Acquisition of pluripotency in the chick embryo occurs during intrauterine embryonic development via a unique transcriptional network

Background: Acquisition of pluripotency by transcriptional regulatory factors is an initial developmental event that is required for regulation of cell fate and lineage specification during early embryonic development. The evolutionarily conserved core transcriptional factors regulating the pluripotency network in fishes, amphibians, and mammals have been elucidated. There are also species-specific maternally inherited transcriptional factors and their intricate transcriptional networks important in the acquisition of pluripotency. In avian species, however, the core transcriptional network that governs the acquisition of pluripotency during early embryonic development is not well ***: We found that chicken nAnOG(c nAnOG) was expressed in the stages between the pre-ovulatory follicle and oocyte and was continuously detected in Eyal-Giladi and Kochav stage I(EGK.I) to X. However, c POUV was not expressed during fol iculogenesis, but began to be detectable between EGK.V and VI. Unexpectedly, c SOX2 could not be detected during fol iculogenesis and intrauterine embryonic development. Instead of c SOX2, c SOX3 was maternally inherited and continuously expressed during chicken intrauterine development. In addition, we found that the pluripotency-related genes such as c EnS-1, c KIT, c LIn28 A, c MYC, c PRDM14, and c SALL4 began to be dramatical y upregulated between *** and ***: These results suggest that chickens have a unique pluripotent circuitry since maternally inherited c nAnOG and c SOX3 may play an important role in the initial acquisition of pluripotency. Moreover, the acquisition of pluripotency in chicken embryos occurs at around *** to VI I.

White organic light-emitting diodes were fabricated with two separated emissive layers using a blue fluorescent dopant and a red phosphorescent *** L-V characteristics of the device A and B showed maximum luminance of 26 100 and 21 100 cd/m at 14V and the maximum luminous efficiency of the devices showed 11.3 and 9.7 cd/A,respectively. The CIE coordinates of device A and B showed (x=0.27,y=0.35)and (x=0.33,y=0.33) at 10 *** device used BAlq as a host demonstrated proper white CIE coordinates and also showed white emission at operating voltages (4V-14V) without shifting of CIE coordinates.

Changes in spatial variations of sap flow in Korean pine trees due to environmental factors and their effects on estimates of stand transpiration

It is difficult to scale up measurements of the sap flux density(J_S) for the characterization of tree or stand transpiration(E) due to spatial variations in J_S and their temporal *** assess spatial variations in the sap flux density of Korean pine(Pinus koraiensis) and their effects on E estimates,we measured the J_S using Granier-type *** trees,the J_S decreased exponentially with the radial depth,and the J_S of the east aspects were higher than those of the west *** trees,there was a positive relationship between J_S and the tree diameter at breast height,and this positive relationship became stronger as the transpiration demand *** spatial variations that caused large errors in E estimates(i.e.,up to 110.8 % when radial variation was ignored) had varied systematically with environmental factors systematic characteristics in relation to environmental ***,changes in these variations did not generate substantial errors in the E *** our study periods,the differences in the daily E(E_D) calculated by ignoring radial,azimuthal and tree-to-tree variations and the measured E_D were fairly constant,especially when the daily vapor pressure deficit(D_D)was higher than 0.6 k *** results imply that the effect of spatial variations changes on sap flow can be a minor source of error compared with spatial variations(radial,azimuthal and tree-to-tree variations) when considering E estimates.

Sex disparity in viral load, inflammation and liver damage in transgenic mice carrying full hepatitis B virus genome with the W4P mutation in the preS1 region

AIM To study sex disparity in susceptibility to hepatocellular carcinoma(HCC), we created a transgenic mouse model that expressed the full hepatitis B virus(HBV) genome with the W4P *** Transgenic mice were generated by transferring the p hy92-1.1 x-HBV-full genome plasmid(genotype A2) into C57 Bl/6 n mice. We compared serum levels of hepatitis B surface antigen(HBs Ag), interleukin(IL)-6, and the liver enzymes alanine aminotransferase(ALT) and aspartate transaminase(AST), as well as liver histopathological features in male and female transgenic(W4PTG) mice and in nontransgenic littermates of 10 mo of age. RESULTS W4PTG males exhibited more pronounced hepatomegaly, significantly increased granule generation in liver tissue, elevated HBs Ag expression in the liver and serum, and higher serum ALT and IL-6 levels compared to W4PTG females or littermate control groups. COnCLUSIOn Together, our data indicate that the W4 P mutation in pre S1 may contribute to sex disparity in susceptibility to HCC by causing increased HBV virion replication and enhanced IL-6-mediated inflammation in male individuals. Additionally, our transgenic mouse model that expresses full HBV genome with the W4 P mutation in pre S1 could be effectively used for the studies of the progression of liver diseases, including HCC.

In this study, we investigated the effects of streptozotocin-induced type 1 diabetes on antioxi- dant-like protein-1 immunoreactivity, protein carbonyl levels, and malondialdehyde formation, a marker for lipid peroxidation, in the hippocampus. For this study, streptozotocin （75 mg/kg） was intraperitoneally injected into adult rats to induce type 1 diabetes. The three experimental pa- rameters were determined at 2, 3, 4 weeks after streptozotocin treatment. Fasting blood glucose levels significantly increased by 20.7-21.9 mM after streptozotocin treatment. The number of antioxidant-like protein-1 immunoreactive neurons significantly decreased in the hippocampal CA1 region, but not the dentate gyrus, 3 weeks after streptozotocin treatment compared to the control group. Malondialdehyde and protein carbonyl levels, which are modified by oxidative stress, significantly increased with a peak at 3 weeks after malondialdehyde treatment, and then decreased 4 weeks after malondialdehyde treatment. These results suggest that neurons in the hippocampal CA1 region, but not the dentate gyrus, are susceptible to oxidative stress 3 weeks after malondialdehyde treatment.

Objective:To investigate the effects of an ethanol extract of Kalopanax septemlobus(Thunb.)***(EEKS) on cell proliferation in human hepatocellular carcinoma cells and its mechanisms of ***:Cells were treated with EEKS and subsequently analyzed for cell proliferation and flow cytometry *** of cell cycle regulators were determined by reverse transcriptase polymerase chain reaction analysis and Western blotting,and activation of eyclin-associaled kinases studied using kinase ***:The EEKS suppressed cell proliferation in both HepG2 and Hep3 B cells,but showed a more sensitive anli-proliferative activity in HepG2 *** cytometry analysis revealed an association between the growth inhibitory effect of EEKS and with G_1 phase cell cycle arrest in HepG2 cells,along with the dephosphorylation of retinoblastoma protein(pRB) and enhanced binding of pRB with the E2 F transcription factor family *** with EEKS also increased the expression of cyclin-dependent kinase(CDK) inhibitors,such as p21WAF1/CIP1 and *** any noticeable changes in G_1 cyclins and CDKs(except for a slight decrease in CDK4).Treatment of HepG2 cells with EEKS also increased the binding of p21 and p27 with CDK4 and *** was paralleled by a marked decrease in the cyclin D- and cyclin E-associated kinase ***:Overall,our findings suggest that EEKS may be an effective treatment for liver cancer through suppression of cancer cell proliferation via G_1,cell cycle arrest Further studies arc required to identify the active compounds in EEKS.

Zircon deposit and rare earth element enriched metasomatic alkaline rocks at Kyemyeongsan Formation, Chungju, Korea: Paleozoic magmatism and Zr-REE-nb mineralization

The zircon ore deposit in metasediments and peralkaline granite of the Kyemyeongsan Formation is located in southwest of Chungju city, Korea. The deposit, closely associated with REE and nb,is composed of metasomatic alkaline rocks and rare metal alkali granite, and was formed in late Carboniferous (340～343Ma). Zircon occurs in different paragenetic sequence: (1) earlier rare metal alkali granite, (2) later metasomatic zircon ore. The metasomatic zone contains abundant microcline, albite and quartz with minor biotite, magnetite, hornblende, allanite and zircon. The alkali granites have high silica (72.13～74.52wt.% as SiO 2), and total iron content (5.95～6.89%), and are characterized by low Al 2O 3 content (7.12～9.74%). They also show variable K 2O content (3.60～6.98%), and high ratios of K 2O/na 2O. The REE patterns of rare metal alkali granite are similar to those of felsic volcanics from rifts, or back arc basins in, or near continental crust. Zircon ores are characterized by high iron content and low Al 2O 3, SiO 2, and K 2O content and have unusually high total REE content (0.18～2.33%). REE patterns show relatively flat to somewhat heavy REE (HREE) depleted characteristics (Ce/Yb=0.39～5.17) with large Eu negative anomaly (Eu/Eu *=0.16～0.29). Laser ablation microprobe inductively coupled plasma mass spectrometer (LAM ICP MS) analyses has been carried on zircon. The REE patterns of mineral zircons are almost the same to those of zircon ores and rare metal alkali granites, which may reflect the inability of zircons to effectively fractionated REE at formation of origin. The Sm nd isochron age of the zircon ore and rare metal alkali granite are 330Ma, and 331Ma, respectively with ε nd(t) being range from -2.00 to -1.84. This data suggest that the ore forming material came from the mantle. Alkali granite has suffered extensive post magmatic metasomatism of a high temperature to produce zircon ores. The intense Cenozoic volcanism of Mt. Changbai provides a natural laboratory for investigating the characteristics of volcanism and the dynamical evolution of the northeast Asian continental margin. Mt. Changbai volcanoes predominantly consist of Wangtian’e volcano in China, Tianchi volcano spanning China and DPR Korea, and namphothe volcano in DPR Korea. Geochronology data and historical records of volcanism indicate that the three eruption centers were formed in the following sequence: Wangtian’e volcano to namphothe and Tianchi volcano, advancing temporally and spatially from southwest to northeast. The three eruption centers of *** volcano are located close together, have similar magma evolution trends, bimodal volcanic rock distribution, and an enriched mantle source, etc. Although the Cenozoic volcanism in Mt. Changbai is thought to be somewhat related to the subduction of the Western Pacific Plate, the regularity of volcanic activity and petrography characteristics have continental rift affinity. We therefore conclude that the occurrence of synchronous and similar volcanic activity on both sides of the Japan Sea(i.e., the Japan Arc and northeast China) likely respond to the rift expansion and the back-arc spreading of Japan Sea. From many perspectives, Mt. Changbai volcano is a giant active volcano with hidden potentially eruptive risks.
