Evaluation of remote radiologist-interpreted point-of-care ultrasound for suspected dengue patients in a primary health care facility in Colombia

impairment of pre-mRNA splicing in liver disease: Mechanisms and consequences

Pre-mRNA splicing is an essential step in the process of gene expression in eukaryotes and consists of the removal ofintrons and the linking of exons to generate mature mRNAs. This is a highly regulated mechanism that allows the alternative usage of exons, the retention ofintronic sequences and the generation of exonic sequences of variable length. Most human genes undergo splicing events, and disruptions of this process have been associated with a variety of diseases, including cancer. Hepatocellular carcinoma (HCC) is a molecularly heterogeneous type of tumor that usually develops in a cirrhotic liver. Alterations in pre-mRNA splicing of some genes have been observed in liver cancer, and although still scarce, the available data suggest that splicing defects may have a role in hepatocarcinogenesis. Here we briefly review the general mechanisms that regulatepre-mRNA splicing, and discuss some examples that illustrate how this process is impaired in liver tumorigenesis, and may contribute to HCC development. We believe that a more thorough examination of pre-mRNA splicing is still needed to accurately draw the molecular portrait of liver cancer. This will surely contribute to a better understanding of the disease and to the development of new effective therapies.

Post-translational modifications of prostaglandin-endoperoxide synthase 2 in colorectal cancer:An update

The biosynthesis of prostanoids is involved in both physiological and pathological processes. The expression of prostaglandin-endoperoxide synthase 2(PTGS2; also known as COX-2) has been traditionally associated to the onset of several pathologies, from inflammation to cardiovascular, gastrointestinal and oncologic events. For this reason, the search of selective PTGS2 inhibitors has been a focus for therapeutic interventions. in addition to the classic non-steroidal anti-inflammatory drugs, selective and specific PTGS2 inhibitors, termed coxibs, have been generated and widely used. PTGS2 activity is less restrictive in terms of substrate specificity than the homeostatic counterpart PTGS1, and it accounts for the elevated prostanoid synthesis that accompanies several pathologies. The main regulation of PTGS2 occurs at the transcription level. in addition to this, the stability of the mRNA is finely regulated through the interaction with several cytoplasmic elements, ranging from specificmicroR NAs to proteins that control mR NA degradation. Moreover, the protein has been recognized to be the substrate for several post-translational modifications that affect both the enzyme activity and the targeting for degradation via proteasomal and non-proteasomal mechanisms. Among these modifications, phosphorylation, glycosylation and covalent modifications by reactive lipidic intermediates and by free radicals associated to the proinflammatory condition appear to be the main changes. identification of these post-translational modifications is relevant to better understand the role of PTGS2 in several pathologies and to establish a correct analysis of the potential function of this protein in diseases progress. Finally, these modifications can be used as biomarkers to establish correlations with other parameters, including the immunomodulation dependent on molecular pathological epidemiology determinants, which may provide a better frame for potential therapeutic interventions.

Phylogeny,taxonomy and diversification events in the Caliciaceae

Bofedal response to climate variability, local management, and water extraction: A case study of Chucuyo, Northern Chile

Andean high-altitude wetlands are important ecosystems that serve a range of socialecological functions. in the Andes, bofedales, a specific type of peat-producing wetland, are essential for the sustainability of mountain ecosystems and indigenous pastoralist communities. The Chucuyo bofedal in northern Chile is affected by climatic variability and water extraction for agricultural uses via the Lauca canal. Herders in the local community also actively manage this wetland according to their traditional ecological knowledge to ensure permanent fodder for their animals. in this article, we analyze the annual behavior of the Chucuyo bofedal after the wet season. Based on precipitation data, extracted water flows, vegetation vigor from satellite images, and an ethnographic approach, we determined that the bofedal’s productivity was negatively correlated with the amount of precipitation and positively correlated with the surface area of the wetland. However, water extraction via the Lauca channel had no significant relationship with either surface area or vegetation vigor. We identify community practices and traditional ecological knowledge as key elements in the maintenance of these ecosystems. This situation is critical in the context of an aging population and the current pattern of migration out of the region to urban areas. The results provide substantial empirical evidence for future decision-making regarding the conservation of these ecosystems.

On the road towards the global analysis of human synapses

Synapses are essential units for the flow of information in the *** the last 70 years,synapses have been widely studied in multiple animal models including worms,fruit flies,and *** comparison,the study of human synapses has evolved significantly slower,mainly because of technical ***,three novel methods allowing the analysis of molecular,morphological,and functional properties of human synapses may expand our knowledge of the human ***,we briefly describe these methods,and evaluate how the information provided by each unique approach may contribute to the functional and anatomical analysis of the synaptic component of human brain *** particular,using tissue from cryopreserved human brains,synaptic plasticity can be studied in isolated synaptosomes by fluorescence analysis of single-synapse long-term potentiation（FASS-LTP）,and subpopulations of synapses can be thoroughly assessed in the ribbons of brain tissue by array tomography（AT）.Currently,it is also possible to quantify synaptic density in the living human brain by positron emission tomography（PET）,using a novel synaptic ***,data provided by FASS-LTP,AT,and PET may significantly contribute to the global understanding of synaptic structure and function in both healthy and diseased human brains,thus directly impacting translational research.

Combination of interleaved Single-input Multiple-output DC-DC Converters

This paper analyses,simulates and verifies an experimental prototype of a four-phase interleaved DC-DC *** is based on a SEPiC-Cuk *** developed prototype has been used in single-input multiple-output(SiMO)*** combined converter allows obtaining dual output voltages of the same value,from a single input DC voltage and with only a power *** interleaved DC-DC converters achieve a better dynamic response and low ripple,maintaining their *** converter is connected in parallel,thereby managing their losses by distributing them between more components,which facilitates the thermal management of the multiphase converter and allows handling high power values in small sizes with respect to solutions for a single *** control strategies were applied:synchronous operation mode(SOM)and interleaved operation mode(iOM).The simulation results allow the comparison of both operational modes,verifying that the iOM presents advantages with respect to the ripple at the input and output *** experimental prototype was designed for a distributed power architecture and bipolar DC microgrid(MG).
