A functional tacrolimus-releasing nerve wrap for enhancing nerve regeneration following surgical nerve repair

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

Physical scientists research biomedicine: a call for caution

It is noticeable that nowadays there are more and more scientists with physical science or engineering background working on or started to work on biomedical projects, but rarely vice versa. This reflects the multi-discipline nature of some projects, however, also at least partially can be explained that biomedical research is seen as trendy, fundable, and likely to get more citations and with results more publishable in high impact journals （1,2）. It is also apparent with some publications and lectures that not all these physical scientists/engineers are well-prepared to work on projects which do not align with their own expertise. Some studies are against very basic principles of pharmacology or biology, while others try to solve medical problems which do not exist clinically. I reviewed a number of submissions including a few from prestigious American universities to Nature journals which made no sense with the study design. Much of manpower and financial resource are being wasteful spent. There are also examples of clinical trials which were ill-planned right from the start and in the end only causing much inconvenience to patients to say at the least. In addition, ample examples exist where experienced physical scientists worked with inexperienced medical scientists and resulted in avoidable failures.

The structure of poly-o-phenylenediamine in the redox process has been studied using in-situ resonance Raman spectroscopy, in-situ UV-Vis subtractive reflectance spectroscopy and cyclic voltammetry. The results show that the structure of PoPD is dominated by a ladder polymer with phenazine rings, and at least three structures of PoPD are formed in the redox process. The three kinds of PoPD have different resonance enhancement effect, and the electronic structure of PoPD molecule is influenced obviously by protonation. The results also show that the most stable state of PoPD is its semi-oxidized state.

Mechanical and hydraulic properties of carbonate rock:The critical role of porosity

Laser and Fluorescence Properties of Organic Dye DCM Doped Sol-Gel-Derived Solid-State Materials

Two kinds of new Ormosil xerogels derived from GPTMS (3-glycidyl propyl trimethoxisilane) and TEOS (tetraethoxysilane) or GPTMS and Ti(OBu)4 (tetrabutoxytitanate) were synthesized viathe sol-gel method as dye laser host. Their optical properties, microstructure characteristics and sudece laser damage thresholds have been reported. The effects of dye matrix combination on the fluorescence emission, slope efficiency, photostability and tunable bandwidth were *** oscillation was easily achieved near 620 nm with a slope efficiency of 29% from DCM dopedSiO2-Ormosil pumped by a frequency-doubled radiation from a Q-switched Nd:YAG laser. Modeling the high-energy emission from the gamma-ray binary 1FGL j1018.6–5856

1FGL j1018.6–5856 is a high mass gamma-ray binary containing a compact object orbiting around a massive star with a period of 16.544 *** the compact object is a pulsar,non-thermal emissions are likely produced by electrons accelerated at the termination shock,and may also originate from the magnetosphere and the un-shocked wind of the *** this paper,we investigate the non-thermal emissions from the wind and the shock with different viewing geometries and study the multi-wavelength emissions from 1FGL j1018.6–*** present the analysis results of the Fermi/LAT using nearly 10 years of *** phase-resolved spectra indicate that the Ge V emissions comprise a rather steady component that does not vary with orbital motion and a modulated component that shows flux maximum around inferior *** ke V/Te V light curves of 1FGL j1018.6–5856 also exhibit a sharp peak around inferior conjunction,which are attributed to the boosted emission from the shock,while the broad sinusoidal modulations could be originating from the deflected shock tail at a larger *** modulations of Ge V flux are likely caused by the boosted synchrotron emission from the shock and the IC emission from the unshocked pulsar wind,while the steady component comes from the outer gap of the pulsar ***,we discuss the similarities and differences of 1FGL j1018.6–5856 with other binaries,like LS 5039.
