Metabolic reprogramming by syntenin-1 directs RA FLs and endothelial cell-mediated inflammation and angiogenesis

A novel rheumatoid arthritis(RA)synovial fluid protein,syntenin-1,and its receptor,syndecan-1(sDC-1),are colocalized on RA synovial tissue endothelial cells and fibroblast-like synoviocytes(FLs).syntenin-1 exacerbates the inflammatory landscape of endothelial cells and RA FLs by upregulating transcription of IRF1/5/7/9,IL-1β,IL-6,and CCL2 through sDC-1 ligation and HIF1α,or mTOR ***,syntenin-1 orchestrates RA FLs and endothelial cell invasion via sDC-1 and/or mTOR *** syntenin-1 reprogrammed endothelial cells,the dynamic expression of metabolic intermediates coincides with escalated glycolysis along with unchanged oxidative factors,AMPK,PGC-1α,citrate,and inactive oxidative ***,RA FLs rewired by syntenin-1 displayed a modest glycolytic-ATP accompanied by a robust mitochondrial-ATP *** enriched mitochondrial-ATP detected in syntenin-1 reprogrammed RA FLs was coupled with mitochondrial fusion and fission recapitulated by escalated Mitofusin-2 and DRP1 *** found that VEGFR1/2 and Notch1 networks are responsible for the crosstalk between syntenin-1 rewired endothelial cells and RA FLs,which are also represented in RA *** to RA explants,morphological and transcriptome studies authenticated the importance of VEGFR1/2,Notch1,RAPTOR,and HIF1αpathways in syntenin-1 arthritic mice and their obstruction in sDC-1 deficient ***,dysregulation of sDC-1,mTOR,and HIF1αnegated syntenin-1 inflammatory phenotype in RA explants,while inhibition of HIF1αimpaired synovial angiogenic imprint amplified by *** conclusion,since the current therapies are ineffective on syntenin-1 and sDC-1 expression in RA synovial tissue and blood,targeting this pathway and its interconnected metabolic intermediates may provide a novel therapeutic strategy.

Coronal magnetic fields evolve quasi-statically over long timescales and dynamically over short *** of now there exist no regular measurements of coronal magnetic fields,and therefore generating the coronal magnetic field evolution using observations of the magnetic field at the photosphere is a fundamental requirement to understanding the origin of transient phenomena from solar active regions(ARs).Using the magneto-friction(MF)approach,we aim to simulate the coronal field evolution in the solar AR *** MF method is implemented in the open source PENCIL CODE along with a driver module to drive the initial field with different boundary conditions prescribed from observed vector magnetic fields at the *** order to work with vector potential and the observations,we prescribe three types of bottom boundary drivers with varying free-magnetic *** MF simulation reproduces the magnetic structure,which better matches the sigmoidal morphology exhibited by Atmospheric Imaging Assembly(AIA)images at the pre-eruptive *** found that the already sheared field further driven by the sheared magnetic field will maintain and further build the highly sheared coronal magnetic configuration,as seen in AR ***-driven MF simulation is a viable tool to generate the coronal magnetic field evolution,capturing the formation of the twisted flux rope and its eruption.

Dialysis plays a crucial role in the purification of nanomaterials but its impact on the structural properties of carbon nanomaterials was never ***,a carbon-based nanomaterial generated electrochemically in potassium phosphate buffer,was characterized before and after dialysis against pure *** is shown that dialysis affects the size of the carbon domains,structural organization,surface functionalization,oxidation degree of carbon,and grade of ***,dialysis drives the nanomaterial organization from discrete roundish carbon domains,with sizes ranging from 70 to 160 nm,towards linear stacking structures of small nanoparticles （surface groups evolve into more oxidized carbon groups （e.g.,ketone and ester groups）.Investigation of the as-prepared nanomaterial by electron paramagnetic resonance （EPR） revealed a resonance signal consistent with carbon-oxygen centred ***,this study brings to light the selective affinity of the carbon nanomaterial under study to capture Na+ions,a property greatly enhanced by the dialysis process,and its high ability to trap oxygen,particularly before *** findings open new perspectives for the application of carbon-based nanomaterials and raise awareness of the importance of structural changes that can occur during the purification of carbon-based nanomaterials.

We present a method to fabricate handcrafted thermoelectric devices on standard office paper *** devices are based on thin films of Ws_(2),Te,and BP(P-type semiconductors)and Tis_(3)and Tis_(2)(N-type semiconductors),deposited by simply rubbing powder of these materials against *** thermoelectric properties of these semiconducting films revealed maximum seebeck coefficients of(+1.32±0.27)mV K^(-1)and(-0.82±0.15)mV K^(-1)for Ws_(2)and Tis_(3),***,Peltier elements were fabricated by interconnecting the P-and N-type films with graphite electrodes.A thermopower value up to 6.11 mV K^(-1)was obtained when the Peltier element were constructed with three *** findings of this work show proof-of-concept devices to illustrate the potential application of semiconducting van der Waals materials in future thermoelectric power generation as well as temperature sensing for low-cost disposable electronic devices.

Detection of the second harmonic response of magnetic nanostructures to an ac current is shown to be a very sensitive probe of the magnetization reversal process. A temperature oscillation is obtained by joule heating instead of using a laser as the heat source, as in thermo-galvanic voltage measurements （TGV）. joule heating is used to produce a large local temperature gradient in asymmetric Co/Cu/Co spin valves. Evidence is found for an effect of a heat current on magnetization.

Methionine(Met)is an essential and first limiting amino acid in the poultry diet that plays a significant role in chicken embryonic development and *** present study examined the effect of in ovo injection of DL-Met and L-Met sources and genotypes on chicken embryonic-intestinal development and *** eggs of the two genotypes,TETRA-sL layer hybrid(TsL)—commercial layer hybrid and Hungarian Partridge colored hen breed(HPC)—a native genotype,were randomly distributed into four treatments for each *** treatment groups include the following:1)control non-injected eggs(NoIn);2)saline-injected(saIn);3)DL-Met injected(DLM);and 4)L-Met injected(LM).The in ovo injection was carried out on 17.5 d of embryonic development;after hatching,eight chicks per group were sacrificed,and the jejunum was extracted for *** results showed that both DLM and LM groups had enhanced intestinal development as evidenced by increased villus width,villus height,and villus area(Psignificantly reduced crypt depth,glutathione(GsH)content,glutathione s-transferase 3 alpha(GsT3),occludin(OCLN)gene expression and increased villus height to crypt depth ratio in the TsL genotype than the LM group(Ps overexpressed insulin-like growth factor 1(IGF1)gene,tricellulin(MD2),occludin(OCLN),superoxide dismutase 1(sOD1),and GsT3 genes than the TsL genotype(Psion,these findings showed that in ovo injection of Met enhanced intestinal development,and function,with genotypes responding differently under normal *** also influenced the expression of intestinal antioxidants,tight junction,and growth-related genes.

Abalone shell-derived Mg-doped mesoporous hydroxyapatite microsphere drug delivery system loaded with icariin for inducing apoptosis of osteosarcoma cells

Hydroxyapatite (HAP) porous microspheres with very high specific surface area and drug loading capacity, as well as excellent biocompatibility, have been widely used in tumour therapy. Mg2+ is considered to be a key factor in bone regeneration, acting as an active agent to stimulate bone and cartilage formation, and is effective in accelerating cell migration and promoting angiogenesis, which is essential for bone tissue repair, anti-cancer, and anti-infection. In this study, abalone shells from a variety of sources were used as raw materials, and Mg2+-doped abalone shell-derived mesoporous HAP microspheres (Mg-HAP) were prepared by hydrothermal synthesis as Mg2+/icariin smart dual delivery system (ICA-Mg-HAP, IMHA). With increasing of Mg2+ doping, the surface morphology of HAP microspheres varied from collapsed macroporous to mesoporous to smooth and non-porous, which may be due to Mg2+ substitution or coordination in the HAP lattice. At 30% Mg2+ doping, the Mg-HAP microspheres showed a more homogeneous mesoporous morphology with a high specific surface area (186.06 m2/g). The IMHA microspheres showed high drug loading (7.69%) and encapsulation rate (83.29%), sustained Mg2+ release for more than 27 days, sustained and stable release of icariin for 60 hours, and good responsiveness to ph (ph 6.4 > ph 5.6). In addition, the IMHA delivery system stimulated the rapid proliferation of bone marrow mesenchymal stem cells and induced apoptosis in MG63 cells by blocking the G2 phase cycle of osteosarcoma cells and stimulating the high expression of apoptotic genes (Bcl-2, caspase-3,-8,-9). This suggests that the abalone shell-based IMHA may have potential applications in drug delivery and tumour therapy. © 2024, Chinese Medical Multimedia Press Co Ltd. All rights reserved.

The incredibly complex soil microbial communities at small scales make their analysis and identification of reasons for the observed structures *** community structure is mainly a result of the inoculum(dispersal),the selective advantages of those organisms under the habitat-based environmental attributes,and the ability of those colonizers to sustain themselves over *** soil is protective,and its microbial inhabitants have long adapted to varied soil conditions,significant portions of the soil microbial community structure are likely ***,a substantial portion of the community will not correlate to often measured soil *** suggest that the drivers be ranked on the basis of their importance to the fundamental needs of the microbes:(i)those that supply energy,i.e.,organic carbon and electron acceptors;(i)environmental effectors or stressors,i.e.,ph,salt,drought,and toxic chemicals;(ii)macro-organism associations,i.e.,plants and their seasonality,animals and their fecal matter,and soil fauna;and(iv)nutrients,in order,N,P,and probably of lesser importance,other micronutrients,and *** relevance of drivers also varies with spatial and time scales,for example,aggregate to field to regional,and persistent to dynamic populations to transcripts,and with the extent of phylogenetic difference,hence phenotypic differences in organismal *** present a summary matrix to provide guidance on which drivers are important for particular studies,with special emphasis on a wide range of spatial and temporal scales,and illustrate this with genomic and population(rRNA gene)data from selected studies.

Revolutionizing plasmonic platform via magnetic field-assisted confined ultrafast laser deposition of high-density,uniform,and ultrafine nanoparticle arrays
