Structure Elucidation of a New Toxin from the Mushroom Cortinarius rubellus Using Gas Chromatography-Mass Spectrometry (GC-MS)

Cortinarius orellanus (Fries) and C. rubellus (Cooke),which were formerly also known as C. speciosissimus, are poisonous mushrooms containing the toxin orellanine and several degradation products of orellanine,includingorelline and orellinine. Mass intoxication by poisonous mushrooms was observed in Poland in 1952-1957 [1]. In 1957, the cause of these outbreaks was described by Grzymala as poisoning by a member of the Cortinarius family. The toxin orellanine was first isolated from C. orellanusby Grzymala in 1962;the chemical structure of orellanine was later determined to be 3,3',4,4'-tetrahydroxy-2,2'-bipyridine-N,N'-dioxide. Poisoning with C. orellanus and C. rubellus has a very specific character. The first symptoms of intoxication usually do not appear until 2-3 days after ingestion, but in some cases intoxication appears after three weeks. The target organ for the toxin is the kidney. Histologically, it is easy to record the specific damage. The presence of degradation products of orellanine in kidney can be confirmed chromatographically, suggesting that the cause of poisoning is orellanine. However, the presence of orellanine in the blood of intoxicated persons has not been directly detected. A specific model was developed by Brondz et al. for the detection of orellanine, orelline, and orellininein animal stomach fluids [2-4]. The hypothesis that the fungal toxin orellanine as a diglucoside can be transported from the digestive system by the blood to the kidney could not be supported. The toxin orellanine as a diglucoside is very unstable in an aqueous acidic environment.[i1]?However, in the present study, it was possible to record an additional substance in animal stomach fluids using GC-MSafter ingestion ofC. rubellus. This substance, which has been namedrubelline, is part of a toxic mixture inC. orellanusandC. rubellusand is closely related to orellanine. Analytic Methods for Quality Control of Scientific Publications Part VI: Presentation in Research Gate, Journal Indexing, and Recognition

In the world of science, recognition of scientific performance is strongly correlated with publication visibility and interest generated among other researchers, which is evident by downloads and citations. A published paper’s number of downloads and citations are the best indices of its importance and are useful measures of the researchers’ performance. However, the published paper should be valuated and indexed independently, and the prestige of the journal in which it is published should not influence the value of the paper itself. By participating in and presenting at congresses and international meetings, scientists strongly increase the visibility of their results and recognition of their research;this also promotes their publications. Status in Research Gate (RG), the so-called RG Score, the Percentile, and the h-index give researchers feedback about their performance, or their place and prestige within the scientific community. RG has become an excellent tool for disseminating scientific results and connecting researchers worldwide. RG also allows researchers to present achievements other than publications (e.g., membership in recognized associations such as the American Chemist Society, a biography in Marquis Who’s Who in the World, awards received, and/or ongoing projects). This paper discusses questions regarding how the RG Score, Percentile, and h-index are calculated, whether these methods are correct, and alternative criteria. RG also lists papers with falsified results and the journals that publish them. Thus, it may be appropriate to reduce the indices for such journals, authors, and the institutions with which these authors are affiliated.

Influence of Modifiers, Extractants, and Trappers on Lipid Composition with Liquids in Standard State Extraction, Supercritical Fluid Extraction and Trapping by Supercritical Fluid Extraction, Part II

Modifiers have a broad array of influences on extraction with liquids in standard state, supercritical fluid extraction (SFE), trapping by SFE and supercritical fluid chromatography (SFC). They can significantly change the qualitative and quantitative results. Quantitative and qualitative results can be influenced by different extractants and modifiers in different ways as it was shown by Brondz et al. at 2007 in “The real nature of the indole alkaloids in Cortinarius infractus: Evaluation of artifact formation through solvent extraction method development”, J. Chromatography A, 1148, 1-7. The choice of correct extractant, modifier, and trapper to the bulk mobile phase for supercritical fluids (SFs) or for liquids in subcritical or in the liquids in standard state is a challenge in any extraction procedure. This is the second paper in a sequence that describes the influence of extractants and modifiers on the performance of SFs and results of extraction with liquids in standard state and SFE. Here, attention is given to possible mistakes in qualitative and quantitative results by poor understanding of the influence of extractants, modifiers, and trappers on extraction and trapping process by a careless choice of extractant, modifier, and trapper for extraction with liquids in standard state and SFE. The SF chosen for discussion in the paper is CO2. However, similar effects can be observed with use of other SFs and fluids in subcritical and standard states. In this paper, the discussion of lipids, fatty and carboxylic acids have been chosen as target analytes for extraction, trapping and analysis. Some examples from extraction with liquids in the standard state and trapping in the supercritical state (collection) have been furnished with the wrong extractant, modifier, or trapper which is presented for illustration of inappropriate choice of extractants, modifiers, and trappers.

The New Type of Liquid Chromatography—Colloidal High-Performance Liquid Chromatography (CHPLC): Further Applications

The recent paper by I. Brondz (2018), “One-Step Procedure for Direct Purification of Pediocin-Like Bacteriocins and Cationic Antimicrobial Peptides from Complex Culture Medium on an Analytical, Semipreparative, and Preparative Scale. A New Type of Liquid Chromatography—Colloidal High-Performance Liquid Chromatography (CHPLC)” (International Journal of Analytical Mass Spectrometry and Chromatography, 6, 41-49, https://***/10.4236/ijamsc.2018.63004), described a new type of CHPLC. This technique allows colloidal liquids and suspensions to be chromatographed directly despite the presence in the liquids of material such as bacteria, fungi, and other soft and hard particles. The significance of this development lies in enabling the single-step cleanup and concentration of the target substance from a complex mixture of soluble molecules in the presence of insoluble particles by high performance liquid chromatography (HPLC). The technique also allows the use of viscose liquids (as described in Brondz (2018) that are not suitable for analysis by conventional HPLC. In the previous paper, emphasis was placed on describing the applications of the techniques for the preparation of target substances such as small peptides, bacteriocins, bacitracin, and lysosome. Normally, the industrial preparation of these substances requires multistep procedures, which are time- and labor-consuming, and typically results in significant loss of target material and specific activity. In the present paper, the application of CHPLC for the isolation of alkaloids from crude raw material such as opium cake is demonstrated. In the opium cake, large amounts of hard vegetable particles and even sand corns are present together with the target alkaloids. Despite this, isolation by CHPLC of the desired compound was achieved in a single step by using a water/ethanol-based liquid. Analytical Methods in the Quality Control of Scientific Publications Part V: The Fraud of Pseudoscientists Based on False Measurements and Method Development

Despite the obvious frauds published in [1] [2] and the involvement of Editors and Editors in Chief of the journals in the publication of these fraudulent papers, the Editors in Chief of publishers have not taken any action to retract the fraudulent papers or to dismiss the corrupt editors. By this passivity, these Editors in Chief are supporting plagiarism, dissemination of fraudulent data, lies and making published information unreliable. In the present paper, we evaluate examples of bold, fraudulent publications and give the names of some Editors directly participating in concealing fraudulent publications, together with the names of Editors in Chief who also concealed and covered the facts of fraud and were reluctant to remove the fraudulent papers from circulation or to remove cheaters from editorial positions. The truth is universal and international;in contrast, the lie is individual and partisan, social, political, confessional, cultural and dirty. Knowledge is a truth that is part of the Universe. Lies are a tool of manipulation and can exist only in distinct environments that produce and support them. In this paper, we will show how artificial prefabricated analytical procedures were used to disseminate false data with the aim of substituting the truth with fraud.

A neurophysiological detector (NPD) is a hybridization of olfactory system neurons of the fish crucian carp, Carassius carassius L., with a computerized electronic device connected to a high-performance liquid chromatograph (HPLC). This system makes it possible to measure neurophysiological activities in the olfactory system of C. carassius L. after exposure of this fish to alarm pheromones. During and after every military conflict or war, there appears to be rapports about military crimes which were done by military and civil personal of conflicting sides. The basis of these offences is different;however the common to these is psychological and psychical background. The psychological background is an indoctrination of troopers that all what they are doing is right, well for country, nation or defends religion, or ideology. The life and property of enemy (opponent) are less worth than their oven. But most important in this indoctrination is the direct or indirect insurances about the absence of responsibility and promises about free for punishment for these actions. These aspects are well known and well-studied. The psychical background is less studied and more diverse;however in all cases the components of stress are present. The stress can be physical, psychical or toxic. The physical and psychical stress of engaged in military conflict personal is well studied, its action both during the war and as postwar syndrome have sufficient explanations. The action of intoxication especially by contaminants in medical forms, or toxins to induce the criminal behavior of military personal is nearly unstudied. The exceptions are alcohol, psychogenic substances as narcotics, LSD and special doping agents. This paper presented the evidences of additional toxic stress by contaminates in medical forms and intoxication by toxins of military personal engaged in different conflicts during last decades. The hypothesis about the influences of the additional toxic stress by medication with low quality pharmaceutical forms and some toxins on inducing the crimes generally and war crimes by military personal is launched in the paper. In this part the investigation will be concentrated on antimalarial drugs, especially on contaminated primaquine. One-Step Procedure for Direct Purification of Pediocin-Like Bacteriocins and Cationic Antimicrobial Peptides from Complex Culture Medium on an Analytical, Semipreparative, and Preparative Scale. A New Type of Liquid Chromatography—Colloidal High-Performance Liquid Chromatography (CHPLC)

A simple time- and labor-saving one-step procedure suitable for analytical-, semipreparative-, and preparative-scale preparation of bacteriocins and other cationic antibacterial peptides (for example as lysozyme) has been developed. Suspensions of bacterial cells, other colloidal- and fast-particle materials, and dissolved anionic molecules passed through the column;however, cationic bacteriocins were separated by chromatography. The purity of the target substance at more than 97% was achieved by the judgment of spectral, chromatographic, and antimicrobial activity. A many-fold concentration increase was achieved for bacteriocins, lysozyme, and bacitracin (a commercial antibiotic) in comparison with the starting solutions (see the text below). In the experiments, homemade cation-exchange columns have been used. The columns have been prepared by the grafting of glass fibers with a diameter of 0.2 μm. In this paper, applications of a new type of liquid chromatography—colloidal high-performance liquid chromatography (CHPLC)—were demonstrated.

Nowadays, physical chemistry is intensely discussing studies of macroscopic, atomic, subatomic, and particulate phenomena in chemical systems that are closely connected to clusters, clusters as host molecules for other guest molecules. The clusters as host molecules for other guest molecules could be described as adducts. Intermolecular forces, reaction kinetics and rates of reaction, surface chemistry and electrochemistry of membranes, the electrical conductivity of materials and the identity of ions, electrochemical cells, and many other phenomena depend on the presence and state of cluster molecules and adducts. This review discusses two popular books that give readers an introduction to the theory behind the subject. Together with this theory, several practical approaches for applications described in the above books are discussed and explained.
