A number of eukaryotic expression vectors have been developed for use as DNA vaccines. They showed varying abilities to initiate immune responses;however, there is little data to indicate which of these vectors will be the most useful and practical for DNA vaccines in different species. This report examines the use of five expression vectors with different promoters and Kozak sequence to express the same hemagglutinin (hA) protein of an h6N2 avian influenza virus for DNA vaccination in chickens. Although intramuscular vaccination with seven DNA constructs elicited no or limited measurable h6 hA antibody responses in hy-Line chickens, variable reduction in virus shedding for either oropharyngeal or cloacal swabs post-virus challenge were observed. This indicated that all DNA constructs generated some levels of protective immunity against homologous virus challenge. Interestingly, lower dose (50 or 100 μg) of plasmid DNAs consistently induced better immune response than higher dose (300 or 500 μg). In the transfection experiments there appeared to be a hierarchy in the in vitro expression efficiency in the order of pCAG-optihAk/ pCAG-hAk > pCI-hAk > VR-hA > pCI-hA > pCI-neo-hA > pVAX-hA. Since the level of in vitro expression correlates with the level of immune response in vivo, in vitro expression levels of the DNA constructs can be used as an indicator for pre-selection of plasmid vaccines prior to in vivo assessment. Moreover, our results suggested that the Kozak sequence could be used as an effective tool for DNA vaccine design.

hydrogen sulfide is synthesized endogenously in mammals and has been shown to have both physiological and pathological functions. So far there has been little agreement as to the actual levels of endogenous sulfide under physiological or pathological conditions;this is partly due to the complexity involved in measuring free sulfides due to h2S volatility, oxidation, reactivity and the presence of bound labile sulfur in tissues. In this report we describe a method of measuring free tissue sulfides using a zinc sulfide precipitation and wash method. It is an indirect method that measures the sulfide difference between samples prepared at ph 9 and ph 6, assuming that at ph 9 free sulfides would be retained in solution, while at ph 6 free sulfides would volatilize during sample preparation. Using this approach we were able to measure appreciable amounts of free sulfides in mouse: lung, pancreas, liver and kidney at 0.036 + 0.006, 0.082 + 0.009, 0.215 + 0.016 and 0.323 + 0.031 nmole per mg of tissue respectively (n = 6).

Free-space optical(FSO)communication technologies constitute a solution to cope with the bandwidth demand of future satellite-ground *** may overcome the RF bottleneck and attain data rates in the order of Tbit/s with only a handful of ground ***,we demonstrate single-carrier Tbit/s line-rate transmission over a free-space channel of 53.42 km between the Jungfraujoch mountain top(3700 m)in the Swiss Alps and the Zimmerwald Observatory(895 m)near the city of Bern,achieving net-rates of up to 0.94 Tbit/*** this scenario a satellite-ground feeder link is mimicked under turbulent *** adverse conditions high throughput was achieved by employing a full adaptive optics system to correct the distorted wavefront of the channel and by using polarization-multiplexed high-order complex modulation *** was found that adaptive optics does not distort the reception of coherent modulation ***,we introduce constellation modulation–a new four-dimensional BPSK(4D-BPSK)modulation format as a technique to transmit high data rates under lowest *** way we show 53 km FSO transmission of 13.3 Gbit/s and 210 Gbit/s with as little as 4.3 and 7.8 photons per bit,respectively,at a bit-error ratio of 1∙10−*** experiments show that advanced coherent modulation coding in combination with full adaptive optical filtering are proper means to make next-generation Tbit/s satellite communications practical.

We have developed a noninv asive imaging method to quantify in viwo drug delivery pharmaco-kinetics without the need for blood or tissue collection to determine drug *** combining the techniques of hy perspectral imaging and a dorsal skinfold window chamber,this method enabled the real-time monitoring of vascular transport and tissue deposition of nano-particles labeled with near infrared(NIR)*** this imaging method,we quantified the delivery pharmacokinetics of the native high-density lipoprotein(hDL)and epidermal growth factor receptor(EGFR)-targeted hDL nanoparticles and demonstrated these hDLs had long circulation time in blood stream(half-life>12h).These hDL nanoparticles could eficiently carry cargo DiR-BOA to extravasate from blood vesels,difuse through extr acellular matrix,and penetrate and be retained in the tumor *** EGFR targeting specificity of EGFR-targeted hDL(EGFR-specific peptide conjugated hDL)was also visualized in vivo by competitive inhi bition with excess EGFR specifc *** summary,this imaging technology may help point the way toward the development of novel imaging based pharmacokinetic assays for preclinical drugs and evaluation of drug delivery eficiency,providing a dynamic window into the devel opment and application of novel drug delivery systems.

AIM： To investigate the effects and mechanism of miR-211 in mediating the antioxidant function of lens epithelial cells affected by age-related cataracts. METhODS： Real-time quantitative polymerase chain reaction （RT-qPCR） was used to detect miR-211 expression in the anterior lens capsules of healthy people, the anterior lens capsules of patients with age-related cataracts, and human epithelial cell line （SRA01/04） cells exposed to oxidative stress. A 2＇, 7＇-dichloro-fluorescein diacetate （DCFh-DA） probe was used to measure the levels of endogenous reactive oxygen species （ROS） in human lens epithelial cells （hLECs） exposed to 400 pmol/L h2O2 for lh. SRA01/04 cells were transfected with either miR-211 mimics, mimic controls, miR-211 inhibitors or inhibitor controls. After 72h, these cells were exposed to 400 IJmollL h2O2 for lh, then p53 and Bax mRNA expression were measured using RT-qPCR. p53 and Bax protein expression were also measured by Western blotting analysis. Finally, cell viability was assessed using an MTS assay. RESULTS： Compared to the control group, expression of miR-211 in the anterior lens capsules of age-related cataract patients and in SRA01/04 cells exposed to oxidative stress was significantly increased （P〈0.001）. Levels of endogenous ROS were significantly elevated in hLECs exposed to oxidative stress （P〈0.001）. Compared to the mimic control group, the hLECs in the miR-211 mimic group expressed significantly higher levels of p53 and Bax mRNA and protein while cell viability was significantly reduced （P〈0.001）. Conversely, p53 and Bax mRNA and protein expression were significantly reduced in the miR-211 inhibitor group as compared to the control group, while the cells in this group had much higher levels of call viability （P〈0.001）. CONCLUSION： miR-211 is upregulatsd in the anterior lens capsules of age-related cataract patients, miR-211 decreased the antioxidative stress capacity of lens epithelial cells by upregulating p53 and Bax, while inhib

Background: Obesity is associated with higher end-stage renal disease incidence, but associations with earlier forms of kidney disease remain incompletely characterized. Methods: We studied the association of body mass index (BMI), waist circumference (WC), and waist-to-hip ratio (WhR) with rapid kidney function decline and incident chronic kidney disease in 4573 non-diabetic adults with eGFR ≥ 60 ml/min/1.73m2 at baseline from longitudinal Multi-Ethnic Study of Atherosclerosis cohort. Kidney function was estimated by creatinine and cystatin C. Multivariate analysis was adjusted for age, race, baseline eGFR, and hypertension. Results: Mean age was 60 years old, BMI 28 kg/m2, baseline eGFRCr 82 and eGFRCys 95 ml/min/1.73m2. Over 5 years of follow up, 25% experienced rapid decline in renal function by eGFRCr and 22% by eGFRCys. Incident chronic kidney disease (CKD) developed in 3.3% by eGFRCys, 11% by eGFRCr, and 2.4% by both makers. Compared to persons with BMI 25, overweight (BMI 25 - 30) persons had the?lowest risk of rapid decline by eGFRCr (0.84, 0.71 - 0.99). In contrast, higher BMI categories were associated with stepwise higher odds of rapid decline by eGFRCys, but remained significant only when BMI ≥ 35 kg/m2 (1.87, 1.41 - 2.48). Associations of BMI with incident CKD were insignificant after adjustment. Large WC and WhR were associated with increased risk of rapid decline only by eGFRCys, and of incident CKD only when defined by both filtration markers. Conclusions: Obesity may be a risk factor for kidney function decline, but associations vary by filtration marker used.

Photonics integrated circuitry would benefit considerably from the ability to arbitrarily control waveguide cross-sections with high precision and low loss,in order to provide more degrees of freedom in manipulating propagating ***,we report a new method for femtosecond laser writing of optical-fiber-compatible glass waveguides,namely spherical phase-induced multicore waveguide(SPIM-WG),which addresses this challenging task with three-dimensional on-chip light *** in the heating regime with high scanning speed,precise deformation of cross-sections is still achievable along the waveguide,with shapes and sizes finely controllable of high resolution in both horizontal and vertical transversal *** observed that these waveguides have high refractive index contrast of 0.017,low propagation loss of 0.14 dB/cm,and very low coupling loss of 0.19 dB coupled from a single-mode ***-WG devices were easily fabricated that were able to perform on-chip beam rotation through varying angles,or manipulate the polarization state of propagating light for target *** also demonstrated SPIM-WG mode converters that provide arbitrary adiabatic mode conversion with high efficiency between symmetric and asymmetric nonuniform modes;examples include circular,elliptical modes,and asymmetric modes from ppKTP(periodically poled potassium titanyl phosphate)waveguides which are generally applied in frequency conversion and quantum light *** inside optical glass,these waveguides and devices have the capability to operate across ultra-broad bands from visible to infrared *** compatibility with optical fiber also paves the way toward packaged photonic integrated circuitry,which usually needs input and output fiber connections.
