Preclinical evaluation of azathioprine plus buthionine sulfoximine in the treatment of human hepatocarcinoma and colon carcinoma

AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in ***: different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, RKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tumor growth in vivo using athymic mice. We measured safety by serological markers such as aminotransferases and creatine ***: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-mercaptopurine, 6-methylmercaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of membrane potential), and proteolysis activation leading to loss of membrane integrity, release of lactate dehydrogenase and dnA degradation. Some of this biochemical and cellular changes could be reversed by n-acetylcysteine (a GSH replenisher). In vivo studies showed that HepG2 tumor growth was inhibited when AZA was combined with ***: Our studies suggest that a combination of AZA plus BSO could be useful for localizedtreatment of hepatocellular carcinoma as in the currently used transarterial chemoembolization method.

The Hunga Tonga Hunga Ha’apai submarine volcano has experienced repeated eruptions in the latest *** recent one,in January 2022,released an enormous amount of energy inducing global perturbations,as tsunamis and atmospheric *** structure of the volcano is poorly understood,especially its internal ***-seated magmatic connections are difficult to define or *** use a high-resolution gravity data set obtained via satellite to calculate the Bouguer anomaly over its structure,to perform a preliminary exploration of its *** 3d gravity inversions,we find a complex plumbing system with various exhaust trajectories and multiple surface pockets of low-density material within the volcanic edifice;some appear to be associated with ring *** is in line with the report of the 2009 eruption,described as beginning from multiple *** found no signs of a magma chamber within 6 km depth,although several volcanic conduits are identified from such depth to the *** variations occur within a plumbing conduit or may vary from one conduit to another in the same *** models yield quantitative estimates for areas of magma-water interaction,constituting a baseline to compare with structural changes to be induced in future eruptions.

The incidence of cardiogenic shock(CS)complicating acute myocardial infarction(AMI-CS)remains between 3%and 10%and in-hospital mortality is hardly less than 30%.[1-3]In addition,the economic cost of caring for these patients is high.[4]Revascularization in the acute phase is the only measure that has demonstrated to modify the prognosis of AMI-CS.

Structure and Function of TPC1 Vacuole SV Channel Gains Shape

Plants and animals in endosomes operate TPCI/SV-type cation channels. All plants harbor at least one TPC1 gene. Although the encoded SV channel was firstly discovered in the plant vacuole membrane two decades ago, its biological function has remained enigmatic. Recently, the structure of a plant TPC1/SV channel protein was determined. Insights into the 3d topology has now guided site-directed mutation ap- proaches, enabling structure-function analyses of TPC1/SV channels to shed new light on earlier findings. Fou2 plants carrying a hyperactive mutant form of TPC1 develop wounding stress phenotypes. Recent studies with fou2 and mutants that lack functional TPC1 have revealed atypical features in local and long-distance stress signaling, providing new access to the previously mysterious biology of this vacuolar cation channel type in planta.

The accurate identification of dinosaur egg accumulations as nests or clutches is crucial for understanding the reproductive behaviour of these extinct ***,existing methods often rely on the presence of complete eggs and embryo remains,and sedimentological criteria that are only applicable to well-structured *** this study,we introduce an innovative approach to characterize egg accumulations in structureless sediments,where traditional nest structures may not be *** methodology employs a unique combination of sedimentological,taphonomic,geochemical,and geophysical proxies for the study of egg *** applied this approach to the egg accumulation from Paimogo(Jurassic,Portugal),traditionally interpreted as a *** findings reveal that the Paimogo egg assemblage is a secondary deposit,resulting from a flooding event in a fluvial plain that dismantled several allosauroid and crocodylomorph *** eggshell vapor conductance results,coupled with sedimentological evidence,suggest that allosauroid dinosaurs buried their eggs in the dry terrain of overbank areas close to a main channel during the breeding season,likely during the dry season to prevent the embryos from *** research underscores the necessity of multidisciplinary approaches in interpreting egg accumulations and offers a novel methodology for studying these accumulations in structureless *** findings provide new insights into the breeding behaviour and nesting preferences of these extinct organisms,contributing to our understanding of dinosaur ecology.

diogenes Laërtius: A Moderate Skeptic in the History of Philosophy (Book IX)

This paper presents the keys and reasons for diogenes Laërtius’ alleged scepticism, based on an analysis of thegeneral design of his work The Lives and Opinions of Eminent Philosophers. I believe that it would be manifestlyerroneous to seek confirmation of this scepticism solely in Book IX without taking into account the overallstructure of the work. A convincing explanation is also provided of one of the most enigmatic and most studiedphrases in this work. What did diogenes mean when he said that Apollonides of nicaea was ὁ παρ’ ἡμῶν (“one ofus”)?

Geophysical Modeling with Satellite Gravity data: Eigen-6C4 vs. GGM Plus

Satellite data sets are an asset in global gravity collections;their characteristics vary in coverage and resolution. new collections appear often, and the user must adapt fast to their characteristics. Their use in geophysical modeling is rapidly increasing;with this in mind we compare two of the most densely populated sets: EIGEn-6C4 and GGMplus. We characterize them in terms of their frequency histograms, Free Air anomalies, power spectrum, and simple Bouguer anomalies. The nature of the digital elevation models used for data reduction is discussed. We conclude that the GGMplus data set offers a better spatial resolution. To evaluate their effect in geophysical modelling, we chose an inland region with a prominent volcanic structure in which we perform 3d inversions of the respective Bouguer anomalies, obtaining density variations that in principle can be associated with the geologic materials and the structure of the volcanic edifice. Model results are analyzed along sections of the inverted data;we conclude that the GGMplus data set offers higher resolution in the cases analyzed.
