Nitric oxide(NO)has emerged as an important signal molecule in plants,having myriad roles in plant *** addition,NO also orchestrates both biotic and abiotic stress responses,during which intensive cellular metabolic reprogramming *** to these responses is the location of NO biosynthetic and scavenging pathways in diverse cellular compartments,enabling plants to effectively organize signal transduction *** regulates plant metabolism and,in turn,metabolic pathways reciprocally regu-late NO accumulation and ***,these diverse cellular processes are inextricably *** re-view addresses the numerous redox pathways,located in the various subcellular compartments that pro-duce NO,in addition to the mechanisms underpinning NO *** focus on how this molecular dance is integrated into the metabolic state of the *** this context,a reciprocal relationship be-tween NO accumulation and metabolite production is often *** also showcase cellular pathways,including those associated with nitrate reduction,that provide evidence for this integration of NO function and ***,we discuss the potential importance of the biochemical reactions governing NO levels in determining plant responses to a changing environment.

The Involvement of Lipids in Alzheimer's Disease

It has been estimated that Alzheimer＇s disease （AD）, the most common form of dementia, will affect approximately 81 million individuals by 2040. To date, the actual cause and cascade of events in the progression of this disease have not been fully determined. Furthermore, there is currently no definitive blood test or simple diagnostic method for AD. Considerable efforts have been put into proteomic approaches to develop a diagnostic blood test, but to date these efforts have not been successful. More recently, there has been a stronger focus on lipidomic studies in the hope of increasing our understanding of the underlying mechanisms leading to AD and developing an AD blood test. It is well known that the strongest genetic risk factor for AD is the e4 variant of apolipoprotein E （APOE）. Evidence suggests that the ApoE protein, a major lipid transporter, plays a key role in the pathogenesis of AD, and its role in both normal and aberrant lipid metabolism warrants further extensive investigation. Here, we review ApoE-lipid interactions, as well as the roles that lipids may play in the pathogenesis of AD.

Convolutional neural network to learn building-shape representations for early-stage energy design

Machine learning(ML)supports energy-efficient design by quickly predicting the building performance at early ***,existing ML approaches cannot easily learn geometrical representations and their interactions with technical specifications for predicting *** approaches of artificial neural networks(ANN)use either numerical parameters to represent building geometry or component-based ML(CBML)to train trans-ferrable ML ***,both approaches have high generalisation errors for complex *** used a convolutional neural network(CNN)to learn geometrical representations of buildings and an autoencoder to optimise the training *** test existing approaches of ANN and CBML with a proposed approach of CNN for different sizes of training *** the biggest training dataset,ANN,CBML,and CNN have generalisation errors(root-mean-squared errors)of 1.35,1.60,and 0.77 kWh/a.m 2,*** CBML has lower prediction errors on small datasets,CNN is more suitable for large ***,CNN most often has better prediction accuracy than ANN and trains faster on encoded *** CNN captures the effects of building geometry on energy predictions better than ANN and CBML approaches,it is an attractive choice for early-stage energy design.

Deep learning based classification of sheep behaviour from accelerometer data with imbalance

Classification of sheep behaviour from a sequence of tri-axial accelerometer data has the potential to enhance sheep *** behaviour is inherently imbalanced(e.g.,more ruminating than walking)resulting in underperforming classification for the minority activities which hold *** works have not addressed class imbalance and use traditional machine learning techniques,e.g.,Random Forest(RF).We investigated Deep Learning(DL)models,namely,Long Short Term Memory(LSTM)and Bidirectional LSTM(BLSTM),appropriate for sequential data,from imbalanced *** data sets were collected in normal grazing conditions using jaw-mounted and earmounted *** to this study,alongside typical single classes,e.g.,walking,depending on the behaviours,data samples were labelled with compound classes,e.g.,walking_-*** number of steps a sheep performed in the observed 10 s time window was also recorded and incorporated in the *** designed several multi-class classification studies with imbalance being addressed using synthetic *** models achieved superior performance to traditional ML models,especially with augmented data(e.g.,4-Class+Steps:LSTM 88.0%,RF 82.5%).DL methods showed superior generalisability on unseen sheep(i.e.,F1-score:BLSTM 0.84,LSTM 0.83,RF 0.65).LSTM,BLSTM and RF achieved sub-millisecond average inference time,making them suitable for real-time *** results demonstrate the effectiveness of DL models for sheep behaviour classification in grazing *** results also demonstrate the DL techniques can generalise across different *** study presents a strong foundation of the development of such models for real-time animal monitoring.

Nicotiana attenuata's capacity to interact with arbuscular mycorrhiza alters its competitive ability and elicits major changes in the leaf transcriptome

To study the local and systemic effects of arbuscular mycorrhizal fungal （AMF） colonization, Nicotiana attenuata plants impaired in their interactions with AMF due to silencing of a calcium- and calmodulin dependent protein kinase （inverted repreat （ir）CCaMK） were grown competitively in pairs with empty vector （EV） plants, with and without two different types of inoculum. When inoculated, EV plants strongly outperformed irCCaMK plants. Foliar transcript profiling revealed that AMF colonization significantly changed gene expression of P-starvation and -transporter genes in irCCaMK plants. The Phtl family phosphate transporter NaPT5 was not only specifically induced in roots after AMF colonization, but also in leaves of AMF-colonized irCCaMK plants, and in plants grown under low Pi conditions in the absence of AMF. The P-starvation signature of inoculated irCCaMK plants corresponded with increases in selected amino acids and phenolic compounds in leaves. We also found a strong AMF-induced increase in amino acids and phenolic metabolites in roots. Plants impaired in their interactions with AMF clearly have a fitness disadvantage when competing for limited soil nutrients with a fully functional isogenic line. The additional role of the AMF-induced Phtl family transporter NaPT5 in leaves under P-starvation conditions will require further experiments to fully resolve.

Over 100 Million Years of Enzyme Evolution Underpinning the Production of Morphine in the Papaveraceae Family of Flowering Plants

Phylogenomic analysis of whole genome sequences of five benzylisoquinoline alkaloid(BIA)-producing species from the Ranunculales and Proteales orders of flowering plants revealed the sequence and timing of evolutionary events leading to the diversification of these compounds.(S)-Reticuline is a pivotal intermediate in the synthesis of many BIAs and our analyses revealed parallel evolution between the two orders,which diverged122 million years ago(MYA).Berberine is present in species across the entire Ranunculales,and we found co-evolution of genes essential for production of the protoberberine *** benzophenanthridine class,which includes the antimicrobial compound sanguinarine,is specific to the Papaveraceae family of Ranunculales,and biosynthetic genes emerged after the split with the Ranunculaceae family110 MYA but before the split of the three Papaveraceae species used in this study at77 *** phthalideisoquinoline noscapine and morphinan class of BIAs are exclusive to the opium poppy *** estimation of paralogous pairs indicates that morphine biosynthesis evolved more recently than 18 MYA in the Papaver *** the preceding 100 million years gene duplication,neofunctionalization and recruitment of additional enzyme classes,combined with gene clustering,gene fusion,and gene amplification,resulted in emergence of medicinally valuable BIAs including morphine and noscapine.

Enhanced Electroactivity of Facet-Controlled Co3O4 Nanocrystals for Enzymeless Biosensing

In this work, we report enhanced electroactivity of Co304 nanocrystals （nanocubes, NCs and truncated nano-octahedra, TNO） on the exposed {111} facets as compared to the {001} facets in relation to the surface density and the activity of the octahedral Com species. Transmission electron microscopy, X-ray powder diffraction, X-ray photoelectron spectroscopy and Fourier-transform infrared spectroscopy were em- ployed to characterize the crystal facets and materials properties. The enhanced electroactivity of {111 } crystal facets was evaluated by cyclic voltammetry and amperometric titration. Our results indicate that the {111 } facets in TNO has a better electroactivity for enzymeless glucose sensing with a decent glucose sensitivity of 32.54 μA （mmol/L）-1 cm-2.

Seasonal variation of tidal prism and energy in the Changjiang River estuary： a numerical study

Tidal rivers are intrinsically complex because tidal propagation is influenced by river discharge. This study aims to examine the seasonal variation of tidal prism and energy variance in the tidal river of the Changjiang(Yangtze) River estuary in China. In order to quantify the behaviour of river and tide,we use numerical modelling that has been validated using measured data. We conduct our analysis by quantifying the discharge and energy variance in separate components for both the river and the tide,during wet and dry seasons. We note various definitions of tidal prism and explore the difference between tidal discharge on the flood and ebb and tidal storage volume. The results show that the river discharge attenuates the tidal motion and reduces the tidal flood discharge but the tidal storage volume is approximately constant with different riverine discharge since part of the fresh water discharge is intercepted and captured in the estuary due to the backwater effect. It appears that the tidal discharge adjusts according to the variation of river discharge to keep a constant tidal storage volume. An analysis of the hydraulics shows that the transition from tidal dominance(at the mouth) to river dominance(upstream) depends on the location of tidal current reversal which varies from wet season to dry season. Duringthe wet season,the Changjiang River estuary is totally dominated by energy from fresh water discharge.

Evidence for the ef?cacy and safety of herbal weight loss preparations

Rising rates of obesity across the globe have been associated with an increase in the use of herbal preparations for weight control. However, the mechanisms of action for these substances are often not known,as is the potential for interaction with other herbal preparations or prescription pharmaceutical drugs. To investigate the reported efficacy and safety of herbal weight loss preparations, we conducted a review of the literature focusing on herbs that are most commonly used in weight loss preparations, specifically,Garcinia cambogia, Camellia sinensis, Hoodia gordonii, Citrus aurantium and Coleus forskohlii. There was no clear evidence that the above herbal preparations would cause sustained long-term weight loss in humans in the long term. Serious illness and even death have occasionally resulted from the use of herbal weight loss preparations. Few clinical trials have been undertaken to evaluate the efficacy and/or safety of herbal weight loss preparations. In addition, potential issues of herb–herb and herb–drug interactions are often not considered. Regulation of these products is much less rigorous than for prescription medications, despite documented cases of associated hepatotoxicity.
