the cell-surface ATP synthase.We examined here the associations between chronic PA and plasma IF1 concentrations among older adults,and we investigated whether apoA-I mediated these associations.Methods:In the present work,1096 healthy adults(63.8%females)aged 70 years and over who were involved in the Multidomain Alzheimer Prevention Trial study were included.IF1 plasma concentrations(square root of ng/mL)were measured at the 1-year visit of the Multidomain Alzheimer Prevention Trial,while PA levels(square root of metabolic equivalent task min/week)were assessed using questionnaires administered each year from baseline to the 3-year visit.Multiple linear regressions were performed to investigate the associations between the first-year mean PA levels and IF1 concentrations.Mediation analyses were conducted to examine whether apoA-I mediated these associations.Mixedeffect linear regressions were carried out to investigate whether the 1-year visit IF1 concentrations predicted subsequent changes in PA.Results:Multiple linear regressions indicated that first-year mean PA levels were positively associated with IF1 concentrations(B=0.021;se=0.010;p=0.043).Mediation analyses revealed that about 37.7%of this relationship was mediated by apoA-I(B_(ab)=0.008;se=0.004;p=0.023).Longitudinal investigations demonstrated that higher concentrations of IF1 at the 1-year visit predicted a faster decline in PA levels over the subsequent 2 years(time×IF1:B=0.148;se=0.066;p=0.025).Conclusion:This study demonstrates that regular PA is associated with plasma IF1 concentrations,and it suggests that apoA-I partly mediates this association.Additionally,this study f

Osteogenesis imperfecta（OI） comprises a group of heritable connective tissue disorders generally defined by recurrent fractures, low bone mass, short stature and skeletal fragility. Beyond the skeletal complications of OI,many patients also report intolerance to physical activity, fatigue and muscle weakness. Indeed, recent studies have demonstrated that skeletal muscle is also negatively affected by OI, both directly and indirectly. Given the well-established interdependence of bone and skeletal muscle in both physiology and pathophysiology and the observations of skeletal muscle pathology in patients with OI, we investigated the therapeutic potential of simultaneous anabolic targeting of both bone and skeletal muscle using a soluble activin receptor 2B（ACVR2B） in a mouse model of type Ⅲ OI（oim）. Treatment of 12-week-old oim mice with ACVR2 B for 4 weeks resulted in significant increases in both bone and muscle that were similar to those observed in healthy,wild-type littermates. This proof of concept study provides encouraging evidence for a holistic approach to treating the deleterious consequences of OI in the musculoskeletal system.

Gastrointestinal(GI)symptoms,such as diarrhea,abdominal pain,vomiting,and anorexia,are frequently observed in patients with coronavirus disease 2019(COVID-19).However,the pathophysiological mechanisms connecting these GI symptoms to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infections remain elusive.Previous studies indicate that the entry of SARS-CoV-2 into intestinal cells leads to downregulation of angiotensin converting enzyme 2(ACE2)receptors resulting in impaired barrier function.While intestinal ACE2 functions as a chaperone for the amino acid transporter B0AT1,the B0AT1/ACE2 complex within the intestinal epithelium acts as a regulator of gut microbiota composition and function.Alternations to the B0AT1/ACE2 complex lead to microbial dysbiosis through increased local and systemic immune responses.Previous studies have also suggested that altered serotonin metabolism may be the underlying cause of GI disorders involving diarrhea.The findings of elevated plasma serotonin levels and high fecal calprotectin in COVID-19 patients with diarrhea indicate that the viral infection evokes a systemic inflammatory response that specifically involves the GI.Interestingly,the elevated proinflammatory cytokines correlate with elevated serotonin and fecal calprotectin levels further supporting the evidence of GI inflammation,a hallmark of functional GI disorders.Moreover,the finding that rectal swabs of COVID-19 patients remain positive for SARS-CoV-2 even after the nasopharynx clears the virus,suggests that viral replication and shedding from the GI tract may be more robust than that of the respiratory tract,further indicating fecal-oral transmission as another important route of viral spread.This review summarized the evidence for pathophysiological mechanisms(impaired barrier function,gut inflammation,altered serotonin metabolism and gut microbiota dysbiosis)underlying the GI symptoms in patients with COVID-19.

Although the process by which the cortical tissues of the brain fold has been the subject of considerable study and debate over the past few decades,a single mechanistic description of the phenomenon has yet to be fully accepted.Rather,two competing explanations of cortical folding have arisen in recent years;known as the axonal tension and the differential tangential expansion models.In the present review,these two models are introduced by analyzing the computational,theoretical,materials-based,and cell studies which have yielded them.Then Four-dimensional bioprinting is presented as a powerful technology which can not only be used to test both models of cortical folding de novo,but can also be used to explore the reciprocal effects that folding associated mechanical stresses may have on neural development.Therein,the fabrication of‘smart’tissue models which can accurately simulate the in vivo folding process and recapitulate physiologically relevant stresses are introduced.We also provide a general description of both cortical neurobiology as well as the cellular basis of cortical folding.Our discussion also entails an overview of both 3D and 4D bioprinting technologies,as well as a brief commentary on recent advancements in printed central nervous system tissue engineering.

There has been increasing interest over recent years in the application of three-dimensional(3 D) printing technologies in the biomedical field. One such method is Direct Ink Writing(DIW); this approach has the potential advantage of allowing room-temperature deposition of materials, presented as an ink, to build complex architectures. DIW offers the ability to process biomaterials containing temperature-sensitive components. Due to the fabrication principles of DIW, there are specific rheological requirements that the ink must exhibit for the 3 D construction. For this reason, hydrogel-based liquid feed stocks have been the focal point of ink development. As a consequence, studies based on inks comprising hydrophobic biomaterials, which are insoluble in water and hence unsuited to the hydrogel approach, have been limited.In this study, we investigate novel inks that utilize polycaprolactone(PCL), a hydrophobic polymer, as the primary constituent by dissolving the polymer in solvent systems based on dichloromethane(DCM) and acetone(ACE). Moreover, polyethylene oxide(PEO) was incorporated into the PCL systems in order to extend the range of hydrophilicity of the systems. The rheological properties of the inks were investigated as a function of polymer composition and solvent system. Woodpile constructs of PCL and PCL/PEO were fabricated using DIW method and were assessed by a series of material characterisation. The type of solvent system had a noticeable impact on the ink rheology, which ultimately affected the surface properties. The incorporation of PEO particularly enhanced the roughness and wettability of the constructs. Our results support the use of DIW as a new means to process hydrophobic polymers for biomedical applications.

Recent proteogenomic approaches have led to the discovery that regions of the transcriptome previously annotated as non-coding regions[i.e.,untranslated regions(UTRs),open reading frames overlapping annotated coding sequences in a different reading frame,and non-coding RNAs]frequently encode proteins,termed alternative proteins(altProts).This suggests that previously identified protein–protein interaction(PPI)networks are partially incomplete because altProts are not present in conventional protein databases.Here,we used the proteogenomic resource OpenProt and a combined spectrum-and peptide-centric analysis for the re-analysis of a highthroughput human network proteomics dataset,thereby revealing the presence of 261 altProts in the network.We found 19 genes encoding both an annotated(reference)and an alternative protein interacting with each other.Of the 117 altProts encoded by pseudogenes,38 are direct interactors of reference proteins encoded by their respective parental genes.Finally,we experimentally validate several interactions involving altProts.These data improve the blueprints of the human PPI network and suggest functional roles for hundreds of altProts.
