AIM:To compare gene targeting efficiencies,expression profiles,and Ca2+ handling potentials in two widely used mouse embryonic stem cell ***:The two widely used mouse embryonic stem cell lines,r1 and HM-1,were cultured and maintained on Mitomycin C treated mouse embryonic fibroblast feeder cell layers,following standard culture *** were incubated with primary and secondary antibodies before fluorescence activated cell sorting analysis to compare known pluripotency ***,cells were harvested by trypsinization and transfected with a kinase-inactive murine Tyk2 targeting construct,following the Biorad and Amaxa transfection ***,the cells were cultured and neomycin-resistant cells were picked after 13 d of *** clones were screened twice by polymerase chain reaction(PCr) and finally confirmed by Southern blot analysis before *** gene expression profiles of more than 20 400 probes were also compared and significantly regulated genes were conf irmed by real time PCr *** handling potentials of these cell lines were also compared using various ***:We found signif icant differences in transfection eff iciencies of the two cell lines(91% ± 6.1% vs 75% ± 4.2%,P = 0.01).Differences in the targeting efficiencies were also significant whether the Amaxa or Biorad platforms were used for *** did not observe signif icant differences in the levels of many known pluripotency ***,our genomewide expression analysis using more than 20 400 spotted cDNA arrays identified 55 differentially regulated transcripts(P rious important biological processes,including binding molecular functions(particularly Ca2+ binding roles).Subsequently,we measured Ca2+ signals in these cell lines in response to various calcium agonists,both in high and low Ca2+ solutions,and found signif icant differences(P regulation of Ca2+ homeostasis betwee

Black locust(robinia pseudoacacia L.) was the first North-American tree species imported to Europe at the beginning of the seventeenth century. It is commonly planted worldwide because of its adaptability to environmental stresses, its valuable wood, easy propagation, frequent and abundant seed production, excellent coppicing,high seedling survival, and relatively high wood yield. In Europe, romania and Hungary have the most highly-developed black locust growing techniques and *** a result of increasing interest in black locust in many countries, this review aims to provide a comprehensive overview of state-of-the-art site requirements, propagation, improvement and management(including growth and yield as well as use in energy plantations).

Diarrhea after bariatric procedures:Diagnosis and therapy

Diarrhea after bariatric procedures, mainly those with malabsorptive elements including roux-Y Gastric Bypass and Biliopancreatic Diversion, is common and an essential determinant of quality of life and microand macronutrient deficiencies. Bariatric surgery is the only sustainably successful method to address morbid obesity and its comorbidities, particularly gaining more and more importance in the specific treatment of diabetic patients. Approximately half a million procedures are annually performed around the world, with numbers expected to rise drastically in the near future. A multitude of factors exert their influence on bowel habits; preoperative comorbidities and procedure-related aspects are intertwined with postoperative nutritional habits. Diagnosis may be challenging owing to the characteristics of postbariatric surgery anatomy with hindered accessibility of excluded segments of the small bowel and restriction at the gastric level. Conventional testing measures, if available, generally yield low accuracy and are usually not validated in this specific population. Limited trials of empiric treatment are a practical alternative and oftentimes an indispensable part of the diagnostic process. This review provides an overview of causes for chronic post-bariatric surgery diarrhea and details the particularities of its diagnosis and treatment in this specific patient population. Topics of current interest such as the impact of gut microbiota and the influence of bile acids on morbid obesity and especially their role in diarrhea are highlighted in order to provide a better understanding of the specific problems and chances of future treatment in post-bariatric surgery patients.

Successful elimination of extended-spectrum beta-lactamase (ESBL)-producing nosocomial bacteria at a neonatal intensive care unit

Background:Extended-spectrum beta-lactamase (ESBL)-producing Gram-negative bacteria are highly dangerous to *** our Neonatal Intensive Care Unit (NICU),the presence of these bacteria became so threatening in 2011 that immediate intervention was ***:This study was conducted during a nearly two-year period consisting of three phases:retrospective (9 months),educational (3 months) and prospective (9 months).Based on retrospective data analysis,a complex management plan was devised involving the introduction of the INSUrE protocol,changes to the antibiotic regimen,microbiological screening at short intervals,progressive feeding,a safer bathing protocol,staff hand hygiene training and continuous monitoring of the number of newly infected and newly colonized *** these intervals,a total of 355 patients were ***:Both ESBL-producing Enterobacter cloaceae and Klebsiella pneumoniae were found (in both patients and environmental samples).In the prospective period a significant reduction could be seen in the average number of both colonized (26/167 patients;P=0.029) and infected (3/167 patients;P=0.033) patients compared to data from the retrospective period regarding colonized (72/188 patients) and infected (9/188 patients) *** was a decrease in the average number of patientdays (from 343.72 to 292.44 days per months),though this difference is not significant (P=0.058).During the prospective period,indirect hand hygiene compliance showed a significant increase (from the previous 26.02 to 33.6 hand hygiene procedures per patient per hospital day,Pre rolled back successfully in a multi-step effort that required an interdisciplinary approach.

Antimicrobial properties of sea anemone Anthopleura nigrescens from Pacific coast of Costa rica

Objective: To evaluate the antimicrobial and antifungal activities of the aqueous and partitioned extract of sea anemone Anthopleura nigrescens(A. nigrescens).Methods: The sea anemone A. nigrescens was collected, minced, homogenized,lyophilized and then further partitioned with diethyl ether, acetone, ethanol and *** fractions were evaluated for antimicrobial activity against bacterial and fungal ***: Acetone extract was found to produce a pronounced inhibition of 7.0 mm against Proteus vulgaris and diethyl ether extract inhibited Pseudomonas aeruginosa with an inhibition zone of 6.5 mm. In antifungal activity, ethanol extract showed good activity against Botrytis cinerea, Trichoderma harzianum and rhizopus oryzae compared with other strains. Acetone and ethanol extract of A. nigrescens showed activity against all of pathogens tested. Slight activity was observed in the water extract with inhibition zone of 1.5 ***: The present study revealed that sea anemone A. nigrescens may also contain some biologically active agents which have potential activity against pathogenic microorganisms.

自主制造系统的研究既受益于信息物理融合系统这一新技术的推动，也响应工业实际需求的号召。目前，先进的传感器技术、数字孪生、人工智能和新型通信技术可以支持半结构化工业环境中的自主操作，可实现对生产过程的实时监控、情况识别和预测、自动和自适应（重新）规划、团队合作，以及通过学习提高性能。本文总结了实现自主工业机器人技术的主要需求，提出了实现此类系统的通用工作流程，并介绍了HUN-rEN SZTAKI最近在装配、焊接、打磨、拾取和放置以及机械加工等多个应用领域的广泛实践。这些解决方案的共同点是以通用的数字孪生概念为核心。最后，本文提出实现工业自主机器人解决方案的一般建议，并讨论了未来要研究的一些开放性问题。

Simultaneous Growth of <i>Chaetoceros muelleri</i>and Bacteria in Batch Cultures

The relationship between bacterial load and the microalga Chaetoceros muelleri was analyzed in a scale-up experiment. The microalga was grown during five days in a 0.4-L Erlenmeyer flask, 2-L Fernbach flask, 18-L Carboy and 400-L column, during which the cell density of C. muelleri, the abundance of heterotrophic bacteria, Vibrio spp., and total bacteria were determined. The highest specific growth rates (μ) of C. muelleri occurred during the first day of culture (0.88 to 2.29 d-1). Highest cell density was recorded on the fifth day at the 2-L (7.62 × 106 cells·mL-1) and 18-L (6.32 × 106 cells·mL-1), coinciding with the maximum counts of heterotrophic bacteria (16.55 × 105 and >30 × 105 CFU·mL-1, respectively). There was a high correlation (0.80, 0.75, 0.85;p?5 cells·mL-1) during the five days occurred at 18-L Carboy. The concentration of total bacteria at all levels was always higher than that of heterotrophic bacteria. The average ratio of heterotrophic to total bacteria was higher in the 2-L (0.0108) and 18-L (0.0172) cultures. The high biomass of C. muelleri and the presence of Vibrio spp. at the 18-L and 400-L levels indicate that it is necessary to establish programs to prevent diseases and economic losses caused by pathogenic bacteria in penaeid shrimp farming.

Nutritional Status,Sarcopenia and the Importance of Prehabilitation of Gastrointestinal Tumor Patients:a Surgical Point of View
