Comparative molecular docking studies of lupeol and lupenone isolated from Pueraria lobata that inhibits BACE1: Probable remedies for Alzheimer's disease

Objective: To discover lead lupane triterpenoid's potential isolated from Pueraria lobata roots against b-site amyloid precursor protein cleaving enzyme 1(BACE1), which serve as a rate limiting step in amyloid beta(Aβ) production altering the course of Alzheimer's disease. In addition, enzyme kinetics study and molecular docking were conducted to establish the inhibition type and structure activity ***: A systematic study of 70% ethanolic P. lobata root extract was employed to identify its BACE1 inhibitory potential. Further, BACE1 inhibitory potential of two lupane terpenoids, yielded from ethanolic extract, was assessed. In order to determine their inhibition mode, Lineweaver–Burk plots and Michaelis–Menten model for BACE1 was performed. Auto Dock 4.2 program in addition determined the molecular interaction of BACE1 with isolated ***: Considering the inhibitory potential of 70% ethanolic extract of P. lobata against BACE1(IC_(50)= 80.35 mg/mL), lupeol and lupenone were subsequently isolated and exhibited notable or moderate BACE1 inhibitory activity with IC_(50) values of 5.12 and 62.98 mmol/L, respectively, as compared to the positive control quercetin(IC_(50)= 21.28 mmol/L). The enzyme kinetics study enabled us to identify both compounds as competitive inhibitors, where lupeol displayed a very potent inhibition against BACE1 with low inhibition constant(Ki) value of 1.43 mmol/L, signifying greater binding *** order to understand the binding mechanism and structure–activity relationship of two triterpene-based BACE1 inhibitors, we employed computer aided docking studies which evidently revealed that hydroxyl group of lupeol formed two hydrogen bonds with the ASP32(catalytic aspartic residue) and SER35 residues of BACE1 with the binding energy of(-8.2 kcal/mol), while the ketone group of lupenone did not form any hydrogen bonds with BACE1 giving evidence for less binding affinity. field attraction test of (7R,8S)-cis-7,8-epoxy-2- methyloctadec-17-ene (= 7R,8S-epo-2me-17ene-18Hy), a trace component of the sex pheromone gland of the Asian gypsy moth,(Lymantria dispar), traps that were baited with the trace component attracted more L. dispar than traps baited with (7S,8R)-cis-7,8-epoxy-2-methyloctadec-17-ene (= 7S,8R-epo-2me-17ene-18Hy) and unbaited traps. However, traps baited with 7R,8S-epo-2me-17ene-18Hy were less attractive than traps baited with (+)-disparlure [=(7R,8S)-cis-7,8-epoxy-2-methyloctadecane], the sex pheromone of L. dispar. Combination tests with (?)-disparlure, 7R,8S-epo-2me-17ene-18Hy, and 7S,8R-epo-2me- 17ene-18Hy revealed that 7R,8S-epo-2me-17ene-18Hy acted synergistically with (?)-disparlure.

Green Nanocomposites Made With Polyvinyl Alcohol and Cellulose Nanofibers Isolated From Recycled Paper

This paper reports green nanocomposites made by blending polyvinyl alcohol(PVA)with cellulose nanofiber(CNF)isolated from recycled deinked copy/printing paper(DIP).The reinforcement effect of DIPCNF in the nanocomposites is compared with other CNFs isolated from native cotton and hardwood by means of *** prepared PVA-CNF nanocomposites are characterized in terms of morphology,chemical interaction,structural,thermal and mechanical properties.X-ray diffraction and Fourier transform infrared spectroscopy confirm the reinforcing ability of cellulose nanofibers into *** blending CNFs into PVA matrix,the thermal stability of the nanocomposites is improved and DIPCNF shows similar enhancement effect with COCNF and *** prepared nanocomposites exhibit 50%Young’s modulus improvement by adding 6%of CNF and DIPCNF exhibits similar enhancement of the mechanical properties to COCNF and HWCNF in the *** indicates that the use of DIPCNF is beneficial for environment protection,resource retaining and save energy in comparison with COCNF and HWCNF.

AIM To investigate the effects of a water extract of Hwangryunhaedok-tang(HHTE) on the pacemaker potentials of mouse interstitial cells of Cajal(ICCs).METHODS We dissociated ICCs from small intestines and cultured. ICCs were immunologically identified using an antic-kit antibody. We used the whole-cell patch-clamp configuration to record the pacemaker potentials generated by cultured ICCs under the current clamp mode(I = 0). All experiments were performed at 30 ℃-32 ℃RESULTS HHTE dose-dependently depolarized ICC pacemaker potentials. Pretreatment with a 5-HT_3 receptor anta-gonist(Y25130) or a 5-HT_4 receptor antagonist(RS39604) blocked HHTE-induced pacemaker potential depolarizations, whereas pretreatment with a 5-HT7 receptor antagonist(SB269970) did not. Intracellular GDPβS inhibited HHTE-induced pacemaker potential depolarization and pretreatment with a Ca^(2+)-free solution or thapsigargin abolished the pacemaker potentials. In the presence of a Ca^(2+)-free solution or thapsigargin, HHTE did not depolarize ICC pacemaker potentials. In addition, HHTE-induced pacemaker potential depolarization was unaffected by a PKC inhibitor(calphostin C) or a Rho kinase inhibitor(Y27632). Of the four ingredients of HHT, Coptidis Rhizoma and Gardeniae Fructus more effectively inhibited pacemaker potential *** These results suggest that HHTE dose-dependently depolarizes ICC pacemaker potentials through 5-HT_3 and 5-HT_4 receptors via external and internal Ca^(2+) regulation and via G protein-, PKC-and Rho kinase-independent pathways.

Predicting Bitcoin Trends Through Machine Learning Using Sentiment Analysis with Technical Indicators

Predicting Bitcoin price trends is necessary because they represent the overall trend of the cryptocurrency *** the history of the Bitcoin market is short and price volatility is high,studies have been conducted on the factors affecting changes in Bitcoin *** have been conducted to predict Bitcoin prices using Twitter ***,the amount of data was limited,and prices were predicted for only a short period(less than two years).In this study,data from Reddit and LexisNexis,covering a period of more than four years,were *** data were utilized to estimate and compare the performance of the six machine learning techniques by adding technical and sentiment indicators to the price data along with the volume of *** accuracy of 90.57%and an area under the receiver operating characteristic curve value(AUC)of 97.48%were obtained using the extreme gradient boosting(XGBoost).It was shown that the use of both sentiment index using valence aware dictionary and sentiment reasoner(VADER)and 11 technical indicators utilizing moving average,relative strength index(RSI),stochastic oscillators in predicting Bitcoin price trends can produce significant ***,the input features used in the paper can be applied on Bitcoin price ***,this approach allows investors to make better decisions regarding Bitcoin-related investments.

S-doped TiO2(S-TiO2) films were immobilized on flexible low-cost aluminum sheets(S-TiO2-AS) using a sol-gel dipping process and low post-processing temperatures. The photocatalytic degradation of toxic organic vapors using the prepared films was evaluated using a continuous-flow glass tube under visible light exposure. The surface properties of the S-TiO2-AS and TiO2-AS films were examined by scanning electron microscopy, energy-dispersive X-ray spectroscopy, X-ray diffraction, and ultraviolet-visible spectroscopy. The photolysis of benzene, toluene, ethyl benzene, and xylene(BTEX) did not occur on the bare AS. In contrast, the photocatalytic degradation efficiencies of the target pollutants using S-TiO2-AS were higher than those obtained using reference TiO2-AS photocatalyst. In particular, the average photocatalytic degradation efficiencies of BTEX using S-TiO2-0.8-AS(S/Ti ratio = 0.8) over a 3-h process were 34%, 78%, 91%, and 94%, respectively, whereas those of TiO2-AS were 2%, 11%, 21%, and 36%, respectively. The photocatalytic decompo-sition efficiencies of BTEX under visible irradiation using S-TiO2-AS increased with increasing S/Ti ratios from 0.2 to 0.8, but decreased when the ratio further increased to 1.6. Thus, S-TiO2-AS can be prepared using optimal S/Ti ratios. The degradation of BTEX over S-TiO2-AS depended on the air flow rates and initial concentrations of the target chemical. Overall, under optimal conditions, S-TiO2-AS can be effectively applied for the purification of toxic organic vapors.

Porous titanium(Ti)scaffolds have been extensively utilized as bone substitute scaffolds due to their superior biocompatibility and excellent mechanical ***,naturally formed TiO2 on the surface limits fast *** biomolecules have been widely utilized to overcome this issue;however,homogeneous porous Ti scaffolds could not simultaneously deliver multiple biomolecules that have different release *** this study,functionally graded porous Ti scaffolds(FGPTs)with dense inner and porous outer parts were fabricated using a two-body combination and densification *** with growth factor(BMP-2)and antibiotics(TCH)exhibited suitable mechanical properties as bone substituting material and presented good structural *** release of BMP-2 was considerably prolonged,whereas the release of TCH was comparable to that of homogenous porous titanium scaffolds(control group).The osteogenic differentiation obtained using FGPTs was maintained due to the prolonged release of *** antimicrobial properties of these scaffolds were verified using *** in terms of prior release *** addition,various candidates for graded porous Ti scaffolds with altered pore characteristics were presented.

Anti-MMP-2 and MMP-9 activity of Salsola komarovii Iljin extract and its solvent fractions

Objective: To investigate matrix metalloproteinases(MMP)-2 and MMP-9 inhibitory effect of Salsola komarovii Iljin, an edible halophyte with health beneficial ***: Salsola komarovii crude extracts(SKI), and solvent(n-hexane, 85% aq. MeOH, n-BuOH, and H2O) fractionated extracts of SKI were prepared. Gelatin zymography was carried out to observe MMP enzymatic activity. The release of the MMP enzymes was measured by enzyme-linked immunosorbent assay. Expression of MMPs in m RNA and protein level were investigated by polymerase chain reaction analysis and immunoblotting, ***: SKI and SKI fractions inhibited active MMP-2 and MMP-9 amount in the treated cell culture medium. Also, SKI suppressed the release of MMP-2 and MMP-9 from stimulated HT1080 human fibrosarcoma cells. Furthermore, SKI suppressed the m RNA and protein expression of MMP-2 and MMP-9. SKI fractions showed parallel effects except for H2O fraction which did not yield any significant MMP inhibitory effect. Among fractions, 85% aq. Me OH was the most active fraction to inhibit both the enzymatic effect and expression of MMP-2 and ***: SKI may contain potential MMP release inhibitory compounds. Salsola komarovii is a promising source of compounds against MMP and could be utilized in the development of antitumor agents.
