Online single-session interventions for Kenyan adolescents:study protocol for a comparative effectiveness randomised controlled trial

Background Mental health problems are the leading cause of disability among adolescents worldwide,yet access to treatment is *** digital interventions have been shown to improve youth mental health,but little is known about which digital interventions are most *** To evaluate the effectiveness of two digital single-session interventions(Shamiri-Digital and Digital-CBT(cognitive-behavioural therapy))among Kenyan *** We will perform a school-based comparative effectiveness randomised controlled *** 926 Kenyan adolescents will be randomly assigned to one of three conditions:Shamiri-Digital(focused on gratitude,growth mindsets and values),Digital-CBT(focused on behavioural activation,cognitive restructuring and problem solving)or a study-skills control condition(focused on note-taking and essay writing skills).The primary outcomes include depressive symptoms(measured by the patient Health Questionnaire-8),anxiety symptoms(Generalized Anxiety Disorder Screener-7)and subjective well-being(Short Warwick-Edinburgh Mental Well-being Scale).The secondary outcomes include acceptability,appropriateness,primary control and secondary *** and appropriateness will be measured immediately post-intervention;other outcomes will be measured 2 weeks,4 weeks and 12 weeks *** We hypothesise that adolescents assigned to Shamiri-Digital and adolescents assigned to Digital-CBT will experience greater improvements(assessed via hierarchical linear models)than those assigned to the control *** will also compare Shamiri-Digital with Digital-CBT,although we do not have a preplanned *** Our findings will help us evaluate two digital single-session interventions with different theoretical *** effective,such interventions could be disseminated to reduce the public health burden of common mental health problems.

Hydrogen sulfide intervention in cystathionine-β-synthase mutant mouse helps restore ocular homeostasis

AiM: To investigate the applications of hydrogen sulfide(H2 S) in eye-specific ailments in ***: Heterozygous cystathionine-β-synthase(CBS+/-) and wild-type C57 BL/6 j(WT) mice fed with or without high methionine diet(HMD) were administered either phosphate buffered saline(pBS) or the slow-release H2 S donor: GYY4137. Several analyses were performed to study GYY4137 effects by examining retinal lysates for key protein expressions along with plasma glutamate and glutathione estimations. intraocular pressure(iOp) was monitored during GYY4137 treatment; barium sulfate and bovine serum albumin conjugated fluorescein isothiocyanate(BSA-FiTC) angiographies were performed for examining vasculature and its permeability post-treatment. Visionguided behavior was also tested employing novel object recognition test(NORT) and light-dark box test(LDBT) ***: CBS deficiency(CBS+/-) coupled with HMD led disruption of methionine/homocysteine(Hcy) metabolism leading to hyperhomocysteinemia(HHcy) in CBS+/-mice as reflected by increased Hcy, and s-adenosylhomocysteine hydrolase(SAHH) levels. Unlike CBS, cystathionine-γ lyase(CSE), methylenetetrahydrofolate reductase(MTHFR) levels which were reduced but compensated by GYY4137intervention. Heightened oxidative and endoplasmic reticulum(ER) stress responses were mitigated by GYY4137 effects along with enhanced glutathione(GSH) levels. increased glutamate levels in CBS+/-strain were prominent than WT mice and these mice also exhibited higher iOp that was lowered by GYY4137 treatment. CBS deficiency also resulted in vision-guided behavioral impairment as revealed by NORT and LDBT findings. interestingly, GYY4137 was able to improve CBS+/-mice behavior together with lowering their glutamate levels. Blood-retinal barrier(BRB) appeared compromised in CBS+/-with vessels' leakage that was mitigated in GYY4137 treated group. Over the past one year and three months, 150 out patients with COViD 19 have been treated with early initiation of Oseltamevir 75 mg × 2 [150 mg] twice daily, with azithromycin, cephalosporin [to give atypical bacterial cover, gram positive bacterial cover, respectively] enabling cure. Corona virus pandemic was enabled by global contraception/abortion [non evidence based non medical practice] with its blood/necrotic tissue environmental pollution-documented by rising environmental oestrogen/alpha feto protein/beta human chorionic gonadotropin. Global contraception abortion with its blood, necrotic tissue environmental pollution provides a rich environmental media for Corona virus-a fastidious organism to become pandemic, with recurrent mutations. Guaranteed elimination of this fastidious organism-corona virus [and other microbes like Ebola, Nipah…] by implementing many childbirths per mother [earth bus fare/ticket] as God ordained abolition of global contraception, abortion, one child policy/small family norms [jumping off cliff to fly-marred science] with its blood/necrotic tissue pollution, mandatory reversal of contraception. Steroids are deleterious in any viral infection, being immunosuppressants-secondary mucormycosis, other fungal [less virulent] infections can follow;corona virus itself can mutate, gain virulence, drug resistance;but prior contraception-destroying germ cells, prior abortion [destruction of tissue] in the victim host result in auto-immunity of corresponding gradations including fatal HLH [Hemophagocytic lymphohistiocytosis], cytokine storm, requiring immunomodulation with plasmapheresis, steroids, hydroxychloroquine without much avail.
