Environmental control of the alcohol dehydrogenase (Adh) and other stress response genes in plants is in part brought about by transcriptional regulation involving the G-box cis-acting DNA element and bZIP G-box Binding Factors (GBFs). The mechanisms of GBF regulation and requirements for additional factors in this control process are not well understood. In an effort to identify potential GBF binding and control partners, maize GBF1 was used as bait in a yeast two-hybrid screen of an A. thaliana cDNA library. GBF Interacting Protein 1 (GIP1) arose from the screen as a 496 amino acid protein with a predicted molecular weight of 53,748 kDa that strongly interacts with GBFs. Northern analysis of A. thaliana tissue suggests a 1.8-1.9 kb GIP1 transcript, predominantly in roots. Immunolocalization studies indicate that GIP1 protein is mainly localized to the nucleus. In vitro electrophoretic mobility shift assays using an Adh G-box DNA probe and recombinant A. thaliana GBF3 or maize GBF1, showed that the presence of GIP1 resulted in a tenfold increase in GBF DNA binding activity without altering the migration, suggesting a transient association between GIP1 and GBF. Addition of GIP1 to intentionally aggregated GBF converted GBF to lower molecular weight macromolecular complexes and GIP1 also refolded denatured rhodanese in the absence of ATP. These data suggest GIP1 functions to enhance GBF DNA binding activity by acting as a potent nuclear chaperone or crowbar, and potentially regulates the multimeric state of GBFs, thereby contributing to bZIP-mediated gene regulation.

尽管学校的领导技能水平符合区、县及学校层面对学生学业成绩所设目标的落实要求,但教育领导者同时还影响着学校教师的教学效果。本研究以领导技能对教师教学效果的影响为研究课题。在姆万扎区,教师的教学效果不容乐观,其前景也并不明朗。本研究采用定性研究的方法,以揭示领导技能在提升教师教学效果方面的应用现状。同时,本研究采用归纳和探索性研究设计,采用随机抽样方法确定研究对象。本研究从1326名教师和校长为母体的研究对象中抽取了其中的9.8%来分析。本研究涉及32名学校校长和98名中学教师,共计130名受访者。本研究涉及32所随机抽选的中学,每所中学有10%的教师参与了此次研究。本研究采用定性研究方法,通过访谈、焦点小组讨论和问卷调查的形式收集所需信息。研究数据的呈现、分析、阐释及讨论以两个章节进行分述。第四章主要分析了学校校长和坦桑尼亚国家考试委员会(NECTA)的统计数据以及有关学生学业成绩的各方数据,而这些成绩是对教师教学效果的反映;第五章则主要讨论了收集自焦点小组讨论和访谈对话的信息。此外,本研究采用描述性和探索性的内容分析方法进行数据分析,数据内容以表格形式呈现。本研究认为,在提高中学教师的教学效果方面,人事、概念、技术和专业领导技能没有得到合适有效的应用。在提高教学效果方面,中学教师高度依赖概念、人事、技术和专业领导技能,而不是经验和过往背景。这并不意味着除了受到学校领导的关注以外,领导技能就无益于提高姆万扎地区教师的教学效果,而是领导技能未能有效地用于改善教师的教学效果。因此,本研究建议对人事,技术,专业和概念领导技能进行有效应用。通过问卷调查、访谈和焦点小组讨论,受访者主要提出了四种针对学校校长和教师的领导技能培养方法,分别是在职培训、向教学效果有所提升的教师提供激励性补偿、制定领导技能政策和为教师创造有利的工作环境。上述技能将用于提高教师的教学效果,而提高教学效果的途径便是使学生取得优异成绩。教师的优异教学效果体现于学生的I、II、III级得分,教师的不佳教学效果则体现于学生的IV和0级得分。本研究以Ketz(1995)的技能理论和Mumford的领导技能模型(2000)为理论基础。这两种理论与本研究所提出的四种学校领导技能(人事、概念、技术和专业)直接相关。学校校长及教育领导者需共同运用这四种领导技能,以使教师的教学效果提高至最佳水平。但是,由于所管教师的背景和秉性不同,在运用领导技能时,教育领导者若取其一而舍其他,则可能无法实现预期效果。

Overview of surface ozone variability in East Asia-North Pacific region during IGAC/APARE(1994—1996)

Surface ozone(O\-3) was measured at Oki Island(japan), Cheju Island(South Korea), Lanyu Island(Taiwan Province, China), Cape D'Aguilar(Hong Kong SAR) and Lin'an, Longfenshan, Waliguan(China mainland) during january 1994—December 1996 as a component of IGAC/APARE(International Global Atmospheric Chemistry/East Asia-North Pacific Regional Experiment). This paper gave a joint discussion on the observational results at these stations over the study region. Investigations showed that the average of surface O 3 mixing ratios at the seven sites are 47.9±15.8, 48.1±17.9, 30.2±16.4, 31.6±17.5, 36.3±17.5, 34.8±11.5 and 48.2±9.5 ppbv, respectively. Significant diurnal variations of surface O 3 have been observed at Oki, Cheju, D’Aguilar, Lin'an and Longfenshan. Their annual averaged diurnal differences range from 8 to 23 ppbv and differ in each season. Surface O 3 at Lanyu and Waliguan do not show strong diurnal variability. Seasonal cycles of surface O 3 showed difference at the temperate and the subtropical remote sites. Oki has a summer minimum-spring maximum, while Lanyu has a summer minimum-autumn maximum. The suburban sites at D'Aguilar and Lin'an report high-level O 3 in autumn and low level O 3 in summer. Surface O 3 remains high in autumn and low in winter at the rural site Longfenshan. For the global background station Waliguan, surface O 3 exhibits a broad spring-summer maximum and autumn-winter minimum. The backward air trajectories to these sites have shown different pathways of long-range transport of air pollution from East Asia Continent to North Pacific Ocean. Surface O 3 was found to be strongly and positively correlated with CO at Oki and Lanyu, especially in spring and autumn, reflecting the substantial photochemical buildup of O\-3 on a regional scale. It is believed that the regional sources of pollution in East Asia have enhanced the average surface O 3 concentrations in the background atmosphere of North Pacific.

Heavy Metal Dispersion in Stream Sediments in River Iyiudene, Abakaliki South-Eastern Nigeria: Source, Distribution Pattern, and Contamination Assessment

River Iyiudene is a vital distributary resource in Abakaliki, southeastern Nigeria and conveys an abundant amount of sediments to provincial and residual ecosystems. Although the importance of the river cannot be overemphasized, the geochemistry of its stream sediments is less investigated. Twenty (20) stream sediment samples were taken at the centre of the river channels to represent the entire drainage area well and avoid collapsed bank materials. The stream sediment samples were used to determine the dispersion, contamination status and sources of heavy metal concentrations. Total elemental digestion accompanied this with the use of aqua regia, an admixture of Hydrochloric acid (HCl) and Nitric acid (HNO3) in the ratio of 3:1 using the atomic absorption spectrometer (AAS). The heavy metal concentration levels in River Iyiudene were low compared with sediments from Imo River, Gulf of California, Upper continental crust, Average shale and surface horizons, excluding Cd, which showed high concentration levels than the other reference studies. The results delineated a wide contrast in the concentration levels of the heavy metals, with the mean contents in the order Zn > Cu > Pb > Cd > Ni > As. The pollution evaluation utilizing the Effect range low (ERL), Effect range median (ERM), single pollution index, and geo-accumulation index revealed Cd contamination. This study indicates that the heavy metals were sourced from the natural geological background of the river basin and possibly from agricultural runoff and atmospheric pollutants.

Dispersion remains an enduring challenge for the characterization of wavelength-dependent transmission through optical multimode fiber(MMF).Beyond a small spectral correlation width,a change in wavelength elicits a seemingly independent distribution of the transmitted *** we report on a parametric dispersion model that describes mode mixing in MMF as an exponential map and extends the concept of principal modes to describe the fiber's spectrally resolved transmission matrix(TM).We present computational methods to fit the model to measurements at only a few,judiciously selected,discrete *** validate the model in various MMF and demonstrate an accurate estimation of the full TM across a broad spectral bandwidth,approaching the bandwidth of the best-performing principal modes,and exceeding the original spectral correlation width by more than two orders of *** model allows us to conveniently study the spectral behavior of principal modes,and obviates the need for dense spectral measurements,enabling highly effcient reconstruction of the multispectral TM of MMF.

Vitamin D deficiency in cirrhosis relates to liver dysfunction rather than aetiology

AIM: To examine the vitamin D status in patients with alcoholic cirrhosis compared to those with primary biliary cirrhosis. METHODS: Our retrospective case series comprised 89 patients with alcoholic cirrhosis and 34 patients with primary biliary cirrhosis who visited our outpatient clinic in 2005 and underwent a serum vitamin D status assessment. RESULTS: Among the patients with alcoholic cirrhosis, 85% had serum vitamin D levels below 50 nmol/L and 55% had levels below 25 nmol/L, as compared to 60% and 16% of the patients with primary biliary cirrhosis, respectively (P < 0.001). In both groups, serum vitamin D levels decreased with increasing liver disease severity, as determined by the Child-Pugh score. CONCLUSION: Vitamin D deficiency in cirrhosis relates to liver dysfunction rather than aetiology, with lower levels of vitamin D in alcoholic cirrhosis than in primary biliary cirrhosis.

Osmotic stress caused by drought and high salinity is a significant environmental threat that limits plant growth and agricultural yield. Redox regulation plays an important role in plant stress responses, but the mechanisms by which plants perceive and transduce redox signals are still underexplored. Here, we report a critical function for the thiol peroxidase GPX1 in osmotic stress response in rice, where it serves as a redox sensor and transducer. GPX1 is quickly oxidized upon exposure to osmotic stress and forms an intramolecular disulfide bond, which is required for the activation of bZIP68, a VRE-like basic leucine zipper (bZIP) transcription factor involved in the ABA-independent osmotic stress response pathway. The disulfide exchange between GPX1 and bZIP68 induces homo-tetramerization of bZIP68 and thus positively regulates osmotic stress response by regulating osmotic-responsive gene expression. Furthermore, we discovered that the nuclear translocation of GPX1 is regulated by its acetylation under osmotic stress. Taken together, our findings not only uncover the redox regulation of the GPX1-bZIP68 module during osmotic stress but also highlight the coordination of protein acetylation and redox signaling in plant osmotic stress responses.

The integrated use of chemical insecticides and the entomopathogenic nematode, Steinemema carpocapsae （Nematoda： Steinernematidae）, for the control of sweetpotato whitefly, Bemisia tabaci （Hemiptera： Aleyrodidae）

The integration of chemical insecticides and infective juveniles of the entomopathogenic nematode Steinernema carpocapsae （Wesier）（Nematoda： Steinernematidae）, to control second instars of the sweetpotato whitefly, Bemisia tabaci Gennadius （Hemiptera： Aleyrodidae） was investigated. Using a sand bioassay, the effects of direct exposure of S. carpocapsae for 24 h to field rate dilutions of four insecticides （spiromesifen, thiacloprid, imidacloprid and pymetrozine） on infectivity to Galleria mellonella larvae were tested. Although all chemicals tested, except spiromesifen, produced acceptable nematode infectivity rates, they were all significantly less than the water control. The effect of insecticide treatment （dry residues of spiromesifen, thiacloprid and pymetrozine and soil drench of imidacloprid） on the efficacy of the nematode against B. tabaci was also investigated. Nematodes in combination with thiacloprid and spiromesifen gave higher B. tabaci mortality （86.5% and 94.3% respectively） compared to using nematodes alone （75.2%） on tomato plants. There was no significant difference in B. tabaci mortality when using the chemicals imidacloprid, pymetrozine and spiromesifen in conjunction with nematodes compared to using the chemicals alone. However, using thiacloprid in combination with the nematodes produced significantly higher B. tabaci mortality than using the chemical alone. The integration of S. carpocapsae and these chemical agents into current integrated pest management programmes for the control of B. tabaci is discussed.

Land use changes profoundly affect the equilibrium of soil organic carbon(soC)sequestration and greenhouse gas *** the current global climatic changes,it is vital to understand the influence of ecological restoration and conservation management on the dynamics of soC under different land uses,especially in erosion-endangered Loess ***,we investigated changes in soc through a suit of labile fractions,namely:light fraction organic C(LFOC),heavy fraction organic C(HFOC),coarse particulate organic C(CPOC),fine particulate organic C(FPOC),and dissolved organic C(DOC),from two forests i.e.,Robinia pseudoacacia(RP)and Platycladus orientalis(PO),with different ages,in comparison with farmland(FL).The SOC and STN contents significantly increased over 42 years in the RP forest where the contents of CPOC and FPOC were significantly higher than in the ***,total SOC and its labile fractions,in the studied land use types,significantly correlated with soil CacO3,pH,and STN contents,indicating their key roles in SoC *** results reported here from different vegetation with different ages provide a better understanding of sOC and STN alterations at different stages of vegetation *** findings suggest that long-term natural vegetation restoration could be an effective approach for SoC sequestration and soil conservation on the Loess soil.
