Transforming growth factor-beta(TGFβ)is a highly potent immunosuppressive *** TGFβis a tumor suppressor in early/premalignant cancer lesions,the cytokine has several tumor-promoting effects in advanced cancer;abrogation of the antitumor immune response is one of the most important tumor-promoting *** several immunoregulatory mechanisms have recently been shown to be targets of specific T cells,we hypothesized that TGFβis targeted by naturally occurring specific T cells and thus could be a potential target for immunomodulatory cancer ***,we tested healthy donor and cancer patient T cells for spontaneous T-cell responses specifically targeting 3820-mer epitopes derived from TGFβ*** identified numerous CD4^(+)and CD8^(+)T-cell responses against several epitopes in TGFβ.Additionally,several ex vivo responses were *** enriching specific T cells from different donors,we produced highly specific cultures specific to several TGFβ-derived *** CD8^(+)T-cell clones specific for both a 20-mer epitope and a 9-mer HLA-A2 restricted killed epitope peptide were pulsed in HLA-A2+target cells and killed the HLA-A2+cancer cell lines THP-1 and ***,stimulation of THP-1 cancer cells with cytokines that increased TGFβexpression increased the fraction of killed *** conclusion,we have shown that healthy donors and cancer patients harbor CD4^(+)and CD8^(+)T cells specific for TGFβ-derived epitopes and that cytotoxic T cells with specificity toward TGFβ-derived epitopes are able to recognize and kill cancer cell lines in a TGFβ-dependent manner.

Assessing School-Based Water, Sanitation and Hygiene (WASH) Facilities in Peri-Urban Settings of Kinshasa, DR Congo

Background: Kinshasa’s peri-urban settings have a low rate of water access, which has significant consequences for the WASH infrastructures in schools and preventative measures against the spread of waterborne diseases and pathogens. This study aimed to assess the availability, functionality, and gender sensitivity of WASH infrastructures of Kinshasa’s peri-urban schools. Methods: A cross-sectional study was performed in three of the four educational provinces of Kinshasa, targeting 165 peri-urban schools. Data were collected using a questionnaire and an observation grid. Results: An overall proportion of 10.9% of schools possessed a water point, and therefore time-consuming water chores are a necessity in 89.1% of schools. Girl students provided 30% of the labor collecting water during punishments. A total of 98.2% of schools had functional latrines of which 3.6% were found hygienic, associated with water reserve next to the latrines (P = 0.040). Only 2.4% of schools displayed posters raising awareness of latrine hygiene, and 3.6% displayed posters on hand hygiene. The ratios of latrines units for girls were 58:1 for toilets and 115:1 for urinals, justifying open defecation and urination reported in 62.4% of schools. Also, 43% of schools had hand-washing facilities whose functionality was significantly associated with the presence of water points in the school’s inner courtyard (P = 0.032), with water (P P Conclusion: The majority of schools had limited drinking water services, which negatively impact the functionality and gender sensitivity of other WASH provisions. The current evidence as a public health concern would raise government and school authorities’ attention to address these environmental threats.

Background: Hereditary transthyretin(ATTRv) amyloidosis is an autosomal dominant disease linked to transthyretin gene mutations which cause instability of the transthyretin tetramer. After dissociation and misfolding they reassemble as insoluble fibrils(i.e. amyloid). Apart from the common Val30 Met mutation there is a very heterogeneous group of non-Val30 Met mutations. In some cases, the clinical picture is dominated by a rapidly evolving restrictive and hypertrophic cardiomyopathy. Methods: A case series of four liver recipients with the highly clinically relevant, rare and particularly aggressive Val122 del mutation is presented. Medical and surgical therapeutic options, waiting list policy for ATTRv-amyloidosis, including the need for heart transplantation, and status of heart-liver transplantation are discussed. Results: Three patients needed a staged(1 patient) or simultaneous(2 patients) heart-liver transplant due to rapidly progressing cardiac failure and/or neurologic disability. Domino liver transplantation was impossible in two due to fibrotic hepatic transformation caused by cardiomyopathy. After a follow-up ranging from 3.5 to 9.5 years, cardiac(allograft) function was maintained in all patients, but neuropathy progressed in three patients, one of whom died after 80 months. Conclusions: This is the first report in(liver) transplant literature about the rare Val122 del ATTRv mutation. Due to its aggressiveness, symptomatic patients should be prioritized on the liver and, in cases with cardiomyopathy, heart waiting lists in order to avoid the irreversible neurological and cardiac damage that leads to a rapid lethal outcome.

In the past few years there has been a growth in the use of nanoparticles for stabilizing lipid membranes that contain embedded proteins. These bionanoparticles provide a solution to the challenging problem of membrane protein isolation by maintaining a lipid bilayer essential to protein integrity and activity. We have previously described the use of an amphipathic polymer （poly（styrene-co-maleic add）, SMA） to produce discoidal nanoparticles with a lipid bilayer core containing the embedded protein. However the structure of the nanoparticle itself has not yet been determined. This leaves a major gap in understanding how the SMA stabilizes the encapsulated bilayer and how the bilayer relates physically and structurally to an unencapsulated lipid bilayer. In this paper we address this issue by describing the structure of the SMA lipid particle （SMALP） using data from small angle neutron scattering （SANS）, electron microscopy （EM）, attenuated total reflection Fourier transform infrared spectroscopy （ATR-FTIR）, differential scanning calorimetry （DSC） and nuclear magnetic resonance spectroscopy （NMR）. We show that the particle is disc shaped containing a polymer ＂bracelet＂ encircling the lipid bilayer. The structure and orientation of the individual components within the bilayer and polymer are determined showing that styrene moieties within SMA intercalate between the lipid acyl chains. The dimensions of the encapsulated bilayer are also determined and match those measured for a natural membrane. Taken together, the description of the structure of the SMALP forms the foundation for future development and applications of SMALPs in membrane protein production and analysis.

Informing Food Consumption Choices: Innovations in Measuring and Labelling

The need to inform consumers about the health impact of their food choices is ever more pressing in a world where obesity is a growing problem. Concerns over food safety, its origins and its environmental impacts are also growing, as frequently reported in the popular press in many parts of the world. Nutritional and health information on food labels is quite well developed, but the complex nature of the information presented may hinder widespread use of the existing labels. In comparison, there has been little widespread success of carbon labels on food, and their usefulness in reducing carbon emissions from consumption is uncertain. In an attempt to address the need for clearer information on health and environmental impacts of food purchases, we present a novel dual-purpose food labelling system which provides information on both health and environmental impacts of food items. This paper presents results from a pilot study introducing a novel approach to food labelling: a simplified, combined carbon and health label to inform consumers simultaneously about the environmental and health impacts of their choices. Environmental impacts of various food categories were calculated on the basis of their relative energy use along the supply chain by using a newly designed Food Energy Index. Health impacts were based on the NuVal system developed in the USA in 2010. As part of the Norfolk Island Carbon and Health Evaluation study (NICHE), labels were designed, tested and displayed on 25 food categories for a 3-month period in the main supermarket on Norfolk Island (Australia). The in-store labelling trial was followed by a consumer survey on their attitudes to the labels. The results from this pilot trial indicate that consumers were supportive of food labelling including both environmental and health impacts, but the information provided in the dual labels was not sufficient to induce changes in consumption between food categories.
