Dissipation of S-metolachlor and butachlor in agricultural soils and responses of bacterial communities:Insights from compound-specific isotope and biomolecular analyses

The soil dissipation of the widely used herbicides S-metolachlor(SM)and butachlor(BUT)was evaluated in laboratory microcosms at two environmentally relevant doses(15 and 150μg/g)and for two agricultural soils(crop and paddy).Over 80%of SM and BUT were dissipated within 60 and 30 days,respectively,except in experiments with crop soil at 150μg/*** on compound-specific isotope analysis(CSIA)and observed dissipation,biodegradation was the main process responsible for the observed decrease of SM and BUT in the paddy *** SM,biodegradation dominated over other dissipation processes,with changes of carbon isotope ratios(Δδ13C)of up to 6.5‰after 60 days,and concomitant production of ethane sulfonic acid(ESA)and oxanilic acid(OXA)transformation *** crop soil experiments,biodegradation of SM occurred to a lesser extent than in paddy soil,and sorption was the main driver of apparent BUT *** of the 16 S rRNA gene showed that soil type and duration of herbicide exposure were the main determinants of bacterial community *** contrast,herbicide identity and spiking dose had no significant *** paddy soil experiments,a high(4:1,V/V)ESA to OXA ratio for SM was observed,and phylotypes assigned to anaerobic Clostridiales and sulfur reducers such as Desulfuromonadales and Syntrophobacterales were dominant for both *** soil microcosms,in contrast,were associated with a reverse,low(1:3,V/V)ratio of ESA to OXA for SM,and Alphaproteobacteria,Actinobacteria,and Bacillales dominated regardless of the *** results emphasize the variability in the extent and modes of SM and BUT dissipation in agricultural soils,and in associated changes in bacterial communities.

5-Fluorouracil(5-FU)is an anticancer drug extensively used for different *** metabolic activation leads to several nucleoside and nucleotide metabolites essential to exert its cytotoxic activity on multiple cellular targets such as enzymes,DNA and *** this paper,we describe the development of a method based on liquid chromatography coupled with high resolution mass spectrometry suitable for the simultaneous determination of the ten anabolic metabolites(nucleoside,nucleotide and sugar nucleotide)of *** chromatographic separationwas optimized on a porous graphitic carbon column allowing the analysis of the metabolites of 5-FU as well as endogenous *** detection was performed on an Orbitrap■tandem mass *** of the method was verified in intracellular content and in RNA *** limit of detection was equal to 12 pg injected on column for nucleoside metabolites of 5-FU and 150 pg injected on column for mono-and tri-phosphate nucleotide *** effect was evaluated in cellular contents,DNA and RNA extracts for nucleoside and nucleotides *** method was successfully applied to i)measure the proportion of each anabolic metabolite of 5-FU in cellular contents,ii)follow the consequence of inhibition of enzymes on the endogenous nucleotide pools,iii)study the incorporation of metabolites of 5-FU into RNA and DNA,and iv)to determine the incorporation rate of 5-FUrd into 18 S and 28 S sub-units of rRNA.

采用异佛尔酮二异氰酸酯、聚丙二醇经1,4-丁二醇扩链的聚氨酯(PU),再经过甲醇或甲基丙烯酸羟乙酯封端后溶解在丙烯酸酯单体中,超声乳化后进行自由基细乳液聚合。NCO和OCH3为端基的PU构成的复合乳液GPC谱图都呈现两个峰,DSC和FT-IR分析发现,复合后增加了PU软段和硬段相分离的程度。甲基丙烯酸封端的复合乳液生成交联型聚合物,PU的硬段和硬段间的氢键也受到破坏。复合聚合物膜表面元素含量和本体接近,表明复合细乳液粒子未形成核-壳型结构。

The reduced diameter of skeletal myofibres is a hallmark of several congenital myopathies,yet the underlying cellular and molecular mechanisms remain *** this study,we investigate the role of HACD1/PTPLA,which is involved in the elongation of the very long chain fatty acids,in muscle fibre *** humans and dogs,HACD1 deficiency leads to a congenital myopathy with fibre size disproportion associated with a generalized *** of HACD1-deficient Labradors,Hacd1-knockout mice,and Hacd1-deficient myoblasts,we provide evidence that HACD1 promotes myoblast fusion during muscle development and *** further demonstrate that in normal differentiating myoblasts,expression of the catalytically active HACD1 isoform,which is encoded by a muscle-enriched splice variant,yields decreased lysophosphatidylcholine content,a potent inhibitor of myoblast fusion,and increased concentrations of≥C18 and monounsaturated fatty acids of *** lipid modifications correlate with a reduction in plasma membrane *** conclusion,we propose that fusion impairment constitutes a novel,non-exclusive pathological mechanism operating in congenital myopathies and reveal that HACD1 is a key regulator of a lipid-dependent muscle fibre growth mechanism.

The 800-m(m)run is part of Physical Education classes in Cameroon,after which arrhythmias may occur during ***,this study aimed at determining relationship between 800-m run loads on cardiac autonomic recovery among school ***-two male adolescents(aged[17-1]years)performed ***-exercise heart rate variability(HRV)was recorded during 5-min(min)(HRV5-min)and 15-min(HRV15-min)in time:Standard deviation of normal to normal(SDNN);Root mean square of successive differences(RMSSD)and frequency domain(LH:Low frequency,HF:High frequency,TP:Total power).Rating of Perceived Exertion(RPE)and blood lactate concentration(BLa)were measured after *** HRV5-min,RPE was associated with SDNN(r=-0.44,p<0.01)and RMSSD(r=-0.38,p<0.05).BLa was correlated with SDNN(r=-0.38,p<0.05)and RMSSD(r=-0.56,p<0.001)in the time-domain,LF(r=-0.64,p<0.001),HF(r=-0.58,p<0.001)and TP(r=-0.61,p<0.001)in frequency***,RPE was correlated with LF(r=-0.44,p<0.01),TP(r=-0.49,p<0.01)while exercise duration with HF(r=-0.38,p<0.05).In HRV15-min,BLa was correlated with RMSSD(r=-0.53,p<0.001)and SDNN(r=-0.68,p<0.001).RPE was negatively correlated SDNN(r=-0.53,p<0.01)and RMSSD(r=-0.44,p<0.01).BLa was associated with HF(r=-0.55,p<0.001),TP(r=-0.50,p<0.01)and RPE with LF(r=-0.51,p<0.01),HF(r=-0.50,p<0.01),TP(r=-0.49,p<0.01).In addition,exercise duration was negatively linked to HF(r=-0.36,p<0.05).This study outlined that in untrained adolescents an increase of 800-m loads is associated with a slow vagal indexes of HRV during the recovery.
