Transcript shortening via alternative polyadenylation promotes gene expression during fracture healing

Maturation of the 3′end of almost all eukaryotic messenger RNAs(m RNAs)requires cleavage and *** mammalian m RNAs are polyadenylated at different sites within the last exon,generating alternative polyadenylation(APA)isoforms that have the same coding region but distinct 3′untranslated regions(UTRs).The 3′UTR contains motifs that regulate m RNA metabolism;thus,changing the 3′UTR length via APA can significantly affect gene *** ossification is a central process in bone healing,but the impact of APA on gene expression during this process is ***,we report the widespread occurrence of APA,which impacts multiple pathways that are known to participate in bone ***,the progression of endochondral ossification involves global 3′UTR shortening,which is coupled with an increased abundance of shortened transcripts relative to other transcripts;these results highlight the role of APA in promoting gene expression during endochondral bone *** mechanistic studies of transcripts that undergo APA in the fracture callus revealed an intricate regulatory network in which APA enhances the expression of the collagen,type I,alpha 1(Col1a1)and Col1a2 genes,which encode the 2 subunits of the abundantly expressed protein collagen *** exerts this effect by shortening the 3′UTRs of the Col1a1 and Col1a2 m RNAs,thus removing the binding sites of mi R-29a-3p,which would otherwise strongly promote the degradation of both *** together,our study is the first to characterize the crucial roles of APA in regulating the 3′UTR landscape and modulating gene expression during fracture healing.

Advantages & Disadvantages of Online CAS

Online Computer Algebra Systems (CAS) have become increasingly popular among students and teachers. The reasons are many such as being more flexible, simple to use, accessible from anywhere, etc. However, as with any educational tool, they also have some disadvantages that we should know. The purpose of this study is to analyze advantages and disadvantages of online CASs and propose some techniques to optimize CAS performance in order to reduce weaknesses. The research results reveal that online CAS versions are on the rise but lag in some capabilities in comparison with desktop versions.

High performance corrosion and wear resistant composite titanium nitride layers produced on the AZ91D magnesium alloy by a hybrid method

Composite,diffusive titanium nitride layers formed on a titanium and aluminum sub-layer were produced on the AZ91D magnesium *** layers were obtained using a hybrid method which combined the PVD processes with the final sealing by a hydrothermal *** microstructure,resistance to corrosion,mechanical damage,and frictional wear of the layers were *** properties of the AZ91D alloy covered with these layers were compared with those of the untreated alloy and of some engineering materials such as 316L stainless steel,100Cr6 bearing steel,and the AZ91D alloy subjected to commercial *** has been found that the composite diffusive nitride layer produced on the AZ91D alloy and then sealed by the hydrothermal treatment ensures the corrosion resistance comparable with that of 316L stainless *** layers are characterized by higher electrochemical durability which is due to the surface being overbuilt with the titanium oxides formed,as shown by the XPS examinations,from titanium nitride during the hydrothermal *** composite titanium nitride layers exhibit high resistance to mechanical damage and wear,including frictional wear which is comparable with that of 100Cr6 bearing *** performance properties of the AZ91D magnesium alloy covered with the composite titanium nitride coating are substantially superior to those of the alloy subjected to commercial anodizing which is the dominant technique employed in industrial practice.

Social parasitic Hymenopterans have evolved morphological, chemical, and behavioral adaptations to overcome the sophisticated recognition and defense systems of their social host to invade host nests and exploit their worker force. In bumblebees, so- cial parasitism appeared in at least 3 subgenera independently： in the subgenus Psithyrus consisting entirely of parasitic species, in the subgenus Alpinobombus with Bombus hy- perboreus, and in the subgenus Thoracobombus with B. inexspectatus. Cuckoo bumblebee males utilize species-specific cephalic labial gland secretions for mating purposes that can impact their inquiline strategy. We performed cephalic labial gland secretions in B. hyperboreus, B. inexspectatus and their hosts. Males of both parasitic species exhibited high species specific levels of cephalic gland secretions, including different main com- pounds. Our results showed no chemical mimicry in the cephalic gland secretions between inquilines and their host and we did not identify the repellent compounds already known in other cuckoo bumblebees.

The Crisis, Responsibility and Regulation

Any observer of current economic development in the world and in the Europe should ask about the causes of the current crisis situation. The authors of this article perceive current situation in the European Union and many of its member states associated with long-term developmental tendencies still less respecting the objective differences in various economic situation of the member states. Establishment and further functioning of monetary union in Europe is gradually becoming a burden for many countries, which prevents full utilization of instruments of monetary and fiscal policy by daily, and in many cases qualitative differences in the problems of economies of individual member states. The fulfillment of these tendencies in particular in the Euro Area monetary policy is confronted with the dominant position of member states in fiscal policy and in the mutual differences among member states. As considerations of differences in economic level and in member states fiscal policies, both of them are based on official figures, some of them are part of the state. In addition, EU institutions face to face to crisis they are increasingly resorting to deepening statist centralization and enter more often to the processes which are usually domains of business subjects, at most of nation states. All these trends lead to a deepening asymmetry between the decision-making processes and responsibilities for economic development and they could lead to even larger failure of national economies or at European or global level. Since the accession of the Czech Republic to the European Union in 2004 our department has dealing with issues connecting to the economic management of insurance companies and in relation to the ongoing integration process research interest is extended also to other segments of financial markets.

Obsessive-Compulsive Cognitions, Symptoms and Religiousness in an Iranian Population

Individual differences in obsessive-compulsive (OC) behavior in various cultures appear to be associated with religiosity. The purpose of this study was to evaluate the role of religion in OC symptoms and cognitions in distinctly low and high religious groups from a normal community sample of 119 Iranian Muslims. Specifically, we compared the two groups on OC cognitions and symptoms, and we examined the correlations between the cognitive and symptom measures within each group. There was a trend for the high religious group to produce greater scores than those in the low religious group on the Obsessive Beliefs Questionnaire (OBQ) subscale of threat overestimation and responsibility. Furthermore, participants that were more religious achieved significantly higher scores on the Penn Inventory of Scrupulosity and on its Fear of God subscale. Although a number of significant correlations were observed between OBQ and Padua Inventory total and subscale scores, particularly in the low religious group, there was no conclusive relationship between religiosity and OC behavior and obsessional beliefs. Religion appears to be one more arena where OC symptoms expressed, rather than being a determinant of the disorder.

Mesenchymal stromal cells from human perinatal tissues: From biology to cell therapy

Cell-based regenerative medicine is of growing interest in biomedical research. The role of stem cells in this context is under intense scrutiny and may help to define principles of organ regeneration and develop innovative therapeutics for organ failure. Utilizing stem and progenitor cells for organ replacement has been conducted for many years when performing hematopoietic stem cell transplantation. Since the first successful transplantation of umbilical cord blood to treat hematological malignancies, non-hematopoietic stem and progenitor cell populations have recently been identified within umbilical cord blood and other perinatal and fetal tissues. A cell population entitled mesenchymal stromal cells (MSCs) emerged as one of the most intensely studied as it subsumes a variety of capacities: MSCs can differentiate into various subtypes of the mesodermal lineage, they secrete a large array of trophic factors suitable of recruiting endogenous repair processes and they are *** on perinatal tissues to isolate MSCs, we will discuss some of the challenges associated with these cell types concentrating on concepts of isolation and expansion, the comparison with cells derived from other tissue sources, regarding phenotype and differentiation capacity and finally their therapeutic potential.

Impact of International Financial Reporting Standards （IFRS） Adoption on Key Financial Ratios＂ The Case of the Czech Republic

This contribution analyzes the impact of new International Financial Reporting Standards （IFRS） reporting rules on financial ratios prepared in the Czech companies. Using a sample of 16 Czech firms, we attempt to measure the scope and size of the differences in the selected set of financial ratios as calculated with data reported according to the traditional Czech accounting standards （CAS） and under the IFRS provisions. Our study discovers that there are important differences resulting from the two reporting formats. Our research comes to a conclusion that translation of Czech statements to IFRS may cause changes in the values of financial indicators without relationship to the real change in the firms＇ value, performance, and stability. Even though the findings were not statistically significant, the indicative results of our measurements disclosed an important fact that the transition to IFRS could cause deterioration of key indicators and thereby could impact on the overall rating of companies. One needs to be cautious with generalization due to the small sample size.

A Knowledge Management Framework of the Information Technology-Based Approach for Improving Occupational Safety Training of Young Workers

This paper aims to develop a novel knowledge management framework for improving the system of pro-safety attitudes among young worker population in Poland. This is done through an efficient use of existing sources of explicit and tacit knowledge in the field of vocational occupational safety and health （OSH） training. This project identifies three specific research steps for the development of the knowledge management framework of the information technology-based approach for improving occupational safety training of young workers, including：（1） the development of the conceptual model of the flow of explicit knowledge of OSH; （2） evaluation of the effectiveness of formal and explicit knowledge sources for OSH and training methods using these sources in the context of attitudes towards health and safety at work; and （3） verification of the proposed model of vocational training with the use of combined explicit and hidden knowledge. The proposed framework includes consideration of the effectiveness of the formal and informal sources of safety knowledge. A mapping from the formal sources of explicit knowledge about the occupational safety to expected attitudes （hidden knowledge） towards OSH outcomes, including emotional, cognitive, and behavioral aspects has been used. The above framework should help to improve the system of vocational training for young workers in Poland.

The current bumblebee decline leads to inbreeding in populations that fosters a loss of allelic diversity and diploid male production. As diploid males are viable and their offspring are sterile, bumblebee populations can quickly fall in a vortex of extinction. In this article, we investigate for the first time a potential premating mechanism through a major chemical reproductive trait （male cephalic labial gland secretions） that could prevent monandrous virgin queens from mating with diploid males. We focus our study on the cephalic labial gland secretions of diploid and haploid males ofBombus terrestris （L.）. Contrary to initial expectations, our results do not show any significant differentiation of cephalic labial gland secretions between diploid and haploid specimens. Queens seem therefore to be unable to avoid mating with diploid males based on their compositions of cephalic labial gland secretions. This suggests that the vortex of extinction of diploid males could not be stopped through premating avoidance based on the cephalic labial gland secretions but other mechanisms could avoid mating between diploid males and queens.
