Exposure to persistent organic pollutants(POPs)has been reported as a potential risk factor for diabetes in ***,effect modifications by sex and age,especially among Asian populations,have seldom been *** the present study,we assessed associations of major POPs exposure including organochlorine pesticides(OCPs),polychlorinated biphenyls(PCBs),and polybrominated diphenyl ethers(PBDEs),with diabetes in Korean adults(n=1,295),a subset of the Korean National Environmental Health Survey(KoNEHS)Cycle 3(2015−2017).In the adult population,serum levels of dichlorodiphenyltrichloroethanes(DDTs)and PCB167 showed an increased odds ratio(OR)for *** stratified by gender,associations became stronger and more consistent in women:In women,serum levels of OCPs including p,p′-DDT,p,p′-dichlorodiphenyldichloroethylene(p,p′-DDE),andΣOCPs and mono-ortho PCBs,such as PCB118 and PCB157,were significantly associated with *** the male participants,however,no significant positive associations were *** stratified by a cutoff age of 50 years,most studied POPs except PBDEs,i.e.,p,p′-DDT,p,p′-DDE,PCB118,PCB157,PCB167,PCB138,and PCB153,showed significant associations with diabetes,in the older *** the younger group,however,no measured POPs showed a significant positive *** of endogenous sex hormones or interaction of POPs on aryl hydrocarbon receptors may partly explain the observed differences by gender and age,although further experimental confirmations should *** findings show that POPs exposure at the current levels is associated with diabetes among general Korean adults,and such associations could be modified by gender and age.

Elicitation of strong immune responses by a DNA vaccine expressing a secreted form of hepatitis C virus envelope protein E2 in murine and porcine animal models

AIM： To characterize the immunogenicity of a hepatitis C virus （HCV） E2 DNA vaccine alone or with a protein vaccine boost in murine and porcine animal models. METHODS： A DNA vaccine expressing a secreted form of HCV E2 protein was constructed and used to vaccinate mice and piglets with or without boosting with a recombinant E2 protein vaccine formulated with CpG ODN and 10% Emulsigen. The immunogenicity of HCV E2 vaccines was analyzed by EliSA for antibody responses, MTT assay for lymphocyte proliferation, EliSPOT for the number of interferon-γ secreting cells, and cytotoxic T lymphocyte assays. RESULTS： Intradermal injection of E2 DNA vaccine induced strong Th1-1ike immune responses in mice. In piglets, E2 DNA vaccine elicited moderate and more balanced immune responses. A DNA vaccine prime and protein boost vaccination strategy induced significantly higher E2-specific antibody levels and shifted the immune response towards Th2-1ike ones in piglets. CONCLUSION： A DNA vaccine expressing a secreted form of HCV E2 protein elicited E2-specific immune responses in mice and piglets. Recombinant E2 protein vaccination following DNA immunization significantly increased the antibody response in piglets. These HCV E2 vaccines may represent promising hepatitis C vaccine candidates for further investigations.

Clinical algorithm for preventing missed diagnoses of occult cervical spine instability after acute trauma: A case report

BACKGROUND Missed or delayed diagnosis of cervical spine instability after acute trauma can have catastrophic consequences for the patient,resulting in severe neurological ***,however,there is no consensus on the optimal strategy for diagnosing occult cervical spine ***,we present a case of occult cervical spine instability and provide a clinical algorithm to aid physicians in diagnosing occult instability of the cervical *** SUMMARY A 57-year-old man presented with cervical spine pain and inability to stand following a serious fall from a height of 2 *** obvious vertebral fracture or dislocation was found at the time on standard lateral X-ray,computed tomography,and magnetic resonance imaging(MRI).Subsequently,the initial surgical plan was unilateral open-door laminoplasty(C3-7)with alternative levels of centerpiece mini-plate fixation(C3,5,and 7).However,the intraoperative C-arm fluoroscopic X-rays revealed significantly increased intervertebral space at C5-6,indicating instability at this level that was previously unrecognized on preoperative *** finally performed lateral mass fixation and fusion at the C5-6 *** back at the preoperative images,we found that the preoperative T2 MRI showed non-obvious high signal intensity at the C5-6 intervertebral disc and posterior interspinous *** MRI of cervical spine trauma patients should be carefully reviewed to detect disco-ligamentous injury,which will lead to further cervical spine *** patients with highly suspected cervical spine instability indicated on MRI,lateral X-ray under traction or after anesthesia and muscle relaxation needs to be performed to avoid missed diagnoses of occult cervical instability.

CXCR1 is a G-protein coupled receptor, transducing signals from chemokines, in particular the interleukin-8 （1L8） molecules. This study combines homology modeling and molecular dynamics simulation methods to study the structure of CXCRI-IL8 complex. By using CXCR4-vMIP-II crystallography structure as the homologous template, CXCRI-IL8 complex structure was constructed, and then refined using all-atom molecular dynamics simulations. Through extensive simulations, CXCRI-IL8 binding poses were investigated in detail. Furthermore, the role of the N-terminal of CXCR1 receptor was studied by comparing four complex models differing in the N-terminal sequences. The results indicate that the receptor N-terminal affects the binding of IL8 significantly. With a shorter N-terminal domain, the binding of IL8 to CXCR1 becomes unstable. The homology modeling and simulations also reveal the key receptor-ligand residues involved in the electrostatic interactions known to be vital for complex formation.

Intravoxel incoherent motion diffusion-weighted imaging for monitoring chemotherapeutic efficacy in gastric cancer

AIM: To assess intravoxel incoherent motion diffusion-weighted imaging (IVIM-DWI) for monitoring early efficacy of chemotherapy in a human gastric cancer mouse ***: IVIM-DWI was performed with 12 b-values (0-800 s/mm2) in 25 human gastric cancer-bearing nude mice at baseline (day 0), and then they were randomly divided into control and 1-, 3-, 5- and 7-d treatment groups (n = 5 per group). The control group underwent longitudinal MRI scans at days 1, 3, 5 and 7, and the treatment groups underwent subsequent MRI scans after a specified 5-fluorouracil/calcium folinate treatment. Together with tumor volumes (TV), the apparent diffusion coefficient (ADC) and IVIM parameters [true water molecular diffusion coefficient (D), perfusion fraction (f) and pseudo-related diffusion coefficient (D*)] were measured. The differences in those parameters from baseline to each measurement (ΔTV%, ΔADC%, ΔD%, Δf% and ΔD*%) were calculated. After image acquisition, tumor necrosis, microvessel density (MVD) and cellular apoptosis were evaluated by hematoxylin-eosin (HE), CD31 and terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL) staining respectively, to confirm the imaging findings. Mann-Whitney test and Spearman’s correlation coefficient analysis were ***: The observed relative volume increase (ΔTV%) in the treatment group were significantly smaller than those in the control group at day 5 (ΔTVtreatment% = 19.63% ± 3.01% and ΔTVcontrol% = 83.60% ± 14.87%, P = 0.008) and day 7 (ΔTVtreatment% = 29.07% ± 10.01% and ΔTVcontrol% = 177.06% ± 63.00%, P = 0.008). It is required to understand the skin color in developing makeup cosmetics. In this study, we aimed to develop the customized BB cream color for Chinese women by understanding their skin color and color preference. It is known that Chinese women have light yellow color, and yellowness and darkening induced by age are evident for the Chinese women. Based on previous studies, we developed two colors and evaluated color preference. The result was that Chinese women preferred pinkish color to yellowish color. We applied this result to BB cream and proposed Chinese customized color. This study demonstrates that the sensuous and rational approach for satisfying color needs and expectations is beneficial in developing makeup cosmetics.

In this study, we tried to verify the neuroprotective effect of Chrysanthemum indicum linne（CIL） extract, which has been used as a botanical drug in East Asia, against ischemic damage and to explore the underlying mechanism involving the anti-inflammatory approach. A gerbil was given CIL extract for 7 consecutive days followed by bilateral carotid artery occlusion to make a cerebral ischemia/reperfusion model. Then, we found that CIL extracts protected pyramidal neurons in the hippocampal CA1 region（CA1） from ischemic damage using neuronal nucleus immunohistochemistry and Fluoro-Jade B histofluorescence. Accordingly, interleukin-13 immunoreactivities in the CA1 pyramidal neurons of CIL-pretreated animals were maintained or increased after cerebral ischemia/reperfusion. These findings indicate that the pre-treatment of CIL can attenuate neuronal damage/death in the brain after cerebral ischemia/reperfusion via an anti-inflammatory approach.

The utility of a next generation biosynthetic bacterial 6-phytase(PhyG)in restoring bone ash,bone phosphorus(P)content and performance in piglets depleted in P was evaluated.A total of 9 treatments were tested as *** 1,a negative control(NC)diet;treatments 2,3,4,NC supplemented with 250,500 or 1,000 FTU/kg of PhyG;treatments 5,6,NC supplemented with 500 or 1,000 FTU/kg of a commercial Buttiauxella sp phytase(PhyB);treatments 7,8,9,NC supplemented with monocalcium phosphate(MCP)to provide 0.7,1.4 and 1.8 g/kg digestible P,equating to a digestible P content of 1.8,2.5 and 2.9 g/*** latter constituting the positive control(PC)diet with adequate P and calcium(Ca).The NC was formulated without inorganic P(1.1 g digestible P/kg)and reduced in Ca(5.0 g/kg).Additional limestone was added to treatments 7 to 9 to maintain Ca-to-P ratio between 1.2 and 1.3.A total of 162 crossed Pietrain x(Large White×Landrace)21-d-old piglets(50%males and 50%females)were fed adaptation diets until 42 d old and then assigned to pens with 2 pigs/pen and 9 pens/treatment in a completely randomized block *** were fed mash diets based on corn and soybean meal ad libitum for 28 *** the end of the study,one piglet perpen was euthanized and the right feet collected for determination of bone strength,bone ash and mineral *** with the PC,the NC group had reduced average daily gain(ADG)and increased feed conversion ratio(FCR)during all growth phases and overall,and at d 28(70 d old)NC pigs had bones with reduced ash,Ca and P content(Plinearly or quadratically improved bone ash,ADG and FCR(P500 FTU/kg,both PhyG and PhyB maintained ADG and FCR equivalent to *** regression analysis was done to compare the measured response parameters to increasing digestible P from *** on this analysis it was shown that PhyG and PhyB at 1,000 FTU/kg could replace 1,83 and 1.66 g/kg dige
