Human idiopathic hypercalciuria(IH)is the most common cause of calcium oxalate nephrolithiasis with perturbed calcium metabolism with increased bone resorption and decreased renal calcium reabsorption,which can be phenotype-copied in the genetic hypercalciuric stone-forming(GHS)rat *** previously demonstrated that high VdR expression plays important roles in the development of hypercalciuria in the GHS ***,the underlying mechanism through which VdR impact hypercalciuria development remains to be fully ***,we sought to determine how VdR regulated its target genes that are implicated in calcium homeostasis and potentially *** found that VdR expression in the GHS rats was elevated in the calcium transporting tissues,as well as in the thymus and prostate,but not in lung,brain,heart,liver and spleen,when compared with control Sd *** expression in the GHS rats was significantly downregulated in kidney,intestine,thymus and *** injection of 1,25(OH)2d3 significantly upregulated the expression of renal calcium sensing receptor(CaSR),intestinal calcium transporters transient receptor potential vanilloid type 6(TRPV6),and VdR in GHS rats,compared with that in control Sd *** assays revealed that VdR specifically bound to the proximal promoters of target genes,followed by histone H3 hyperacetylation or ***,our results suggest that elevated VdR expressi on may con tribute to the development of hypercalciuria by sensi・tizing VdR target genes to 1,25(OH)2d3 through histone modifications at their promoter regions in a genetic hypercalciuric stone-forming(GHS)rat model.

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.

Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express Cd90, Cd105, Cd73 but not Cd45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

Plants require a number of essential elements in different proportions for ensuring their growth and *** elemental concentrations in leaves reflect the functions and adaptations of plants under specific environmental ***,less is known about how the spectrum of leaf elements associated with resource acquisition,photosynthesis and growth regulates forest biomass along broad elevational *** examined the influence of leaf element distribution and diversity on forest biomass by analyzing ten elements(C,N,P,K,Ca,Mg,Zn,Fe,Cu,and Mn)in tree communities situated every 100 meters along an extensive elevation gradient,ranging from the tropical forest(80 meters above sea level)to the alpine treeline(4200 meters above sea level)in the Kangchenjunga Landscape in eastern Nepal *** calculated communityweighted averages(reflecting dominant traits governing biomass,i.e.,mass-ratio effect)and functional divergence(reflecting increased trait variety,i.e.,complementarity effect)for leaf elements in a total of 1,859 trees representing 116 *** increasing mass-ratio effect and decreasing complementarity in leaf elements enhance forest biomass accumulation.A combination of elements together with elevation explains biomass(52.2%of the variance)better than individual elemental trait diversity(0.05%to 21%of the variance).Elevation modulates trait diversity among plant species in biomass *** promotes biomass at lower elevations,but reduces biomass at higher elevations,demonstrating an interaction between elevation and *** interaction between elevation and mass-ratio effect produces heterogeneous effects on biomass along the elevation *** research indicates that biomass accumulation can be disproportionately affected by elevation due to interactions among trait diversities across vegetation *** higher trait variation enhances the adaptation of species to environmental changes,it reduces

Most vascular plants interact with arbuscular mycorrhizal fungi （AMF） and are thereby provided significant advantages in nutrient acquisition, especially phosphate. The widespread retention of this symbiosis in vascular plants is testament to its importance and ancient origins. This highlight focuses on the reports of two of the first genes identified through for- ward genetic screens for AMF symbiotic mutants： Required for Abuscular Mycorrhization 1 （RAM1） and RAM2, which respectively encode a GRAS transcription factor and a glyc- erol-3-phosphate acyl-transferase （GPAT） that are required for the formation of fungal entry structures （hyphopodia） on the root surface.

Agglomerate behavior in a recirculating fluidized bed with sheds： Effect of ring baffles

The radioactive particle tracking technique was used to study the effect of internal ring baffles on wet agglomerate motion inside a cold flow recirculating fluidized bed. The study found that using such a baffle on its own or above the regular sheds helps reduce the fouling of the stripper section by increasing the residence time that the agglomerates spend above the baffle, thereby reducing the release of the vapors below the baffles that cause fouling of the sheds. Adding down-comers, or flux tubes, to the ring baffles degrades the performance of the baffles, Reducing the length of the flux tubes, so that they do not reach the bottom of the baffle lip results in a further degradation in baffle performance.

Modulation of human global/local perception by low spatial frequency filtering

We investigated the effect of low spatial fre-quency (SF) filtering on neural substrates underlying global and local processing in the peripheral vision by measuring hemodynamic responses with functional magnetic resonance imaging (fMRI). Subjects identified global or local shapes of compound letters that were either broadband in spatial- frequency spectrum or contrast balanced (CB) to removed low SFs and displayed randomly in the left or right visual fields. Attention to both broadband and CB global shapes generated stronger activation over the medial occipital cor-tex relative to local attention. Lateralized activations in association with global processing were observed over the right temporal-parietal junction for broadband stimuli whereas over the right fusiform gyrus for CB stimuli. Atten-tion to CB local shapes resulted in activations in the medial frontal cortex, bilateral inferior frontal and superior tempo-ral cortices. The results were discussed in terms of the competition between global and local information in determining brain activations in association with global/local processing of compound stimuli.

TCR-pMHC: may the force be of you?

Theα/βcomponent of the T-cell antigen receptor(TCR)is a transmembrane heterodimer containing genetically variable(V)domains and constant domains within each polypeptide chain.1 The Vαand Vβdomains bind with a clonally unique(clonotypic)interface angle,2 and the complementary determining regions(CdR1,CdR2,and CdR3)manifest as‘loops’between anti-parallelβ-strands of the overall sandwich ofβ-sheets with a central disulfide *** with immunoglobulins,the CdR1 and CdR2 loops are encoded in the germline via the particular V-region dNA segment involved in the RAG1/RAG2 recombination mechanism responsible for somatic construction,together with the d-and/or j-segment,of the third loop,CdR3.1,3 As shown by multiple solved X-ray crystallography structures,the CdRs contain the closest amino acid(a.a.)contacts with the peptide(p)plus the major histocompatibility complex(MHC)protein(together abbrev.,pMHC)composite *** rigorous selection in the thymus for only those TCRs that should function properly in the particular individual after birth(MHC restriction),4 T cells function clonally as well,i.e.,different T-cell clones display differential responses to not only different pMHC ligands but also perhaps more importantly,to the same pMHC ligands.5,6 A long-standing question is how(not whether)TCR V-domain binding to pMHC is ultimately responsible for such vast biological outcomes as protection *** to infectious diseases,atopy,the onset of autoimmune diseases,and the success of various types of transplantation.

AIM: To investigate the function of NOd2 in colonic epithelial cells (CEC). METHOdS: A combination of in vivo and in vitro analyses of epithelial cell turnover in the presence and absence of a functional NOd2 protein and, in response to enteric Salmonella typhimurium infection, were used. shRNA interference was also used to investigate the consequences of knocking down NOd2 gene expression on the growth and survival of colorectal carcinoma cell lines. RESULTS:In the colonic mucosa the highest levels of NOd2 expression were in proliferating crypt epithelial cells. Muramyl dipeptide (MdP), that is recognized by NOd2, promoted CEC growth in vitro . By contrast,the growth of NOd2-deficient CECs was impaired. In vivo CEC proliferation was also reduced and apoptosis increased in Nod2-/- mice, which were also evident following enteric Salmonella infection. Furthermore, neutralization of NOd2 mRNA expression in human colonic carcinoma cells by shRNA interference resulted in decreased survival due to increased levels of apoptosis. CONCLUSION: These findings are consistent with the involvement of NOd2 protein in promoting CEC growth and survival. defects in proliferation by CECs in cases of Cd may contribute to the underlying pathology of disrupted intestinal homeostasis and excessive inflammation.
