The role of ultrasonic bat detectors in improving inventory and monitoring surveys in Vietnamese karst bat assemblages

Bats account for 30% of mammal diversity in SE Asia and are potential bioindicators of wider biodiversity impacts resulting from habitat loss and climate change. As existing sampling techniques in the region typically fail to record bats that habitually fly in open areas and at higher altitudes, current inventory efforts are less than comprehensive. Acoustic sampling with bat detectors may help to overcome these limitations for insectivorous bats, but has yet to be tested in mainland SE Asia. To do so, we sampled bats while simultaneously recording the echolocation calls of insectivorous species commuting and foraging in a variety of karst habitats in north Vietnam. Monitoring of cave-dwelling bats was also undertaken. Discriminant function analysis of 367 echolocation calls produced by 30 insectivorous species showed that acoustic identification was feasible by correctly classifying 89. 1% of calls. In all habitats, acoustic sampling and capture methods recorded significantly more species each night than capture methods alone. Capture methods consequently failed to record 29% （ten spp. of aerial insectivores） of the bat fauna in commuting and foraging habitats and 11% （two spp. ） of that in our cave sample. Only four of these species were subsequently captured following significantly greater sampling effort. This strongly suggests that acoustic methods are indispensable for maximizing bat inventory completeness in SE Asia. As accurate inventories and monitoring are essential for effective species conservation, we recommend the inclusion of acoustic sampling in future studies of bat assemblages across the region [ Current Zoology 55 （5）： 327 - 341, 2009].

Comparison of RF3 <i>B. juncea</i>to RF3 <i>B. napus</i>

RF3 Brassica napus L. (InVigorTM Canola) has been in commercial cultivation since 1997. RF3 B. napus was produced by Agrobacterium-mediated transformation to provide glufosniate ammonium herbicide tolerance to the crop. Canola quality (CQ) varieties of Brassica juncea L. (Czern & Coss), a close relative of B. napus, have been available in Canada for more than 20 years. Currently, BASF Agricultural Solutions is developing CQ RF3 B. juncea via conventional breeding with RF3 B. napus to provide similar herbicide tolerance in the B. juncea species. The insertion event RF3 contains the bar gene (origin Streptomyces hygroscopicus) coding for phosphinothricin acetyl transferase (PAT/bar) protein which confers tolerance to glufosinate-ammonium herbicides. RF3 also contains the barstar gene (origin Bacillus amyloliquefaciens), coding for the Barstar protein, which is an inhibitor of the Barnase protein. In the absence of Barnase, there is no impact of Barstar. Many safety studies have been conducted during the development of the canola quality lines of RF3 B. napus and RF3 B. juncea. This report seeks to compare the two species based on the scientific data generated during the safety assessments of RF3 B. napus and RF3 B. juncea and discusses the utility of using studies with one species to support the safety of the other species both containing the same insertion event.

DNA was extracted from 52 thick primary melanomas and mutations sought in exon 15 of the BRAF (v-raf murine sarcoma viral oncogene homolog B1) gene using denaturing high performance liquid chromatograph (dHPLC) fragment analysis, sequencing, and allele-specific PCR. Exon 15 BRAF mutations were found in 13 of 52 (25% ) primary melanomas. These comprised five of 17 (29% ) superficial spreading melanomas, three of 11 (27% ) nodular melanomas, two of 13 (15% ) acral lentiginous melanomas, one of one (100% ) mucosal melanoma and two of 10 (20% ) lentigo maligna melanomas. In common with other groups, our findings show a relative concentration of the exon 15 BRAF mutation in superficial spreading and nodular melanomas, but add further evidence that this mutation not necessary for malignant transformation of the melanocyte.

Background: Up to 5% of patientswith melanoma have a family history of a first-degree relative also being affected. Objectives: To study such families for germline mutations, to help clarify the gene-environment interaction in melanoma aetiology. Methods: Thirty-two families in Scotland with melanoma in two or more first-degree relatives are reported for the first time. Peripheral blood DNA was extracted, and denaturing high-performance liquid chromatography analysis performed on exons 1 α and 2 of the CDKN2A gene and their splice junctions. The coding sequences and splice junctions of these exons were sequenced in all samples as confirmation of the chromatographic pattern observed. Results: Seven of the 32 melanoma families (22% ) have CDKN2A mutations. One mutation, H83N, which has not previously been described in melanoma families, was found in one family. In addition, two families have R112G mutations, one family has a G67R mutation, one has an exon 1 α 24-bp duplication where bases 9- 32 are duplicated between bases 32 and 33, and two families have M53I mutations, bringing the total of known Scottish families with the M53I mutation to six. Conclusions: This study brings the total of Scottish families investigated for germlinemutations to 48, and strongly suggests that the M53I mutation originated in Scotland.

Objective: The risk of stroke in patients with recently symptomatic carotid stenosis is considerably higher than in patients with asymptomatic stenosis. In the present study it was hypothesised that excessive platelet activation might partly contribute to this difference. Methods: A full blood count was done and whole blood flow cytometry used to measure platelet surface expression of CD62P, CD63, and PAC1 binding and the percentage of leucocyte-platelet complexes in patients with acute (0-21 days, n = 19) and convalescent (79-365 days) symptomatic (n = 16) and asymptomatic (n = 16) severe (≥70%) carotid stenosis. Most patients were treated with aspirin (37.5-300 mg daily) although alternative antithrombotic regimens were more commonly used in the symptomatic group. Results: The mean platelet count was higher in patients with acute and convalescent symptomatic compared with asymptomatic carotid stenosis. There were no significant differences in the median percentage expression of CD62P and CD63, or PAC1 binding between the acute or convalescent symptomatic and asymptomatic patients. The median percentages of neutrophil-platelet (p = 0.004), monocyte-platelet (p = 0.046), and lymphocyte-platelet complexes (p = 0.02) were higher in acute symptomatic than in asymptomatic patients. In patients on aspirin monotherapy, the percentages of neutrophil-platelet and monocyte-platelet complexes (p=0.03) were higher in acute symptomatic (n=11) than asymptomatic patients (n = 14). In the convalescent phase, the median percentages of all leucocyte-platelet complexes in the symptomatic group dropped to levels similar to those found in the asymptomatic group. Conclusion: Increased platelet count and leucocyte-platelet complex formation may contribute to the early excess risk of stroke in patients with recently symptomatic carotid stenosis.
