Microglial glutaminase 1 mediates chronic restraint stress-induced depression-like behaviors and synaptic damages

Dear Editor,Major depressive disorder(MDD)is one of the most common psychiatric illnesses that significantly increase the risk of suicide.1 Stress-triggered dysfunctions of microglia have been identified as a commonly occurred pathological feature of MDD.1–3 Microglial dysfunction contributes to the pathogenesis of MDD via immunoresponses/neuroinflammation-mediated neural damage and pathological synapse loss-mediated neural circuit disruption.1 Although the involvement of microglia in MDD has been widely investigated,the molecular mechanisms underlying microglial dysfunction remain largely ***,we identified glutaminase 1(Gls1)as one key protein that participates in microglial dysfunction.4–6 Gls1 catalyzes the hydrolysis of glutamine to produce glutamate in the brain.4 Besides its well-known role in excitatory neurotoxicity,we found Gls1 up-regulation in microglia in animal models of Alzheimer’s disease and ischemic stroke.4,7 Gls1 activates microglia to overproduce cytokines and release inflammatory extracellular vesicles,therefore leading to neuroinflammation in animal models of Alzheimer’s disease and ischemic stroke.4–6 More importantly,Gls1 has been found to be up-regulated in the brains of MDD patients,and microglial Gls1 deficiency mitigated LPS-induced depression-like behaviors.8 However,LPS exposure is not an appropriate model to mimic MDD phenotypes,leaving the involvement of Gls1 in MDD an undetermined question.

该研究以脂多糖(LPS)+三磷酸腺苷(ATP)刺激的j774A.1巨噬细胞建立细胞焦亡体外模型,探讨大黄游离蒽醌(free total rhubarb anthraquinones,FTRAs)对细胞焦亡的干预机制。体外培养j774A.1巨噬细胞,实验分组为空白对照组、不同质量浓度脂多糖(LPS,0.25、0.5、1μg·mL^(-1))+ATP(1.25、2.5、5 mmol·L^(-1))组,通过CCK-8法检测细胞活力、碘化丙锭(PI)凋亡细胞染色法、乳酸脱氢酶(LDH)及白细胞介素(IL)-18和肿瘤坏死因子(TNF)-α释放,建立巨噬细胞焦亡体外模型。之后将j774A.1巨噬细胞随机分为6组:空白对照组,LPS+ATP组,FTRAs高剂量单独作用组,FTRAs低、中、高剂量预保护组。检测上述细胞焦亡的表型特征和关键指标作为评判FTRAs对LPS+ATP诱导的细胞焦亡的影响依据。Western blot法和RT-PCR法检测半胱氨酸蛋白酶(caspase)-1/11细胞焦亡通路相关蛋白和mRNA表达水平,阐明其抗焦亡效应的分子机制。结果显示巨噬细胞焦亡体外模型以0.50μg·mL^(-1)LPS+5.00 mmol·L^(-1)ATP的刺激条件效果最佳。FTRAs预保护细胞24 h,能够提高焦亡条件下细胞的活力、减轻细胞的受损程度、降低PI染色阳性率并减少LDH、IL-18和TNF-α的释放。FTRAs能够明显抑制GSDMD蛋白的活化,并且显著下调焦亡通路特征分子TLR4、NLRP3、cleaved-caspase-1、cleaved-caspase-11的蛋白表达,但是对ASC蛋白无明显影响。FTRAs还能够明显抑制caspase-1、caspase-11和GSDMD的mRNA表达。这些研究结果表明,FTRAs对LPS+ATP诱导的细胞焦亡模型具有抑制作用,且FTRAs通过调控经典和非经典细胞焦亡信号通路,减少炎性炎症因子的产生,发挥抗焦亡效应。

A recent study by Adams and colleagues published in Nature unveiled more than one hundred genes that are involved in micronucleus formation,1 which is a biomarker for genomic instability and associated with aging,cancer,and other disorders.2 As an outstanding achievement,the authors identified DSCC1(DNA Replication And Sister Chromatid Cohesion 1)as a critical gene protecting against early developmental defects,genomic instability,reduced proliferative capacity,cohesinopathy-related phenotypes,and tumor development in vivo.

Enhancing Ulcerative Colitis Diagnosis:A Multi-Level Classification Approach with Deep Learning

The evaluation of disease severity through endoscopy is pivotal in managing patients with ulcerative colitis,a condition with significant clinical ***,endoscopic assessment is susceptible to inherent variations,both within and between observers,compromising the reliability of individual *** study addresses this challenge by harnessing deep learning to develop a robust model capable of discerning discrete levels of endoscopic disease *** initiate this endeavor,a multi-faceted approach is embarked *** dataset is meticulously preprocessed,enhancing the quality and discriminative features of the images through contrast limited adaptive histogram equalization(CLAHE).A diverse array of data augmentation techniques,encompassing various geometric transformations,is leveraged to fortify the dataset’s diversity and facilitate effective feature extraction.A fundamental aspect of the approach involves the strategic incorporation of transfer learning principles,harnessing a modified ResNet-50 *** augmentation,informed by domain expertise,contributed significantly to enhancing the model’s classification *** outcome of this research endeavor yielded a highly promising model,demonstrating an accuracy rate of 86.85%,coupled with a recall rate of 82.11%and a precision rate of 89.23%.

Intelligent Machine Learning Based Brain Tumor Segmentation through Multi-Layer Hybrid U-Net with CNN Feature Integration

Brain tumors are a pressing public health concern, characterized by their high mortality and morbidity ***, the manual segmentation of brain tumors remains a laborious and error-prone task, necessitatingthe development of more precise and efficient methodologies. To address this formidable challenge, we proposean advanced approach for segmenting brain tumorMagnetic Resonance Imaging (MRI) images that harnesses theformidable capabilities of deep learning and convolutional neural networks (CNNs). While CNN-based methodshave displayed promise in the realm of brain tumor segmentation, the intricate nature of these tumors, markedby irregular shapes, varying sizes, uneven distribution, and limited available data, poses substantial obstacles toachieving accurate semantic segmentation. In our study, we introduce a pioneering Hybrid U-Net framework thatseamlessly integrates the U-Net and CNN architectures to surmount these challenges. Our proposed approachencompasses preprocessing steps that enhance image visualization, a customized layered U-Net model tailoredfor precise segmentation, and the inclusion of dropout layers to mitigate overfitting during the training ***, we leverage the CNN mechanism to exploit contextual information within brain tumorMRI images,resulting in a substantial enhancement in segmentation *** experimental results attest to the exceptionalperformance of our framework, with accuracy rates surpassing 97% across diverse datasets, showcasing therobustness and effectiveness of our approach. Furthermore, we conduct a comprehensive assessment of ourmethod’s capabilities by evaluating various performance measures, including the sensitivity, jaccard-index, andspecificity. Our proposed model achieved 99% accuracy. The implications of our findings are profound.
