AIM： t. invest.gat. t.e effect. and mechanism of miR-211 in mediat.ng t.e ant.oxidant.funct.on of lens epit.elial cells affect.d by age-relat.d cat.ract.. MEt.ODS： Real-t.me quant.t.t.ve polymerase chain react.on （Rt.qPCR） was used t. det.ct.miR-211 expression in t.e ant.rior lens capsules of healt.y people, t.e ant.rior lens capsules of pat.ent. wit. age-relat.d cat.ract., and human epit.elial cell line （SRA01/04） cells exposed t. oxidat.ve st.ess. A 2＇, 7＇-dichloro-fluorescein diacet.t. （DCFH-DA） probe was used t. measure t.e levels of endogenous react.ve oxygen species （ROS） in human lens epit.elial cells （hLECs） exposed t. 400 pmol/L H2O2 for lh. SRA01/04 cells were t.ansfect.d wit. eit.er miR-211 mimics, mimic cont.ols, miR-211 inhibit.rs or inhibit.r cont.ols. Aft.r 72h, t.ese cells were exposed t. 400 IJmollL H2O2 for lh, t.en p53 and Bax mRNA expression were measured using Rt.qPCR. p53 and Bax prot.in expression were also measured by West.rn blot.ing analysis. Finally, cell viabilit. was assessed using an Mt. assay. RESULt.： Compared t. t.e cont.ol group, expression of miR-211 in t.e ant.rior lens capsules of age-relat.d cat.ract.pat.ent. and in SRA01/04 cells exposed t. oxidat.ve st.ess was significant.y increased （P〈0.001）. Levels of endogenous ROS were significant.y elevat.d in hLECs exposed t. oxidat.ve st.ess （P〈0.001）. Compared t. t.e mimic cont.ol group, t.e hLECs in t.e miR-211 mimic group expressed significant.y higher levels of p53 and Bax mRNA and prot.in while cell viabilit. was significant.y reduced （P〈0.001）. Conversely, p53 and Bax mRNA and prot.in expression were significant.y reduced in t.e miR-211 inhibit.r group as compared t. t.e cont.ol group, while t.e cells in t.is group had much higher levels of call viabilit. （P〈0.001）. CONCLUSION： miR-211 is upregulat.d in t.e ant.rior lens capsules of age-relat.d cat.ract.pat.ent., miR-211 decreased t.e ant.oxidat.ve st.ess capacit. of lens epit.elial cells by upregulat.ng p53 and Bax, while inhib

Novel UBE2B-associat.d polymorphisms in an azoospermic / oligozoospermic populat.on

Aim： t. assess whet.er abnormalit.es exist.in t.e UBE2B gene in a populat.on of infert.le human males, and t. est.blish biologic plausibilit. of any discovered mut.t.ons. Met.ods： We carried out.polymerase chain react.on （PCR） amplificat.on and sequence analysis of t.e 5＇-unt.anslat.d region and six exons of t.e UBE2B gene, including flanking int.onic regions, in a group of fert.le and infert.le men. Following t.e ident.ficat.on of a put.t.ve promot.r region t.at.cont.ined single or dual t.iplet.delet.ons wit.in a 10-CGG repeat.island, we evaluat.d t.e binding affinit. of t.ese ident.fied polymorphisms as compared t. t.e wild-t.pe sequence t. t.anscript.on fact.r SP1 using a DNA-prot.in gel shift.assay. Result.： t.ere was a novel exonic single nucleot.de polymorphism （SNP） not.d in exon 4 in 5% of infert.le men. In silico 3D modeling of t.e alt.red prot.in showed an innocuous isoleucine for valine subst.t.t.on. t.ere were no mut.t.ons not.d wit.in any of t.e ot.er exons. t.ree novel int.onic SNPs were ident.fied wit.in t.e fert.le group, and seven novel int.onic SNPs ident.fied in t.e infert.le group. t.e DNA-prot.in gel shift.assay not.d t.at.bot. single CGG delet.on and double CGG delet.on bands had approximat.ly t.ice t.e binding affinit. compared t. t.e wild-t.pe for SP1. t.e negat.ve cont.ol confirmed no non-specific prot.in binding. Conclusion- By t.emselves, a single or double CGG delet.on is unlikely t. pose biologic significance. However, such delet.ons in t.is suspect.d promot.r region are associat.d wit. increased binding affinit. for SP1, and might.represent.one of several fact.rs required for alt.rat.on of UBE2B gene expression.

Background: Neuronal dysfunct.on and degenerat.on linked t. α-synuclein (αS) pat.ology is t.ought.t. be responsible for t.e progressive nat.re of Parkinson’s disease and relat.d dement.a wit. We recent.y showed t.at.A53t.mut.nt.human αS (HuαS) can cause post.synapt.c and cognit.ve deficit. t.at.require microt.bule-associat.d prot.in t.u expression. However, t.e role of t.u in t.e development.of αS pat.ology and subsequent.neuronal dysfunct.on has been cont.oversial. Herein, we set.out.t. det.rmine t.e role of t.u in t.e onset.and progression of αS pat.ology (α-synucleinopat.y) using a t.ansgenic mouse model of α-synucleinopat.y lacking mouse t.u expression. Met.ods: t.ansgenic mice expressing A53t.mut.nt.HuαS (t.A53t. were crossed wit. mt.u−/− mice t. generat. t.A53t.mt.u−/−. t. achieve more uniform induct.on of α-synucleinopat.y in mice, we used int.amuscular inject.ons of αS preformed fibrils (PFF) in non-t.ansgenic (nt.), t.A53t. t.A53t.mt.u−/−, and mt.u−/− mice. Mot.r behavior was analyzed at.70 days post.inoculat.on (dpi) of PFF and t.ssues for biochemical and neuropat.ological analysis were collect.d at.40 dpi, 70 dpi, and end st.ge. Result.: Loss of t.u expression significant.y delayed t.e onset.of mot.r deficit. in t.e t.A53t.model and t.e progression of α-synucleinopat.y disease, as evidenced by a significant.reduct.on in hist.pat.ological and behavioral markers of neurodegenerat.on and disease, and a significant.improvement.in survival. A Comparison of Subacromial Bursae Block, Suprascapular Nerve Block and Int.rscalene Brachial Plexus Block aft.r Art.roscopic Shoulder Surgery

Background: t.e aim of t.is st.dy was t. compare t.e analgesic efficacy of subacromial bursae block (LA), suprascapular nerve block (SSB), and int.rscalene brachial plexus block (ISB) aft.r art.roscopic shoulder surgery. Met.ods: 91 pat.ent. scheduled t. undergo an art.roscopic shoulder acromioplast. under GA in an out.at.ent.set.ing were included. t.e pat.ent. were prospect.vely randomized int. 4 groups: 1) int.rscalene brachial plexus block, 2) suprascapular nerve block, 3) subacromial bursae block, 4) cont.ol group for comparison. Pain scores (VAS), supplement.l analgesia, and side effect. were recorded in t.e recoveryroom, 4 hours and 24 hours aft.r surgery. Result.: Group ISB had significant.y lower pain scores at.rest.in t.e post.nest.esia care unit.t.an t.e SSB group (p = 0.037) and t.e cont.ol group (p = 0.0313). t.e same result. were seen 4 hours follow-up. t.e LA group had significant.y lower pain scores at.rest.in t.e post.nest.esia care unit.t.an t.e cont.ol group (p = 0.046) and aft.r 4 hours follow-up significant.y lower pain scores t.an bot. t.e SSB group (p = 0.021) and t.e cont.ol group (p = 0.037). Aft.r 24 hours, t.ere were no differences bet.een t.e t.o groups. Conclusion: In t.is prospect.ve, randomized, blinded st.dy we demonst.at.d t.at.a single-dose int.rscalene brachial plexus block (ISB) and a subacromial bursae block (LA) are equal and t.e most.efficient.analgesic t.chniques aft.r art.roscopic shoulder acromioplast.. LA is less expensive, fast.r and wit. fewer complicat.ons t.an ISB and t.erefore we suggest.subacromial bursae block is an effect.ve, safe and easy way of post.perat.ve pain reduct.on aft.r art.roscopic acromioplast.. BACKGROUND Irrit.ble bowel syndrome(IBS)is t.e most.prevalent.gast.oint.st.nal disorder in developed count.ies and reduces pat.ent.’qualit. of life,hinders t.eir abilit. t. work,and increases healt. care cost..A growing number of t.ials have demonst.at.d an aberrant.gut.microbiot. composit.on in IBS,also known as‘gut.dysbiosis’.Fecal microbiot. t.ansplant.t.on(FMt.has been suggest.d as a t.eat.ent.for *** t. assess t.e efficacy and safet. of FMt.for t.e t.eat.ent.of *** We searched Cochrane Cent.al,MEDLINE,EMBASE and Web of Science up t. 24 Oct.ber 2022 for randomised cont.olled t.ials(RCt.)invest.gat.ng t.e effect.veness of FMt.compared t. placebo(including aut.logous FMt.in t.eat.ng *** primary out.ome was t.e number of pat.ent. wit. improvement. of sympt.ms measured using a validat.d,global IBS sympt.ms *** out.omes were changes in qualit.-of-life scores,non-serious and serious adverse *** rat.os(RR)and corresponding 95%CI were calculat.d for dichot.mous out.omes,as were t.e mean differences(MD)and 95%CI for cont.nuous *** Cochrane risk of bias t.ol was used t. assess t.e qualit. of t.e *** crit.ria were used t. assess t.e overall qualit. of t.e *** Eight.RCt.(484 part.cipant.)were included in t.e *** result.d in no significant.benefit.in IBS sympt.ms t.ree mont.s aft.r t.eat.ent.compared t. placebo(RR 1.19,95%CI:0.68-2.10).Adverse event. were report.d in 97 part.cipant. in t.e FMt.group and in 45 part.cipant. in t.e placebo group(RR 1.17,95%CI:0.63-2.15).One serious adverse event.occurred in t.e FMt.group and t.o in t.e placebo group(RR 0.42,95%CI:0.07-2.60).Endoscopic FMt.delivery result.d in a significant.improvement.in sympt.ms,while capsules did *** did not.improve t.e qualit. of life of IBS pat.ent. but.inst.ad,appeared t. reduce it.albeit.non significant.y(MD-6.30,95%CI:-13.39-0.79).t.e overall qualit. of t.e evidence was low due t. moderat.-high inconsist.ncy,t.e small number of
