＜正＞Epilepsy is one of the most common neurological disorders and characterized by recurrent seizures,afflicting about 1%of the *** studies show that histaminergic neuron system plays an important role in the pathogenesis of seizure *** brain histamine seems to be involved in mechanisms regulating seizure susceptibility. However,histamine can not penetrate across blood-brain barrier（BBB） and it has been reported to be involved in brain *** presents in muscle and brain of mammalian in high concentrations,camosine serves as a reservoir for histidine,which is a precursor of *** is proposed that there exist camosine-histidine-histamine pathway in the brain and plays certain important role in ***,few reports have demonstrated the significant relations between brain camosine and *** there is only limited information about the effects of camosine on kindled seizures. So we first investigate the role of camosine in amygdaloid kindled seizures and PTZ-induce kindled seizure in rats. Camosine significantly decreased seizure stage,afterdischarge duration and generalized seizure duration,and significantly prolonged generalized seizure latency of amygdaloid kindled seizures,in a dose-dependent,and time-related *** protective effect of camosine was completely antagonized by histamine H1-antagonists pyrilamine and diphenhydramine,but not by histamine H2-antagonist *** caused a significant increase of camosine and histidine levels in the hypothalamus,thalamus,hippocampus,amygdala and cortex,as well as histamine-Fluoromethylhistidine, a selective and?levels in the hippocampus and amygdala,irreversible histidine decarboxylase inhibitor,only partially reversed the inhibition of amygdaloid kindled seizures induced by *** addition,camosine significantly decreased glutamate contents in the *** assay,Hoechst 33342 and propidium iodide staining showed that camosine suppressed excitot

反向遗传系统可以对RNA病毒直接进行遗传操作 ,为RNA病毒的分子生物学研究提供了一种强大的工具。在过去 2 0年 ,特别是自 90年代中期第一例负链RNA病毒感染性克隆构建成功以来 ,动物RNA病毒的分子生物学研究取得了长足的进展 ,这很大程度上归功于各种动物RNA病毒反向遗传系统的建立。这里系统总结了人类及动物非反转录RNA病毒中各类代表性成员在建立反向遗传系统时的方案设计、遇到的困难及研究者如何克服这些困难。分类讨论到的代表性病毒种属有脊髓灰质炎病毒、冠状病毒 (包括SARS病毒 )、黄病毒、野田村病毒、流感病毒、传染性法氏囊病病毒以及呼肠孤病毒等。

Effects of CpG oligodeoxynucleotide on transcription factors GATA-3 and T-bet mRNA expression in asthmatic mice

Aim:To investigate effects ofCpG oligod'eoxynucleotide(CpG ODN)on the mRNAexpression of transcription factors GATA binding protein 3(GATA-3)and T-boxexpressed in T cells(T-bet)in asthmatic ***:An asthmatic mousemodel was established and treated with CpG *** inflammatory cells andeosinophils in bronchoalveolar lavage fluid(BALF)were counted and inflamma-tory cell infiltration in lung tissue was ***-y and interleukin-4concentrations in BALF and splenocyte culture supernatants were detected us-ing an enzyme-linked immunosorbent *** factor GATA-3 and T-bet mRNA expression in splenocytes and lung tissue were detected by reversetranscription-polymerase chain ***:Total inflammatory cells andeosinophils in BALF were reduced in the CpG ODN-treated group compared withthe asthma group,and inflammatory cell infiltration in lung tissue was also signifi-cantly *** ODN treatment increased the interferon-γ concentrationbut decreased the interleukin-4 concentration in both BALF and splenocyte cul-ture ***-3 mRNA expression was reduced in both lung tissueand splenocytes in the CpG ODN-treated group,while the mRNA ratio ofT-bet toGATA-3 in splenocytes was ***:CpG ODN treatment inhibitsairway inflammatory cell infiltration and regulates interferon-y/interleukin-4 syn-thesis in asthmatic mice,possibly through a mechanism of downregulation ofGATA-3 mRNA expression in both lung tissue and splenocytes.

Molecular Characteristics of S1 Gene of infectious Bronchitis Virus Isolated from Chicken Proventriculus

infectious bronchitis virus was isolated from swollen proventriculi of clinically ill chicken. The suspected virus samples (2/97, 3/97, 1/98) were adapted in SPF chicken embryos for virus isolation and identification. All the virus isolates were able to agglutinate chicken erythrocytes after treatment with trypsin, and interfer with the reproduction of Newcastle disease virus in chicken embryos, and have low antigenic relat-edness values with reference positive IBV. The isolates 2/97, 3/97, 1/98 RNAs extracted from the allantoic fluid of inoculated embryonated eggs were converted to cDNA by reverse transcription with 3'-primer of S1 gene of (IBV). Polymerase chain reaction (PCR) was performed with two primers which span the S1 gene. Amplified product of 1. 93 kb was subjected to EcoR I and BamH I digestion and the fragments obtained were the same as expected size. The PCR product was ligated to pBlueScript-SK ( + ) vector, and its nucleotide sequence was determined by the dideoxy-mediated chain termination method. Nucleotide sequence analysis showed 73. 6 - 99. 7% homology between the isolated IBV and the IBV strains in GenBank. The homology of amino acid was 71. 4 - 99.4%.

New Concepts in Tumor Antigens:Their Significance in Future Immunotherapies for Tumors

The identification and molecular characterization of self-antigens expressed by human malignancies that are capable of elicitation of anti-tumor immune responses in patients have been an active field in tumor immunology. More than 2,000 tumor antigens have been identified, and most of these antigens are self-antigens. These significant progresses have led to the renaissance of tumor immunology and studies on anti-tumor immunotherapy. However, despite of the progress in the identification of self-tumor antigens, current antigen-specific immunotherapies for tumors are far less satisfied than expected, which reflects the urgent need to improve our understanding on self-tumor antigens. In order to develop more effective antigen specific anti-tumor immunotherapies and to monitor the responses to these immunotherapies in patients with tumors, many important fundamental questions need to be addressed. We propose for the first time that the studies in addressing the characteristics of self-tumor antigens and autoantigens are grouped as a new subject termed ＂antigenology＂. In this brief review, we would outline the progress in the identification of tumor antigens in solid tumors and hematologic malignancies, and overview the new concepts and principles of antigenology and their significance for future immunotherapies to these malignancies.

Rapid accumulation of bacterial genomic data offered an unprecedented opportunity to understand bacterial biology from a holistic view of *** can thus closely look at the way in which a pathogen is evolved,and these data has been applied to molecular epidemiology and microbial forensics,and screening of novel diagnostic,vaccine and drug *** newly developed high-throughput low-cost sequencing technologies,such as 454,Solexa and SOLiD,will promote the acquisition and application of genomic data in new research areas that we dared not imagine previously,such as the metagenomics of human gastric-intestinal tract,for better and comprehensive understanding of human health and disease.

＜正＞Postsynaptic density（PSD）-95 is originally isolated from glutamatergic synapse where it serves as a physical tether to allow neuronal nitric oxide synthase（nNOS） signaling by N-methyl-D-aspartate receptor（NMDAR） activity. Considering the physiological importance of glutamate receptor and nitric oxide（NO） during development,we examined the spatiotemporal expression of PSD-95 and nNOS in the lumbar spinal cord at a postnatal ***,both gene and protein levels of them gradually increased with age after birth,peaked at the postnatal day 14（P14）,and then decreased to an adult *** addition,the enhanced coimmunoprecipitations between PSD-95 and nNOS were detected in developing spinal ***,PSD-95 staining codistributed with nNOS in NeuN-positive motor neurons and sensory neurons at *** findings indicate that PSD-95 and nNOS might collectively participate in spinal cord development.
