Type I strain of Toxoplasma gondii from chicken induced different immune responses with that from human,cat and swine in chicken

In this study,four strains of Toxoplasma gondii with the same genetic type（Type I） originated from chicken,human,cat and swine were used to compare the immune responses in resistant chicken host to investigate the relationships between the parasite origins and the pathogenicity in certain host.A total of 300,10-day-old chickens were allocated randomly into five groups which named JS（from chicken）,CAT（from cat）,CN（from swine）,RH（from human） and a negative control group（—Ve） with 60 birds in each *** of four different *** strains（JS,CAT,CN and RH） were inoculated intraperitoneally with the dose of 1×10~7 in the four designed groups,*** negative control（-Ve） group was mockly inoculated with phosphate-buffered saline（PBS） *** and spleen samples were obtained on the day of inoculation（day 0） and at days 4,11,25,39 and 53 post-infection to screen the immunopathological *** results demonstrated some different immune characters of *** infected chickens with that of mice or swine previous *** differences included up-regulation of major histocompatibility complex class Ⅱ（MHC Ⅱ） molecules in the early stage of infection,early peak expressions of interleukin（IL）-12（IL-12） and-10（IL-10） and long keep of *** might partially contribute to the resistance of chicken to *** *** to chickens infected with strains from human,cat and swine,chickens infected with strain from chicken showed significant high levels of CD4~＋ and CD8~＋ T cells,interferon gamma（IFN-γ）,IL-12 and *** suggested that the strain from chicken had different ability to stimulate cellular immunity in chicken.

Investigation on the co-infections of Toxoplasma gondii with PRRSV, CSFV or PCV-2 in swine in part of China

The objective of the present investigation was to estimate the prevalence of Toxoplasma gondii infection and co-infection with porcine reproductive and respiratory syndrome virus(PRRSV), classical swine fever virus(CSFV) and porcine circovirus type 2(PCV-2) in pigs in China. A total of 372 tissues or serum samples collected from pigs distributed in 9 provinces/municipalities of China during the period from February 2011 to November 2012 were assayed for T. gondii antigens and antibodies using enzyme linked immunosorbent assay(ELISA) technique, while the PCR was designed for the detection of the PRRSV, CSFV and PCV-2, respectively. The total positive rate of T. gondii, PRSSV, CSFV and PCV-2 was 9.14%(34/372), 50.00%(186/372), 37.10%(138/372) and 3.23%(12/372), respectively. Among the 34 T. gondii positive samples, 26 samples were simultaneously infected with T. gondii and viruses, while the remaining eight samples were infected with T. gondii alone. In addition, the co-infection rate of T. gondii with PRSSV, T. gondii with PRSSV and CSFV, T. gondii with PRSSV and PCV-2, T. gondii with CSFV and PCV-2, T. gondii with PRSSV, CSFV and PCV-2 was 1.61%(6/372), 4.03%(15/372), 0.27%(1/372), 0.27%(1/372) and 0.81%(3/372), respectively. The results of the present survey revealed that PRRSV and CSFV were the common pathogens co-existing with porcine toxoplasmosis in China, and both of them could increase the chances of T. gondii infection in pig. This is the first report of T. gondii co-infections with viruses in pigs. It is very important to understand the interactions of parasite and virus, and can be used as reference data for the control and prevention of co-infections of T. gondii and viruses in pigs.

Ion-Acoustic Rogue Waves in Multi-ion Plasmas

The basic properties of nonlinear ion-acoustic(ia) waves(iaWs), particularly finite amplitude ia rogue waves(iaRWs) in a plasma medium(containing pair ions, iso-thermal positrons, and non-thermal electrons) are theoretically investigated by deriving the nonlinear Schr¨odinger equation(NLSE).The criteria for the modulational instability of iaWs, and the basic features of finite amplitude iaRWs are *** modulationally stable and unstable regions are determined by the sign of the ratio of the dispersive coefficient to the nonlinear coefficient of *** latter is analyzed to obtain the region for the existence of the iaRWs, which corresponds to the unstable *** shape of the profile of the rogue waves depends on the non-thermal parameter α and the ratio of electron temperature to positron *** is found that the increase in the value of the non-thermal parameter enhances both the amplitude and width of iaRWs, and that the enhancement of electron(positron) temperature reduces(enhances) the amplitude and width of *** is worth to mention that our present investigation may be useful for understanding the salient features of iaRWs in space(viz., upper region of Titan’s atmosphere, cometary comae, and Earth’s ionosphere, etc.)and laboratory(viz., plasma processing reactor and neutral beam sources, etc.) plasmas.

Liver transplantation for non-resectable colorectal liver metastases

At present,there are no absolute practice guidelines for liver transplants in colorectal liver metastases(CRLM),and treatment protocols for unresectable CRLM are institution-specific(1).One of the first prospective studies evaluating the safety of liver transplants for CRLM was the SECA-I trial(1,2).A 5-year overall survival rate of 60% was observed in this trial,which far outperformed survival with chemotherapy alone(3).In the SECA-Ⅱ trial,enrollment was increased,and patients with better prognostic factors,including at least a 10% response to chemotherapy before the transplant,were selected.

Inflammatory bowel disease in rats:Bacterial and chemical interaction

AIM：To develop a novel model of colitis in rats, using a combination of iodoacetamide and enteropathogenic E. coli（EPEC）, and to elucidate the pathophysiologic processes implicated in the development of ulcerative colitis （UC）. METHODS： Hale Sprague-Dawley rats （/7 = 158） were inoculated intrarectally on a weekly basis with 4 different combinations：（a） 1% methylcellulose （HC）, （b） 100 μL of 6% iodoacetamide （ia） in 1% HC, （c） 200 p.L containing 4×10^8 colony factor units （CFU） of EPEC, and （d） combined treatment of （ia） followed by bacteria （13） after 2 d. Thirty days post treatment, each of the four groups was divided into two subgroups; the inoculation was stopped for one subgroup and the other subgroup continued with biweekly inoculation until the end of the experiment. Colitis was evaluated by the clinical course of the disease, the macroscopic and microscopic alterations, activity of myeloperoxidase （HPO）, and by TNF-α gene expression. RESULTS： Findings indicative of UC were seen in the combined treatment （ia ＋ B） as well as the ia continued treatment groups： the animals showed slow rate of increase in body weight, diarrhea, bloody stools, high colonic ulcer score, as well as histological alterations characteristic of UC, with an extensive inflammatory reaction. During the course of the experiment, the MPO activity was consistently elevated and the TNF-α gene expression was upregulated compared to the control animals. CONCLUSION： The experimental ulcerative colitis model used in the present study resembles, to a great extent, the human disease. It is reproducible with characteristics indicative of chronicity.

Objective:Well-defined germ-line mutations in thePTCH1 gene are associated with syndromic multiple basal cell carcinomas(BCCs).Here,we used whole exome sequencing(WES)to identify the role of patched-1 in patients with multiple,unusually large ***:A 72-year old patient presenting with numerous BCCs progressing to large ulcerating lesions was *** was used to identify the pathogenic gene ***:Genetic work-up by WES identified a homozygousPTCH1 nonsense mutation in the tumor tissue but not present in her blood cells or in non-lesional *** addition,heterozygous missense mutations were identified in three cancer-associated genes(EPHB2,RET,andGALNT12)in blood cells as well as in lesional and non-lesional *** also tested systemic immune therapy as a potentially beneficial approach to treat patients with numerous large BCCs on scatted areas of involvement.A rapid and sustained response to nivolumab was noted,suggesting that it is an efficacious drug for long-term therapeutic ***:PTCH1,EPHB2,RET,andGALNT12 may potentially contribute to the synergistic oncogene driven malignant transformation manifesting as multiple,unusually large BCCs.
