The potential benefits of generating and using transgenic cattle range from improvements in agriculture to the production of large quantities of pharmaceutically relevant proteins. Previous studies have attempted to produce transgenic cattle and other livestock by pronuclear injection and somatic cell nuclear transfer, but these approaches have been largely ineffective; however, a third approach, lentivirus-mediated transgenesis, has successfully produced transgenic livestock. In this study, we generated transgenic （TG） Korean native cattle using perivitelline space injection of viral vectors, which expressed enhanced green fluorescent protein （EGFP） systemically. Two different types of lentiviral vectors derived from feline immunodeficiency virus （FIV） and human immunodeficiency virus （HIV） carrying EGFP were injected into the perivitelline space of MII oocytes. EGFP expression at 8-cell stage was significantly higher in the FIV group compared to the HIV group （47.5% ± 2.2% v.s. 22.9% 4± 2.9%）. Eight-cell embryos that expressed EGFP were cultured into blastocysts and then transferred into 40 heifers. Ten heifers were successfully impregnated and delivered 10 healthy calves. All of these calves expressed EGFP as detected by in vivo imaging, PCR and Southern blotting. In addition, we established an EGFP-expressing cell line from TG calves, which was followed by nuclear transfer （NT）. Recloned 8-cell embryos also expressed EGFP, and there were no differences in the rates of fusion, cleavage and development between cells derived from TG and non-TG calves, which were subsequently used for NT. These results illustrate that FIV-based lentiviruses are useful for the production of TG cattle. Moreover, our established EGFP cell line can be used for additional studies that involve induced pluripotent stem cells.

Abscisic acid （ABA） is a major phytohormone involved in important stress-related and developmental plant processes. Membrane-delimited ABA signal transduction plays an important role in early ABA signaling, but the molecular mechanisms connecting core signaling components to the plasma membrane remain un- clear. Plants have evolved a large number of receptor-like kinases （RLKs） to modulate diverse biological processes by perceiving extracellular stimuli and activating downstream signaling responses. In this study, a putative leucine-rich repeat-RLK gene named RECEPTOR DEAD KINASE1 （AtRDK1） was identified and characterized in Arabidopsis thaliana. RDK1 promoter-GUS analysis revealed that RDK1 is expressed ubiq- uitously in the various tissues in Arabidopsis, and its expression is mainly induced by ABA. In the presence of ABA, RDKl-deficient rdkl-1 and rdkl-2 lines showed significant resistance in cotyledon greening and root growth, whereas RDKl-overexpressing lines showed enhanced sensitivity. Consistently, the expres- sion of ABA-responsive genes was significantly downregulated in rdkl mutant seedlings, which were also hypersensitive to drought stress with increased water loss. Interestingly, RDK1 was found to be an atypical kinase localized to the plasma membrane and did not require its kinase activity during ABA-mediated inhi- bition of seedling development. Accordingly, RDK1 interacted in the plasma membrane with type 2C protein phosphatase ABSClSIC ACID INSENSITIVE1 （ABI1）; this interaction was further enhanced by exogenous application of ABA, suggesting that RDKl-mediated recruitment of ABI1 onto the plasma membrane is important for ABA signaling. Taken together, these results reveal an important role for RDK1 in plant responses to abiotic stress conditions in an ABA-dependent manner.

Reactive oxygen species （ROS） attack guanine bases in DNA easily and form 8-hydroxydeoxyguanosine （8-OHdG）, which can bind to thymidine rather than cytosine, based on which, the level of 8-OHdG is gen- erally regarded as a biomarker of mutagenesis conse- quent to oxidative stress. For example, higher levels of 8-OHdG are noted in Helicobacter pylori-associated chronic atrophic gastritis as well as gastric cancer. However, we have found that exogenous 8-OHdG can paradoxically reduce ROS production, attenuate the nuclear factor-KB signaling pathway, and ameliorate the expression of proinflammatory mediators such as interleukin （IL）-I, IL-6, cyclo-oxygenase-2, and induc- ible nitric oxide synthase in addition to expression of nicotinamide adenine dinucleotide phosphate oxidase （NOX）-I, NOX organizer-1 and NOX activator-1 in vari- ous conditions of inflammation-based gastrointestinal （GI） diseases including gastritis, inflammatory bowel disease, pancreatitis, and even colitis-associated carci- nogenesis. Our recent finding that exogenous 8-OHdG was very effective in either inflammation-based or oxidative-stress-associated diseases of stress-related mucosal damage has inspired the hope that synthetic 8-OHdG can be a potential candidate for the treatment of inflammation-based GI diseases, as well as the pre- vention of inflammation-associated GI cancer. In this editorial review, the novel fact that exogenous 8-OHdG can be a functional molecule regulating oxidative- stress-induced gastritis through either antagonizing Rac-guanosine triphosphate binding or blocking the signals responsible for gastric inflammatory cascade is introduced.

Colorectal cancer is a common cancer and the fourth leading cause of death in Korea. The incidence and mortality of colorectal cancer varies according to risk factors, such as age, family history, genetic history, food habits, and physical activities. Some studies have focused on the association between vitamin D and colorectal cancer. Today, there is growing evidence that high vitamin D intake and a plasma level of 25(OH)D3 reduce the incidence of colorectal cancer by modifying cancer angiogenesis, cell apoptosis, differentiation, and proliferation. Taken together, these results suggest that vitamin D supplementation alone, or in combination with anti-cancer agents, might reduce the incidence of colorectal cancer. In this review, we discuss the function and mechanism of vitamin D including the effect of vitamin D on colorectal cancer.

A wavelength-swept laser is constructed using a free space external cavity configuration coupled with a fiber- based ring cavity at the 850 nm region. The external cavity filter employs a galvo-mirror scanner with a diffraction grating for wavelength selection. The filter is connected to a ring cavity through an optical circulator. The ring cavity contains a broadband semiconductor optical amplifier with a high optical output. The perfor- mance of this laser is demonstrated with broad bandwidths and narrow linewidths. The 3 dB linewidth and the bandwidth of this source are 0.05 nm （-20 GHz） and 48 nm, respectively. The maximum output power is 26 mW at 160 mA current.

Fog computing brings computational services near the network edge to meet the latency constraints of cyber-physical System(CPS)*** devices enable limited computational capacity and energy availability that hamper end user *** designed a novel performance measurement index to gauge a device’s resource *** examination addresses the offloading mechanism issues,where the end user(EU)offloads a part of its workload to a nearby edge server(ES).Sometimes,the ES further offloads the workload to another ES or cloud server to achieve reliable performance because of limited resources(such as storage and computation).The manuscript aims to reduce the service offloading rate by selecting a potential device or server to accomplish a low average latency and service completion time to meet the deadline constraints of sub-divided *** this regard,an adaptive online status predictive model design is significant for prognosticating the asset requirement of arrived services to make float ***,the development of a reinforcement learning-based flexible x-scheduling(RFXS)approach resolves the service offloading issues,where x=service/resource for producing the low latency and high performance of the *** approach to the theoretical bound and computational complexity is derived by formulating the system efficiency.A quadratic restraint mechanism is employed to formulate the service optimization issue according to a set ofmeasurements,as well as the behavioural association rate and adulation *** system managed an average 0.89%of the service offloading rate,with 39 ms of delay over complex scenarios(using three servers with a 50%service arrival rate).The simulation outcomes confirm that the proposed scheme attained a low offloading uncertainty,and is suitable for simulating heterogeneous CPS frameworks.

In this paper, the nonlinear state feedback controller has been developed to control the pressures of the oxygen and the hydrogen in the PEM（Proton Exchange Membrane） fuel cell system. Nonlinear model of the PEM fuel cell system was introduced to study the design problems of the state observer and model based controller. A cascade observer using the filtering technique was used to estimate the pressure derivatives of the cathode and the anode in the system. In order to estimate the pressures of the cathode and the anode, the sliding mode observer was designed by using these pressure derivatives. To estimate the oxygen pressure and the hydrogen pressure in the system, the nonlinear state observer was designed by using the cathode pressure estimates and the anode it. These results will be very useful to design the state feedback controller. The validity of the proposed observers and the controller has been investigated by using a Lyapunov＇s stability analysis strategy.
