Cytosolic HSP90 Cochaperones HOP and FKBP Interact with Freshly Synthesized Chloroplast Preproteins of Arabidopsis

most chloroplast and mitochondrial proteins are synthesized in the cytosol of the plant cell and have to be imported into the organelles post-translationally. molecular chaperones play an important role in preventing protein aggregation of freshly translated preproteins and assist in maintaining the preproteins in an import competent state. Pre- proteins can associate with HSP70, HSP90, and 14-3-3 proteins in the cytosol. In this study, we analyzed a large set of wheat germ-translated chloroplast preproteins with respect to their chaperone binding. Our results demonstrate that the formation of distinct 14-3-3 or HSP90 containing preprotein complexes is a common feature in post-translational protein transport in addition to preproteins that seem to interact solely with HSP70. We were able to identify a diverse and extensive class of preproteins as HSP90 substrates, thus providing a tool for the investigation of HSP90 client protein association. The analyses of chimeric HSP90 and 14-3-3 binding preproteins with exchanged transit peptides indicate an involvement of both the transit peptide and the mature part of the proteins, in HSP90 binding. We identified two partner components of the HSP90 cycle, which were present in the preprotein containing high-molecular-weight complexes, the HSP70/HSP90 organizing protein HOP, as well as the immunophilin FKBP73. The results establish chloroplast preproteins as a general class of HSP90 client proteins in plants using HOP and FKBP as novel cochaperones.

Dear Editor, Phosphoenolpyruvate carboxylase （PEPC） is the essential enzyme for initial carbon fixation in C4 photosynthesis. The C4-PEPC evolved from a non-photosynthetic C3 ancestor by subtle sequence changes resulting in distinctly different kinetic and regulatory characteristics （Svensson et al., 1997）. malate and aspartate, which are formed from the carboxyla- tion product of PEPC, serve as feedback inhibitors of the PEPC （Wedding et al., 1990）. Higher malate or aspartate concen- trations during C4 photosynthesis require a reduced sensitiv- ity towards the feedback inhibitors of the C4-PEPC （Jacobs et al., 2008）.

Ionic liquids(ILs)have been used effectively in many applications for reducing problems related to friction and *** this work,the potential of ILs as an anti-wear and extreme pressure lubricant additive for high load-carrying gearbox applications such as helicopter transmissions has been *** halide-free ILs:P_(8881)(BuO)_(2)PO_(2)^(-)(1)and P_(8881)(meO)_(2)PO_(2)^(-)(2),which are blended at 5 wt%each into a standard non-additivated FVA2 base oil(BO)are *** solid-liquid interface,friction and load-carrying capacity,and wear(scuffing)behavior are studied on the nano-,lab-,and component-scale,respectively,at a different range of temperature and loading conditions by using the atomic force microscopy(AFm),Schwing-Reib-Verschleiβ(SRV)friction tests,and Brugger tests,as well as forschungsstelle für zahnrader und getriebebau(FZG)back-to-back gear test *** AFm analysis shows nearly no change of adhesion over the full range of studied temperature for the IL blends compared to the ***,IL blends demonstrate a very stable coefficient of friction(COF)of around 0.16,which even decreases with increasing test temperatures ranging from 40 to 120℃.A clear reduction in COF up to 25%is achieved by adding only 5 wt%of the investigated Ils in the BO,and the Brugger tests also show a pronounced enhancement of load-carrying ***,on the component-scale,a significant improvement in gear scuffing performance has been observed for both used IL blends.A detailed characterization of the wear tracks from the SRV friction tests via the transmission electron microscopy(TEm)revealed the formation of a phosphate(P-O)-based amorphous tribo-chemical layer of about 20 nm ***,this work may present an approach for Ils to be used as an additive in conventional lubricants due to their ability to enhance the lubrication properties,making them an alternative lubricant solution for high load-carrying gearbox applications.

Evaluating Emissions of Nitrous Oxide from Cropland Soils Under Different Rotations in mato Grosso, Brazil:A Scenario Simulation Study

Expansion of cropland involves immense land use changes, and the resulting intensified management practices have a strong influence on the functioning of the underlying soil. For instance, increased application of nitrogen(N) fertilizer is known to enhance fluxes of nitrous oxide(N2O) from the soil to the atmosphere. The emission factor(EF) proposed by the Intergovernmental Panel on Climate Change(IPCC) assumes a linear relationship between added N and N2O-N fluxes, but it does not account for environmental factors, such as soil properties or climate. Due to the high spatial and temporal variability of N2O-N fluxes, mechanistic models are preferable in terms of extrapolation to larger scales. In this study, we evaluated simulated N2O-N fluxes from soils under agricultural use in the Brazilian state, mato Grosso, using the CANDY(Carbon and Nitrogen Dynamics) model. A control tool was developed in order to enable the simulation of 1 650 scenarios covering different sites(soil + climate) and management regimes(crop rotation +amount of applied fertilizer + sowing and harvesting dates). Results suggested that the sites had a very strong influence on calculated emissions, which is not accounted for by static EF. Furthermore, most fertilizer-induced N2O-N fluxes derived from the scenario simulations were best described by a non-linear function. For sounder budgeting on the federal and national scale, there is still a strong need for long-term observations of continuous crop rotations and spatial distribution of soil types and their specific characteristics. The presented results provide a valuable starting point for developing further scenario simulations and adapting experimental campaigns for N2O emission study.

Cortisol levels in skimmed milk during the first 22 weeks of lactation and response to short-term metabolic stress and lameness in dairy cows

Background： Cortisol is secreted into blood in reaction to acute stress, but also in phases of diminished feed intake and changed animal behavior. As cows do not always show clear signs of discomfort, reliable diagnostic markers could be used to provide information regarding individual cows＇ distress. The objective of this study was to establish an ether free immunoassay for the detection of cortisol and to determine values during the first 22 weeks of lactation. Furthermore, the response in milk cortisol levels was assessed during times of metabolic stress and pain associated symptoms of lameness. methods： milk yield and composition, blood serum glucose, NEFA and BHBA as well as milk cortisol were determined in 24 multiparous Holstein-Friesian cows over the course of the first 22 weeks of lactation. Animals were further checked for signs of clinical diseases on a daily basis. Two feed restrictions over three days （FR; 70 % of precious ad libitum intake） were performed during the 4th wk and the 21st wk, respectively. An ELISA for cortisol measurement in easily accessible bovine skimmed milk was established and applied. Results： On the last day of FR in early lactation, a reduction in milk yield and changes in serum metabolites compared to respective previous values were detected. The FR in mid-lactation resulted in no changes in milk production and serum metabolites. milk cortisol was highest during first wk of lactation and remained on comparable levels thereafter. milk yield and composition were not influenced by FR. Lameness resulted in enhanced milk cortisol levels. Conclusion： milk cortisol could be used as an indicator of painful symptoms such as lameness. Higher values of milk cortisol levels during first wk of lactation should be taken into account for interpretation.
