High omega arachidonic acid/docosahexaenoic acid ratio induces mitochondrial dysfunction and altered lipid metabolism in human hepatoma cells

BACKGROUND Non-alcoholic fatty liver disease(NAFLD) is a common cause of liver disease worldwide and is a growing epidemic. A high ratio of omega-6 fatty acids to omega-3 fatty acids in the diet has been implicated in the development of NAFLD. However, the inflicted cellular pathology remains unknown. A high ratio may promote lipogenic pathways and contribute to reactive oxygen species(ROS)-mediated damage, perhaps leading to mitochondrial ***, these parameters were investigated to understand their contribution to NAFLD *** To examine the effect of increasing ratios of omega-6:3 fatty acids on mitochondrial function and lipid metabolism *** Hep G2-derived VL-17 A cells were treated with normal(1:1, 4:1) and high(15:1,25:1) ratios of omega-6: omega-3 fatty acids [arachidonic acid(AA):docosahexaenoic acid(DHA)] at various time points. Mitochondrial activity and function were examined via MTT assay and Seahorse XF24 analyzer, *** accumulation was determined by using Enzy Chrom? and levels of ROS were measured by fluorescence intensity. Protein expression of the mediators of lipogenic, lipolytic and endocannabinoid pathways was assessed by Western *** High AA:DHA ratio decreased mitochondrial activity(P < 0.01;up to 80%) and promoted intracellular triglyceride accumulation(P < 0.05;40%-70%).Mechanistically, it altered the mediators of lipid metabolism;increased the expression of stearoyl-Co A desaturase(P < 0.05;22%-35%), decreased the expression of peroxisome proliferator-activated receptor-alpha(P < 0.05;30%-40%) and increased the expression of cannabinoid receptor 1(P < 0.05;31%).Furthermore, the high ratio increased ROS production(P < 0.01;74%-115%) and reduced mitochondrial respiratory functions such as basal and maximal respiration, ATP production, spare respiratory capacity and proton leak(P < 0.01;35%-68%).CONCLUSION High AA:DHA ratio induced triglyceride accumulation, increased oxida

Tailored Functional Recipe (TFR) Approach to Delay the Progression of HIV to AIDS among People Living with HIV (PLWH) in Abuja, Nigeria

Background: HIV/AIDS is a pandemic disease and its scourge has had a devastating impact on health, nutrition, food security and overall socioeconomic development in affected countries. Moreover, intervention programmes, which simply employ antiretroviral drugs, have been found to lack effectiveness particularly when the patient is under-nourished. Aim and Purpose: This presented pilot intervention provides evidence that suggests use of local resources as therapeutic nutrition. This can act as a fundamental part of the comprehensive package of care at the country level. Methodology: Local ingredients, which were known for their availability, accessibility, micro and macro-nutrient strengths were selected and optimised into a nutritional functional meal (Amtewa). Daily consumption was ascertained to assess its effects on nutritional status and biomedical indices of the study participants (n = 100) who were/were not taking Highly Active Anti-retroviral Therapy (HAART). Findings: Mean CD4 count for ART-Test group at baseline and sixth months increased by 40.8 cells/mm3 while the ART-Control group decreased 18.12 cells/mm3. This positive outcome qualified Amtewa meal to the next phase of intervention (400 participants) to ascertain its effectiveness on health status of HIV infected subjects and appraise its position within the National Health Services framework as innovative approach to attenuate the progression of HIV to AIDS in Nigeria. Conclusion: Amtewa-based approach in HIV management is innovative, culturally relevant, reliable and requiring low technology in order to assure compliance, sustainability and cost effectiveness. Although the achieved results take the form of specific technology, it suggests that a prolong consumption of the intervention meal (Amtewa) will be suitable to sustain the gained improvements in MUAC and CD4 cell counts thereby slowing the progression of the disease.

Rainfall data is probably one of the longest-recorded climatic parameters in Lebanon. On the central coast of Lebanon, the Beirut weather station started collecting rainfall data in 1876. However, the recorded data is not available at one data provider source. Published data is found in historical documents but it reaches the early 1970s and then appears a data gap till 1990. Still the data is available, but it might be found to be saved privately. This study investigated the SPI variability on annual time scale between the years 1876 and 2021. The SPI was computed using R-Stat software to compare every year between 1876 and 2021. The majority (about 70% of the years) of the years are near normal in the precipitation rate. The Standardized Precipitation Index (SPI) demonstrated a normal distribution of years. Dry and wet years constitute about 15% of the total 146 years (1876-2021). Extremely dry years might appear in two consecutive years between 50 to 60 years count. After 1991, there were no wet years it was only near normal and few dry years. The last 30 years showed a trend of increasing drought years without any occurrence of wet years. This study demonstrated the importance of keeping records of at least rainfall data and it must be recorded on a daily basis or intensity on time. It is highly important on a managerial basis and for water security reasons to understand the drought event occurrence and investigate the changes in rainfall rates. Climate change scenarios always forecast a decrease in rainfall rates which will not appear without such studies.

Methane Flux and Its Environmental Impact Factors in Dajiuhu Wetland of Shennongjia

Methane(CH4) flux was measured by using the Eddy covariance technique in the Dajiuhu Wetland of Shennongjia Forestry District, Hubei Province, China. The seasonal and daily variation of CH4 emissions was investigated, and the correlation of CH4 fluxes with the environmental impact factors were discussed to understand the activity of CH4 flux and the effects of environmental factors including air temperature, soil temperature, photosynthetic photon flux density(PPFD), and rainfall on a wetland ecosystem. The results indicated that CH4 flux was significantly correlated with air temperature and photosynthetic photon flux density(PPFD), which may be the primary driver of controlling peatland CH4 flux.

Is chronic hepatitis C virus infection a risk factor for breast cancer?

AIM:To evaluate the prevalence of breast tumors in adult females with chronic hepatitis C virus(HCV) ***:Prospective,single-center study,based on female outpatients consulting in a liver unit,for 1 *** study group included females with present and/or past history of chronic infection by *** with spontaneous recovery were *** hepatitis had been proved by liver biopsy in the majority of cases and/or biological markers of inflammation and *** control group included female patients with other well documented chronic liver diseases:chronic hepatitis B,alcoholic liver disease,autoimmune hepatitis,hemochromatosis,non alcoholic liver disease,chronic *** patients were prospectively questioned during consultation about past breast history and follow-up by ***:Breast carcinoma was recorded in 17/294 patients with HCV infection(5.8%,95% CI:3.1-8.4) vs 5/107 control patients(4.7%,95% CI:0.67-8.67).Benign tumors of the breast(mastosis,nodules,cysts) were recorded in 75/294 patients with HCV infection(25.5%,95% CI:20.5-30.5) vs 21/107(19.6%,95% CI:12.1-27.1) in the control *** lesion was noted in 202 patients with HCV(68.7%,95% CI:63.4-74) vs 81 control patients(75.7%,95% CI:67.6-83.8).Despite a trend to an increased prevalence in the group with HCV infection,the difference was not significant compared to the control group(P=NS).In patients over 40 years,the results were,respectively,as follows:breast cancer associated with HCV:17/266 patients(6.3%,95% CI:3.4-9.3) vs 5/95 patients(5.2%,95% CI:0.7-9.7) in the control group;benign breast tumors:72/266 patients with HCV infection(27%,95% CI:21.7-32.4) vs 18/95 patients(18.9%,95% CI:11-26.8) in the control group;no breast lesion 177/266(66.5%,95% CI:60.9-72.2) in patients with HCV infection vs 72/95(75.7%,95% CI:67.1-84.4) in the control *** differences were not significant(P=NS).CONCLUSION:These results suggest that chronic HCV infectio
