High-throughput preparation of plasmid DNA libraries for next-generation sequencing(NGS)is an important capability for molecular biology *** particular,it is an essential quality control(QC)check when large numbers of plasmid variants are being ***,we describe the use of the Design of Experiments(DOE)methodology to optimise the miniaturised preparation of plasmid DNA libraries for NGS,using the Illumina®Nextera XT technology and the Labcyte Echo®acoustic liquid dispensing ***,we describe methods which can be implemented as a QC check for identifying the presence of genomic DNA(gDNA)in plasmid DNA samples and the subsequent shearing of the gDNA,which otherwise prevents the acoustic transfer of plasmid *** workflow enables the preparation of plasmid DNA libraries which yield high-quality sequencing data.

A wave staff to be anchored at sea and containing sensing and telemetering equipment is described. This gives a record at the land station of water level changes due to tides and of waves as they pass the staff. The staff is a 13 metre long PVC tube, the upper half comprising a capacitance with inner plate a foil layer, dielectric the tube wall, and outer electrode the sea. 本文研究了pH值,亚硫酸钠浓度,油浴温度和时间对机械浆脱除树脂的影响,把吸附有辐射松(Radiata pine)树脂的滤纸和处理液置于密闭的容器中,然后在油浴器中加热处理,测定留存在滤纸上的树脂的量,在这一系列试验中,发现仅pH值明显影响树脂的脱除,且它们之间成正比,特别在pH值为7至10之间,脱除树脂的量迅速增加。

1945年新成立的Manawatu流域理事会(与土壤和河流管理委员会合作),曾在浦汗吉业(Pohangina)县租用了一块供土壤保持研究的丘陵地.Te Awa试验区正是该丘陵地的一普通溪谷陡峭地带.它的主坡的坡度为25—45度.土质为松砂壤,过去在原始森林覆盖下,自从森林被砍伐,以及十九世纪八十年代森林起火被烧后,这里进入放牧时期.遗留下的残桩使溪谷地表土层得以保持,并有助于牵制坡地上的表土与底土.

The light brown apple moth(LBAM),Epiphyas postvittana(Walker)(Lepidoptera: Tortricidae),invaded the state of California at least by early 2007,if not sooner,according to trapping *** a *** research has been undertaken to develop tools useful in eradicating or suppressing this pest,a native of *** previous work has investigated the potential for employing chemically based means such as bait sprays,lure and kill mass trapping,and,especially,mating disruption to control LBAM and related leafroller pests in Australia and *** desired to add an additional environmentally friendly tool to the arsenal against this pest- the sterile insect technique. In a collaborative effort involving our team plus colleagues from the California *** Food and Agriculture(CDFA) and the USDA’s Animal and Plant Health Inspection Service (APHIS),we are successfully rearing colonies of LBAM and are utilizing the moths obtained for various studies,including:(1) adapting existing rearing procedures to mass rearing conditions,(2) determining effective minimal doses of irradiation needed to sterilize adult moths,and(3) conducting field cage mating and other quality control tests to measure sterile moth *** progress has been made in each of these areas of investigation with satisfactory results,leading to plans to conduct pilot releases of sterile LBAM moths in certain infested areas of California beginning in the summer of 2009.
