Parkinsonism by unilateral,intranigralβ-sitosterolβ-D-glucoside administration in rats is distinguished in that theα-synuclein insult begins unilaterally but spreads bilaterally and increases in severity over time,thus replicating several clinical features of Parkinson’s disease,a typicalα-synucleinopathy.As Nurr1 repressesα-synuclein,we evaluated whether unilateral transfected of rNurr1-V5 transgene via neurotensin-polyplex to the substantia nigra on day 30 after unilateralβ-sitosterolβ-D-glucoside lesion could affect bilateral neuropathology and sensorimotor deficits on day 30 post-transfection.This study found that rNurr1-V5 expression but not that of the green fluorescent protein(the negative control)reducedβ-sitosterolβ-D-glucoside-induced neuropathology.Accordingly,a bilateral increase in tyrosine hydroxylase-positive cells and arborization occurred in the substantia nigra and increased tyrosine hydroxylase-positive ramifications in the striatum.In addition,tyrosine hydroxylase-positive cells displayed less senescence markerβ-galactosidase and more neuron-cytoskeleton markerβIII-tubulin and brain-derived neurotrophic factor.A significant decrease in activated microglia(positive to ionized calcium-binding adaptor molecule 1)and neurotoxic astrocytes(positive to glial fibrillary acidic protein and complement component 3)and increased neurotrophic astrocytes(positive to glial fibrillary acidic protein and S100 calcium-binding protein A10)also occurred in the substantia nigra.These effects followed the bilateral reduction inα-synuclein aggregates in the nigrostriatal system,improving sensorimotor behavior.Our results show that unilateral rNurr1-V5 transgene expression in nigral dopaminergic neurons mitigates bilateral neurodegeneration(senescence and loss of neuron-cytoskeleton and tyrosine hydroxylase-positive cells),neuroinflammation(activated microglia,neurotoxic astrocytes),α-synuclein aggregation,and sensorimotor deficits.Increased neurotrophic astrocytes and brain-derived neurotrophic factor can mediate t

Nitric oxide(NO)is an essential reactive oxygen species and a signal molecule in plants.Although several studies have proposed the occurrence of oxidative NO production,only reductive routes for NO production,such as the nitrate(NO_(3)^(-))-upper-reductase pathway,have been evidenced to date in land plants.However,plants grown axenically with ammonium as the sole source of nitrogen exhibit contents of nitrite and NO3−,evidencing the existence of a metabolic pathway for oxidative production of NO.We hypothesized that oximes,such as indole-3-acetaldoxime(IAOx),a precursor to indole-3-acetic acid,are intermediate oxidation products in NO synthesis.We detected the production of NO from IAOx and other oximes catalyzed by peroxidase(POD)enzyme using both 4-amino-5-methylamino-2′,7′-difluorescein fluorescence and chemiluminescence.Flavins stimulated the reaction,while superoxide dismutase inhibited it.Interestingly,mouse NO synthase can also use IAOx to produce NO at a lower rate than POD.We provided a full mechanism for POD-dependent NO production from IAOx consistent with the experimental data and supported by density functional theory calculations.We showed that the addition of IAOx to extracts from Medicago truncatula increased the in vitro production of NO,while in vivo supplementation of IAOx and other oximes increased the number of lateral roots,as shown for NO donors,and a more than 10-fold increase in IAOx dehydratase expression.Furthermore,we found that in vivo supplementation of IAOx increased NO production in Arabidopsis thaliana wild-type plants,while prx33-34 mutant plants,defective in POD33-34,had reduced production.Our data show that the release of NO by IAOx,as well as its auxinic effect,explain the superroot phenotype.Collectively,our study reveals that plants produce NO utilizing diverse molecules such as oximes,POD,and flavins,which are widely distributed in the plant kingdom,thus introducing a long-awaited oxidative pathway to NO production in plants.This knowledge has essential implications for und

CD74 is a protein whose initial role in antigen presentation was recognized two decades ago. Recent studies have revealed that it has additional functions as a receptor for macrophage migration inhibitory factor and as a receptor for an important human pathogen, Helicobacter pylori (H pylori). The role of CD74 as a receptor is important because after binding of migration inhibitory factor or H pylori, NF-κB and Erk1/2 activation occurs, along with the induction of proinflammatory cytokine secretion. This review provides an up-to-date account of the functions of CD74 and how it might be involved in inflammation and cancer within the gastrointestinal tract.

Acute liver failure secondary to acute antibody mediated rejection after compatible liver transplant: A case report

BACKGROUND The liver has traditionally been regarded as resistant to antibody-mediated rejection(AMR).AMR in liver transplants is a field in its infancy compared to kidney and lung transplants.In our case we present a patient with alpha-1-antitrypsin disease who underwent ABO compatible liver transplant complicated by acute liver failure(ALF)with evidence of antibody mediated rejection on allograft biopsy and elevated serum donor-specific antibodies(DSA).This case highlights the need for further investigations and heightened awareness for timely diagnosis.CASE SUMMARY A 56 year-old woman with alpha-1-antitrypsin disease underwent ABO compatible liver transplant from a deceased donor.The recipient MELD at the time of transplant was 28.The flow cytometric crossmatches were noted to be positive for T and B lymphocytes.The patient had an uneventful recovery postoperatively.Starting on postoperative day 5 the patient developed fevers,elevated liver function tests,distributive shock,renal failure,and hepatic encephalopathy.She went into ALF with evidence of antibody mediated rejection with portal inflammation,bile duct injury,endothelitis,and extensive centrizonal necrosis,and C4d staining on allograft biopsy and elevated DSA.Despite various interventions including plasmapheresis and immunomodulating therapy,she continued to deteriorate.She was relisted and successfully underwent liver retransplantation.CONCLUSION This very rare case highlights AMR as the cause of ALF following liver transplant requiring retransplantation.

Background:Volcanic eruptions have large effects on forest ecosystems and create new substrates,triggering primary succession processes.The Paricutín volcano,born in central-western Mexico,erupted between 1943 and 1952.After the cessation of the eruptive activity,plant colonization began on the lava flows and tephra deposits,including the conifer species that dominate the surrounding mature forests.This study aims to reconstruct the history of the establishment of conifer trees on the substrates created by the Paricutín eruption.Methods:16 sampling plots were established along three transects with northern,southern,and south-western aspects,located every 250 m from the preserved forest to the volcanic cone.Increment cores from 400 conifer trees were extracted and their age was determined by cross-dating annual tree rings.The order of the species colonization and the tree establishment,abundance,and dominance patterns were characterized.Also,the influence of the distance from the mature forests and the inter-annual climatic conditions on the temporal tree establishment pattern was evaluated.Results:Eight pine and one fir species have been established since 1970,only 18 years after the end of the eruptive period.However,tree establishment increased by 12.9%annually after 1995,with the youngest tree in our sample getting established in 2015.We did not find a well-defined temporal and spatial pattern of species arrival,which suggests that colonization occurred randomly,although the four pine species that were established early became the most abundant and dominant.Tree establishment was not influenced by the distance from the mature forest,and wet inter-annual conditions did not enhance pulses of tree recruitment,exhibiting a continuous tree establishment pattern.Conclusions:Conifer species have shown a great capacity for colonizing volcanic substrates created by the Paricutín eruption,which suggests that tropical montane conifers can regenerate rapidly under high-magnitude disturbances.These findings support the use

Globomycin is a cyclic lipodepsipeptide originally isolated from several Streptomyces species which displays strong and selective antibacterial activity against Gram-negative pathogens.Its mode of action is based on the competitive inhibition of the lipoprotein signal peptidase II(LspA),which is absent in eukaryotes and considered an attractive target for the development of new antibiotics.Despite its interesting biological properties,the gene cluster encoding its biosynthesis has not yet been identified.In this study we employed a genome-mining approach in the globomycin-producing Streptomyces sp.CA-278952 to identify a candidate gene cluster responsible for its biosynthesis.A null mutant was constructed using CRISPR base editing where production was abolished,strongly suggesting its involvement in the biosynthesis.The putative gene cluster was then cloned and heterologously expressed in Streptomyces albus j1074 and Streptomyces coelicolor M1146,therefore unambigu-ously linking globomycin and its biosynthetic gene cluster.Our work paves the way for the biosynthesis of new globomycin derivatives with improved pharmacological properties.

Using music to reduce anxiety among older adults in the emergency department:a randomized pilot study

BACKGROUND： An emergency department （ED） visit may be distressing and anxiety-provoking for older adults （age 〉 65 years）. No studies have specifically evaluated the effect of music listening on anxiety in older adults in the ED. OBjECTIVE： The objective of this pilot study was to evaluate the effect of music listening on anxiety levels in older ED patients. DESIGN, SETTING, PARTICIPANTS AND INTERVENTIONS： This was a randomized pilot study in the geriatric ED of an urban academic tertiary medical center. This was a sample of English-speaking adults （age 〉 65 years） who were not deaf （n = 35）. Subjects consented to participate and were randomized to receive up to 60 min of music listening with routine care, while the control group received routine care with no music. Subjects in the music treatment group received headphones and an electronic tablet with pre-downloaded music, and were allowed to choose from 5 selections. MAIN OUTCOME MEASURES： The primary outcome was change in anxiety levels, measured by the state- trait anxiety inventory （STAI）, at enrollment and 1 h later. RESULTS： A total of 35 participants were enrolled： 74% were female, 40% were white, and 40% were black; of these, 32 subjects completed the study protocol. When comparing control （n = 18） against intervention subjects （n = 17）, there were no significant differences in enrollment STAI scores （43.00 ± 15.00 vs. 40.30± 12.80, P = 0.57）. STAI scores 1 hour after enrollment （after the music intervention） were significantly reduced in the intervention subjects compared to the control subjects （with reduction of 10.00± 12.29 vs. 1.88 ± 7.97, P = 0.03）. CONCLUSION： These pilot results suggest that music listening may be an effective tool for reducing anxiety among older adults in the ED.

Generalized Kelvin-Voigt and Maxwell models using Prony series are some of the most well-known models to characterize the behavior of polymers.The simulation software for viscoelastic materials generally implement only some material models.Therefore,for the practice of the engineer,it is very useful to have formulas that establish the equivalence between different models.Although the existence of these relationships is a well-established fact,moving from one model to another involves a relatively long process.This article presents a development of the relationships between generalized Kelvin-Voigt and Maxwell models using the aforementioned series and their respective relaxation and creep coefficients for one and two summations.The relationship between the singular points(maximums,minimums and inflexion points)is also included.

Natural sediment at a dam and its inorganic materials as adsorbents of praseodymium(Pr(Ⅲ))

The sediment deposited behind a dam(DS) and its inorganic materials(IM-DS) were characterized and used for the removal of praseodymium(Pr) from aqueous solutions. Quartz, albite, and kaolinite were found in the sediment by X-ray diffraction. Kinetic data fit well to a pseudo second-order model, the equilibrium time for DS and IM-DS was 2 h and 3 min, respectively, and the adsorption capacity was higher for Pr/DS(qe= 4.91 mg/g) than for Pr/IM-DS(qe= 3.01 mg/g) in these conditions. Pr/DS and Pr/IM-DS data isotherms were in agreement with the Langmuir-Freundlich model and the maximum adsorption capacities were 4.95 and 4.01 mg/g, respectively. Both processes are exothermic(enthalpy change, ΔH0=-22.8 and-21.8 k j/mol for DS and IM-DS, respectively), the increase of the randomness is small, and the feasibility and spontaneity are low. Praseodymium was desorbed by ethylenediaminetetraacetic acid disodium(EDTA-Na2), sodium chloride, nitric, citric, and humic acids solutions, and one hundred percent of praseodymium was desorbed by the EDTA-Na2solution. The Pr2(EDTA)3(praseodymium ethylenediaminetetraacetate) complex was completely desorbed from both materials(DS and IM-DS). Distribution coefficients(KD) depend upon praseodymium concentration,temperature, p H, and the kind of adsorbent. Chemical reactions of praseodymium with kaolinite and iron minerals could explain the adsorption.

A test method for evaluating the thermal environment of underground coal mine refuge alternatives

Since 2009,the Mine Safety and Health Administration(MSHA)has required mines to install refuge alternatives(RAs)in underground coal mines.One of the biggest concerns with occupied RAs is the possible severity of the resulting thermal environment.In 30 CFR 7.504,the maximum allowable apparent temperature(AT)for an occupied RA is specified as 35℃(95°F).Manufacturers must conduct heat/humidity tests to demonstrate that their RAs meet the 35℃(95°F)AT limit.For these tests,heat input devices are used to input the metabolic heat of actual miners.A wide variety of test methods,sensors,and heat input devices could be used when conducting such tests.Since 2012,the National Institute for Occupational Safety and Health(NIOSH)has conducted over thirty 96-hour heat/humidity tests on four different RAs.This paper discusses the test equipment and procedures used during these investigations.This information is useful for RA manufacturers conducting RA heat/humidity tests,for other researchers investigating RA heat/humidity buildup,and for those who need to assess the thermal environment of any confined space where people may be trapped or are seeking refuge.
