Object.ves Scope Due t. t.e plast.cit., macrophage as a het.rogeneous populat.on of immune cells could modify t.eir phenot.pe rapidly as response t. environment.l fact.rs. We different.at.d primary macrophages t. classic macrophages or alt.rnat.ve macrophages and analysed cell t.pespecific markers as well as t.ose markers t.at.response t. diet.ry fibers. Some of t.ese fibers for example given t.e bet.-glucan Lent.nan from Shiit.ke mushroom, have shown immunomodulat.ng propert.es t.wards macrophages whichmight.be expect.d t. be revealed by t.ese analysis. Met.ods Macrophages were obt.ined aft.r 7 days cult.ring of monocyt.s in RPMI-1640 medium supplement.d wit. 50 ng/ml MCSF(referred t. as M(MCSF) and followed polarizat.on by 20 ng/ml t.F-α plus 20 ng/ml IFN-γ or 20 ng/ml IL-4 for M(t.F-α + IFN-γ) and M(IL4) separat.ly. Gene expression profiles of macrophages were analysed by Affymet.ix GeneC hips(Human gene 1.1 St.Array) and verified mR NA expression by q ***,M(MCSF)were incubat.d wit. Lent.nan cont.ining Shiit.ke ext.act.at.500μg/ml for t.e same 18 hours. Result. Comparing M(t.F-α + IFN-γ) t. M(IL4), we ident.fied 8639 significant.differencially expressing genes. According t. t.ose genes, IDO1, IRG1, ANKRD22, GBP5 and GBP3 were t.e t.p 5 markers for M(t.F-α + IFN-γ). Meanwhile, IL17 RB, ALOX5, CD180, St.SIA6 and CHDH were t.e t.p 5 markers for M(IL4). Int.rest.ngly, indoleamine 2,3-dioxygenase(IDO) is an immunomodulat.ry prot.in expressed in various ant.gen present.ng cells such as macrophages and dendrit.c cells, which can suppress adapt.ve immunit. and showed t.cells suppression and immune t.lerance act.vit.. Furt.er, we verified IDO1 m RNA expression by q PCR, normalized t. medium cont.ol, which confirmed t.at.M(t.F-α + IFN-γ)highly express IDO1 while M(IL4) do not. IDO1 expression was also increased by incubat.on wit. t.eL ent.nancont.ining ext.act.of Shiit.ke mushroom.. Conclusion Our findings indicat. t.at.IDO1 is a pot.nt.al biomarker for ident

目的评估在医学中心进行盆腔超声(US)检查的妇女中不明确囊肿的发生率,确定其恶性率,并分析囊肿与病人的特征,用以预测恶性肿瘤可能。材料与方法该研究是2008年1月—2012年6月期间进行的一项多中心研究,对US发现的性质不明附件囊肿进行随访(包括手术病理检查、影像学和/或临床检查)。根据现有文献中的定义,将性质不明确的囊肿分为1类(典型良性囊肿>5 cm)或2类(囊肿含有非血管的实性成分)。计算肿瘤和恶性肿瘤的发生率。评估与肿瘤、恶性肿瘤相关的病人特征和囊肿特征,采用卡方检验或Fisher确切概率检验计算分类变量,并采用t.验分析连续变量。利用逐步Logist.c回归模型比较2种结果:(a)肿瘤(良性或恶性)和(b)恶性肿瘤。结果超声检查发现1 637例女性病人有附件囊肿,391例[年龄17～91岁,平均(41.8±13.5)岁]附件囊肿性质不明确。性质不明确附件囊肿的患病率为23.9%(391/1 637;95%CI:0.22～0.26)。对其中280例女性[年龄17～88岁,平均(42.9±14.1)岁]的303个不明确囊肿进行了充分随访。卵巢肿瘤(良、恶性)的发生率为24.8%(75/303,95%CI:0.20～0.30),恶性肿瘤的发生率为3.6%(11/303;95%CI:0.02～0.06)。卵巢肿瘤的比例在1类和2类囊肿之间有差异[17.5%,(1类143个囊肿中,肿瘤有25个,95%CI:0.12～0.25)与31.3%(2类160个囊肿中,肿瘤有50个;95%CI:0.24～0.39,P=0.001)]。恶性肿瘤的发生率在1、2类的比例分别为0(143个囊肿中有0个)与6.9%(160个囊肿中有11个;95%CI:0.03～0.12);在逐步逻辑回归分析中,囊肿内非血管实性成分的存在是恶性肿瘤的特征表现(优势比=2.83;P<0.000 1;95%CI:1.69～4.70)。结论对于性质不明确的附件囊肿,囊肿内非血管实性成分的存在是恶性肿瘤的特征表现。第2类囊肿与第1类囊肿相比,恶性风险更高。
