Objectives Scope Due to the plasticity, macrophage as a heterogeneous population of immune cells could modify their phenotype rapidly as response to environmental factors. We differentiated primary macrophages to classic macrophages or alternative macrophages and analysed cell typespecific markers as well as those markers that response to dietary fibers. Some of these fibers for example given the beta-glucan Lentinan from Shiitake mushroom, have shown immunomodulating properties towards macrophages whichmight be expected to be revealed by these analysis. methods macrophages were obtained after 7 days culturing of monocytes in RPmI-1640 medium supplemented with 50 ng/ml mCSF(referred to as m(mCSF) and followed polarization by 20 ng/ml TNF-α plus 20 ng/ml IFN-γ or 20 ng/ml IL-4 for m(TNF-α + IFN-γ) and m(IL4) separately. Gene expression profiles of macrophages were analysed by Affymetrix GeneC hips(Human gene 1.1 ST Array) and verified mR NA expression by q ***,m(mCSF)were incubated with Lentinan containing Shiitake extract at 500μg/ml for the same 18 hours. Results Comparing m(TNF-α + IFN-γ) to m(IL4), we identified 8639 significant differencially expressing genes. According to those genes, IDO1, IRG1, ANKRD22, GBP5 and GBP3 were the top 5 markers for m(TNF-α + IFN-γ). meanwhile, IL17 RB, ALOX5, CD180, ST8SIA6 and CHDH were the top 5 markers for m(IL4). Interestingly, indoleamine 2,3-dioxygenase(IDO) is an immunomodulatory protein expressed in various antigen presenting cells such as macrophages and dendritic cells, which can suppress adaptive immunity and showed T cells suppression and immune tolerance activity. Further, we verified IDO1 m RNA expression by q PCR, normalized to medium control, which confirmed that m(TNF-α + IFN-γ)highly express IDO1 while m(IL4) do not. Impact of patch size on woody tree species richness and abundance in a tropical montane evergreen forest patches of south India

We examined the effects of forest patch size on woody tree species richness and abundance in tropical montane evergreen forest patches of the Nilgiri region,south India. We sampled woody trees(>1 cm dbh) from 21 forest patches in the upper Nilgiri hills(> 2000 m elevation) and recorded a total of 35,146 individuals of 61 species, 45 genera and 30 families. Species richness and abundance of sapling/shrubs(≥ 1 to m dbh)increased significantly with increasing patch size, but the species richness and abundance of small, medium and larger trees(≥ 10 to m dbh,respectively) did not. Overall, forest interior species richness and abundance increased significantly with increasing patch size but edge species richness did not. Species richness and abundance of shade-tolerant and shade-demanding tree species also increased with increasing patch size. The abundance of zoochory dispersed tree species was significantly related to increasing patch size, but those dispersed by autochory did not display any clear relationship between patch size and species richness or *** findings suggest that with increasing forest patch area,tree compositional patterns may be driven by species specific shade-tolerance adaptations and dispersal *** responses in these traits by the plant community within the individual habitat zones of forest edge and interiors likely plays a major role in determining the inherent plant community and thus the subsequent ecological processes of forest patches, including their responses to increasing patch area.

Arabinan metabolism during Seed Development and Germination in Arabidopsis

Arabinans are found in the pectic network of many cell walls, where, along with galactan, they are present as side chains of Rhamnogalacturonan I. Whilst arabinans have been reported to be abundant polymers in the cell walls of seeds from a range of plant species, their proposed role as a storage reserve has not been thoroughly investigated. In the cell walls of Arabidopsis seeds, arabinose accounts for approximately 40% of the monosaccharide composition of non- cellulosic polysaccharides of embryos. Arabinose levels decline to -15% during seedling establishment, indicating that cell wall arabinans may be mobilized during germination. Immunolocalization of arabinan in embryos, seeds, and seedlings reveals that arabinans accumulate in developing and mature embryos, but disappear during germination and seedling establishment. Experiments using 14C-arabinose show that it is readily incorporated and metabolized in growing seedlings, indicating an active catabolic pathway for this sugar. We found that depleting arabinans in seeds using a fungal arabinanase causes delayed seedling growth, lending support to the hypothesis that these polymers may help fuel early seedling growth.

An overview of the Czech national R&D project HiLASE(High average power pulsed laser) is presented. The project focuses on the development of advanced high repetition rate, diode pumped solid state laser(DPSSL) systems with energies in the range from mj to 100 j and repetition rates in the range from 10 Hz to 100 kHz. Some applications of these lasers in research and hi-tech industry are also presented.

Background:The incorporation of legumes,specifically alfalfa(medicago sativa L.),into bermudagrass(Cynodon spp.)-based pasture systems in the southeastern United States has increased in recent years as an alternative to synthetic nitrogen(N)***:A small plot evaluation was conducted in Shorter,Alabama,and Tifton,Georgia,USA,to evaluate the impact of harvest height(HH)and harvest frequency(HF)on agronomic characteristics of alfalfa+bermudagrass mixtures in southeastern United ***:Results from both locations revealed that the longer the HF and the shorter the HH,the greater the alfalfa retention was in the stand(pmpact any of the reported nutritive value parameters,while longer HF resulted in lower total digestible nutrients,lower crude protein,higher acid detergent fiber,and lower 48 h in vitro dry matter digestibility(pmpacted forage accumulation at both locations(pmed recent findings from comparable evaluations in the southeastern United States,in that increasing HH and decreasing HF improved alfalfa retention while having a negligible effect on nutritive value or forage accumulation.
