Zebrafish as a possible bioindicator of organic pollutants with effects on reproduction in drinking waters

Organic contaminants can be detected at low concentrations in drinking water, raising concerns for human health, particularly in reproduction. In this respect, we attempted to use the zebrafish as a bioindicator to detect the possible presence of these substances in drinking water, aiming to define the most relevant parameters to detect these substances, which particularly affect the development and reproduction of zebrafish. To this end, batches of30 embryos with the chorion intact were cultured in drinking waters from different sources,throughout their full life-cycle up to 5 months, in 20 L tanks. six replicates were performed in all water groups, with a total of 24 aquariums. Two generations（F0 and F1） were studied and the following parameters were tested： in the F0 generation, survival and abnormality rates evaluated at 5 dpf（days post-fertilization） and at 5 mpf（months post-fertilization）,the onset of spawning and the fertility rate from 3 mpf to 5 mpf, and the sex ratio and underdeveloped specimens at 5 mpf. Furthermore, in the F0 offspring（F1）, survival and abnormality rates were evaluated at 5 dpf and the hatching rate at 72 hpf. These results revealed that the hatching rate is the most sensitive parameter to distinguish different levels of effects between waters during the early life stages, whereas the rate of underdeveloped specimens is more suitable at later life stages. Regarding adult reproduction, fertility rate was the most sensitive parameter. The possible reversibility or accumulative nature of such effects will be studied in future work.

In the last decade,extensive research has been carried out on the microstructural behavior of high-entropy alloys(HEA),for which the in-situ formation of nanoparticles has been ***,studies of the incorporation of nanoparticles in HEA have been rarely *** this work,the addition of zinc oxide nanoparticles(ZnO NP)as reinforcement in a CoCrFeMoNi high-entropy alloy matrix,as well as the morphological,structural,and microstructural evolution of composites synthesized via powder metallurgy,were *** electron microscopy and X-ray diffraction analysis were performed in order to study the microstructural and phase characterization of the *** sintering,it was found that the ZnO NP addition(0.5 wt%,1 wt%and 2 wt%)had a significant influence on the microstructure and hardness of the CoCrFeMoNi high-entropy *** bonding among metal particles was promoted with the additions of Zn O NP.A reduction in porosity as a function of ZnO NP content was also *** microhardness results showed that the composite reached its highest reinforcement in bulk samples with 1 wt%ZnO NP(HV 870),which represented a 20%improvement over the unreinforced HEA matrix.

The aim was to evaluate the potential of mucus-permeating nanoparticles for the oral administration of *** nanocarriers,based on the coating of zein nanoparticles with a polymer conjugate containing PEG,displayed a size of 260 nm with a negative surface charge and an insulin payload of 77 mg/*** intestinal pig mucus,the diffusivity of these nanoparticles(PPA-NPs)was found to be 20-fold higher than bare nanoparticles(NPs).These results were in line with the biodistribution study in rats,in which NPs remained trapped in the mucus,whereas PPA-NPs were able to cross this layer and reach the epithelium *** therapeutic effcacy was evaluated in Caenorhabditis elegans grown under high glucose *** this model,worms treated with insulin-loaded in PPA-NPs displayed a longer lifespan than those treated with insulin free or nanoencapsulated in *** finding was associated with a signifcant reduction in the formation of reactive oxygen species(ROs)as well as an important decrease in the glucose and fat content in *** effects would be related with the mucus-permeating ability of PPA-NPs that would facilitate the passage through the intestinal peritrophic-like dense layer of worms(similar to mucus)and,thus,the absorption of insulin.

This study investigates the suitability of statistical techniques for evaluating the fluoride content and the groundwater quality from Robles Department(RD)and Banda Department(BD)in santiago del Estero(Argentina).For the original statistical study,evaluation of nine parameters(fluoride,pH,conductivity,atmospheric and water temperature,total dissolved solids,chloride,hardness,and alkalinity)of 110 collected underground water samples from 23 dispersed rural areas was *** samples were obtained by sampling taken from wells at different *** levels were determined by a standard colorimetric method in two seasonal periods,the dry(from April to september)and rainy(from October to March)*** analytical results obtained for physicochemical parameters such as pH,total dissolved solids(TDs),and temperature does not reveal any notable difference between the rainy and dry seasons *** both seasons,the atmospheric temperature average was 22℃.With respect to fluoride content,approximately 50%of the analysed groundwater samples exceeded the limit established by current legislation(1.0 mg/L),obtaining concentration levels in the range of 0.01-2.80 mg/*** study demonstrates the usefulness of the univariate statistical method(quartiles calculation,interquartile range IQR),multivariate principal component analysis(PCA),and cluster analysis to establish a better understanding of the state of the contamination of the waters in the region studied.

Objective: To identify the changes in the proteome of U937 cells infected with dengue virus(DENV).Methods: In this study, differentiated U937 cultures were infected with two DENV-2strains, one of which was associated with dengue(DENV-2/NG) and the other one with severe dengue(DENV-2/16681), with the aim of determining the cellular proteomic profiles under different infection conditions. Cellular proteins were extracted and separated by two-dimensional electrophoresis, and those proteins with differential expression profiles were identified by mass spectrometry. The obtained results were correlated with cellular viability, the number of infectious viral particles, and the viral DNA/protein ***: In comparison with non-infected cultures, in the cells infected with the DENV-2/NG strain, nine proteins were expressed differentially(five were upregulated and four were downregulated); in those cultures infected with the DENV-2/16681 strain, six proteins were differentially expressed(two were downregulated and four were upregulated). The downregulated proteins included fatty acid-binding protein, heterogeneous nuclear ribonucleoprotein 1, protein disulfide isomerase, enolase 1, heat shock 70 k Da protein 9, phosphotyrosyl phosphatase, and annexin IV. The upregulated proteins included heat shock 90 k Da protein AA1, tubulin beta, enolase 1, pyruvate kinase,transaldolase and phospholipase ***: Because the monocyte/macrophage lineage is critical for disease pathogenicity, additional studies on these proteins could provide a better understanding of the cellular response to DENV infection and could help identify new therapeutic targets against infection.

Tailing ponds pose environmental hazards, such as toxic metals which can contaminate the surroundings through wind and water erosions and leaching. Various chemical and biochemical properties, together with extractable and soluble metals were measured five years after reclamation of a polluted soil affected by former mining activities. This abandoned mine site contains large amounts of Fe-oxyhydroxides, sulphates, and heavy metals. As a consequence, soils remain bare and the soil organic matter content is low （〈 3 g kg-1）. Marble waste, pig manure and sewage sludge were applied in 2004. Plant cover and richness, and soil chemical, biochemical and biological parameters were analysed five years later. Results showed that all soil biochemical properties as well as vegetation cover and richness were higher in treated soils than in the untreated contaminated plots （control）, although organic matter, pH values and extractable metals concentrations were similar among treatments. soluble cadmium and zinc were lower in the amended plots than in control.
