The scope for manipulating the polyunsaturated fatty acid content of beef:a review

Since 1950, links between intake of saturated fatty acids and heart disease have led to recommendations to limit consumption of saturated fatty acid-rich foods, including beef. Over this time, changes in food consumption patterns in several countries including Canada and the USA have not led to improvements in health. instead, the incidence of obesity, type ii diabetes and associated diseases have reached epidemic proportions owing in part to replacement of dietary fat with refined carbohydrates. Despite the content of saturated fatty acids in beef, it is also rich in heart healthy cis-monounsaturated fatty acids, and can be an important source of long-chain omega-3（n-3） fatty acids in populations where little or no oily fish is consumed. Beef also contains polyunsaturated fatty acid biohydrogenation products,including vaccenic and rumenic acids, which have been shown to have anticarcinogenic and hypolipidemic properties in cell culture and animal models. Beef can be enriched with these beneficial fatty acids through manipulation of beef cattle diets, which is now more important than ever because of increasing public understanding of the relationships between diet and health. The present review examines recommendations for beef in human diets, the need to recognize the complex nature of beef fat, how cattle diets and management can alter the fatty acid composition of beef, and to what extent content claims are currently possible for beef fatty acids.

BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. it is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in *** To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic *** Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase F(PNGase F). Prostaglandin E2(PGE2) levels were determined using a specific ELiSA. 1 H high resolution magic angle spinning(HRMAS) analysis was performed using a Bruker AViii 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-MS/MS) using a QExactive HF orbitrap *** Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. Brassinosteroid （BR） binding activates the receptor kinase BRi1 by inducing heterodimerization with its co- receptor kinase BAK1; however, the mechanisms that reversibly inactivate BRi1 remain unclear. Here we show that cytoplasm-localized protein phosphatase 2A （PP2A） B＇ regulatory subunits interact with BRi1 to mediate its dephosphorylation and inactivation. Loss-of-function and overexpression experiments showed that a group of PP2A B＇ regulatory subunits, represented by B＇η, negatively regulate BR signaling by decreasing BRi1 phosphorylation. BR increases the expression levels of these B＇ subunits, and B/Ti interacts preferentially with phosphorylated BRi1, suggesting that the dynamics of BR signaling are modu- lated by the PP2A-mediated feedback inactivation of BRi1. Compared with PP2A B＇α and B＇β, which promote BR responses by dephosphorylating the downstream transcription factor BZR1, the BRi1- inactivating B＇ subunits showed similar binding to BRi1 and BZR1 but distinct subcellular localization. Alteration of the nuclear/cytoplasmic localization of the B＇ subunits revealed that cytoplasmic PP2A de- phosphorylates BRi1 and inhibits the BR response, whereas nuclear PP2A dephosphorylates BZR1 and ac- tivates the BR response. Our findings not only identify the PP2A regulatory B subunits that mediate the binding and dephosphorylation of BRi1, but also demonstrate that the subcellular localization of PP2A specifies its substrate selection and distinct effects on BR signaling.

Cholangiocyte anion exchange and biliary bicarbonate excretion

Primary canalicular bile undergoes a process of fluidization and alkalinization along the biliary tract that is influenced by several factors including hormones, innervation/neuropeptides, and biliary constituents. The excretion of bicarbonate at both the canaliculi and the bile ducts is an important contributor to the generation of the so-called bile-salt independent flow. Bicarbonate is secreted from hepatocytes and cholangiocytes through parallel mechanisms which involve chloride efflux through activation of Cl- channels, and further bicarbonate secretion via AE2/SLC4A2-mediated Cl-/HCO3- exchange. Glucagon and secretin are two relevant hormones which seem to act very similarly in their target cells （hepatocytes for the former and cholangiocytes for the latter）. These hormones interact with their specific G protein-coupled receptors, causing increases in intracellular levels of cAMP and activation of cAMP-dependent Cl- and HCO3- secretory mechanisms. Both hepatocytes and cholangiocytes appear to have cAMP-responsive intracellular vesicles in which AE2/SLC4A2 colocalizes with cell specific Cl- channels （CFTR in cholangiocytes and not yet determined in hepatocytes） and aquaporins （AQP8 in hepatocytes and AQP1 in cholangiocytes）, cAMP-induced coordinated trafficking of these vesicles to either canalicular or cholangiocyte lumenal membranes and further exocytosis results in increased osmotic forces and passive movement of water with net bicarbonate-rich hydrocholeresis.

Episcleral brachytherapy as an effective alternative in vasoproliferative tumors

The aim of this study is to assess the results of episcleral brachytherapy as treatment of retinal vasoproliferative tumors(RVTs) in a referral intraocular Tumors Unit(iTU). A retrospective review of all patients diagnosed with vasoproliferative tumors of the retina and treated with episcleral brachytherapy in the iTU, University Hospital of Valladolid between 2009 and 2015 was done. Five patients accomplished the inclusion criteria. All of them presented associated exudation and secondary retinal detachments(RD). Four patients had received prior treatments. Decreased tumor size and exudation regression was found in all cases after treatment. Visual acuity remained stable or increased in all patients. No recurrences have been found after twelve-months follow up. The results of the present study suggest that episcleral brachytherapy is an efficient and safe option in the management of vasoproliferative tumors, especially when large tumor or extensive subretinal fluid is present. in these cases episcleral brachytherapy could be considered as a first line treatment.

Study on the structure and composition of aortic valve calcific deposits:Etiological aspects

The structures and chemical compositions of valve calcific deposits were investigated. The deposits was chosen arbitrarily and subjected to chemical analysis, observation with scanning microscope, semi-quantitative determination of Ca, Mg, Na, K, P and C elements by energy dispersive X-ray, X-ray diffraction and Fourier transform infrared spectroscopy carried out. These deposits were found to have non-uniform internal structures composed of layers of a structureless aspidinic inorganic material, substantial amounts of voluminous organic material and in a few samples small spheres were also present. Two groups of deposits with distinctly different chemical compositions were identified: one group with a low Ca/P molar ratio (1.59) and the other group with a high (1.82) Ca/P molar ratio. The deposits belonging to the group with a low Ca/P molar ratio contain higher concentration of magnesium and consist of increased amount of amorphous calcium phosphate. The deposits with a high Ca/P molar ratio contain low concentration of magnesium and consist predominantly of carbonated hydroxyapatite. The inorganic material was identified as a poorly crystalline carbonate hydroxyapatite containing molecular water of the average formula Ca9.1Mg0.4(Na,K)(PO4)5.8(CO3)0.3(OH)2. The actual chemical composition of the apatitic solid phase varies not only from deposit to deposit but also within the same deposit. The non-uniform internal structure of the deposits, the occasional presence of spherical particles and the variable point composition of the individual deposits indicate that their formation did not proceed under more or less constant conditions.

Caspase-3 activation as a bifurcation point between plasticity and cell death

Death-mediating proteases such as caspases and caspase-3 in particular, have been implicated in neurodegenerative processes, aging and Alzheimer＇s disease. However, emerging evidence suggests that in addition to their classical role in cell death, caspases play a key role in modulating synaptic function. it is remarkable that active caspases-3, which can trigger widespread damage and degeneration, aggregates in structures as delicate as synapses and persists in neurons without causing acute cell death. Here, we evaluate this dichotomy, and discuss the hypothesis that caspase-3 may be a bifurcation point in cellular signaling, able to orient the neuronal response to stress down either pathological/apoptotic pathways or towards physiological cellular remodeling. We propose that temporal, spatial and other regulators of caspase activity are key determinants of the ultimate effect of caspase-3 activation in neurons. This concept has implications for differential roles of caspase-3 activation across the lifespan. Specifically, we propose that limited caspase-3 activation is critical for synaptic function in the healthy adult brain while chronic activation is involved in degenerative processes in the aging brain.

The renin angiotensin system(RAS)appears to influence male fertility at multiple *** this work,we analyzed the relationship between the RAS and DNA *** male volunteers were divided into two groups(25 each):control(DNA fragmentation≤20%)and pathological(DNA fragmentation>20%)*** of five peptidases controlling RAS were measured fluorometrically:prolyl endopeptidase(which converts angiotensin[A]i and A ii to A 1–7),neutral endopeptidase(NEP/CD10:A i to A 1–7),aminopeptidase N(APN/CD13:A iii to A iV),aminopeptidase A(A ii to A iii)and aminopeptidase B(A iii to A iV).Angiotensin-converting enzyme(A i to A ii),APN/CD13 and NEP/CD10 were also assessed by semiquantitative cytometry and quantitative flow cytometry assays,as were the receptors of all RAS components:A ii receptor type 1(AT1R),A ii receptor type 2(AT2R),A iV receptor(AT4R or insulin-regulated aminopeptidase[iRAP]),(pro)renin receptor(PRR)and A 1–7 receptor or Mas receptor(MasR)None of the enzymes that regulate levels of RAS components,except for APN/CD13(decrease in fragmented cells),showed significant differences between both *** of RAS receptors revealed no significant differences in immunolabeling patterns between normozoospermic and fragmented *** of AT1R(94.3%normozoospermic vs 84.1%fragmented),AT4R(96.2%vs 95.3%)and MasR(97.4%vs 87.2%)was similar between the ***2R(87.4%normozoospermic vs 63.1%fragmented)and PRR(96.4%vs 48.2%)were higher in non-fragmented *** findings suggest that fragmented DNA spermatozoa have a lower capacity to respond to bioactive RAS peptides.

Challenges Analyzing RNA-Seq Gene Expression Data

The analysis of messenger Ribonucleic acid obtained through sequencing techniques (RNA-se- quencing) data is very challenging. Once technical difficulties have been sorted, an important choice has to be made during pre-processing: Two different paths can be chosen: Transform RNA- sequencing count data to a continuous variable or continue to work with count data. For each data type, analysis tools have been developed and seem appropriate at first sight, but a deeper analysis of data distribution and structure, are a discussion worth. in this review, open questions regarding RNA-sequencing data nature are discussed and highlighted, indicating important future research topics in statistics that should be addressed for a better analysis of already available and new appearing gene expression data. Moreover, a comparative analysis of RNAseq count and transformed data is presented. This comparison indicates that transforming RNA-seq count data seems appropriate, at least for differential expression detection.

Arundo donax L.（Arundo D.） Pennisetum purpureum（Elephant G.）,and Panicum virgatum（Switchgrass） are three perennial C4 crops which have been received particular attention during the last years as bioenergy *** experiment located in Uruguay,were established in 2007 to determine biomass production and nutrient balance on these three energy crops.A combination of four treatments of N and P（0 or 100 kg ha-1） were applied *** *** significantly more biomass than Arundo *** Switchgrass in all *** was response to N（100 kg ha-1） in 2009,especially in Elephant G.（＞42% compared to without N）.On the other hand,there was not response to P applications for any C4 crops for any *** of major nutrient flows have shown that the N and P balance were positive with N and P applications;however,the P,N and K balance without N-P applications were *** removal was greater under Elephant G.（-1227 kg ha-1） largely due to greater K concentrations and biomass *** the component of biomass that will be removed and have to be replaced and changes in soil nutrients,should be very important for the long-term success of the bioenergy system.
