Isolation and phylogenetic assignation of actinomycetes in the marine sediments from the Arctic Ocean

Actinomycetes in five marine sediments collected from the Arctic Ocean atdepths of 43 to 3 050 m were cultivated using a variety of media. A total of 61 actinomycetecolonies with substrate mycelia only were observed, and no colonies with aerial mycelia wereobserved under aerobic conditions at 15℃. From these colonies, 28 were selected to representdifferent morphological types. Denaturing gradient gel electrophoresis (DGGE) was used to check thepurity of isolates and select representatives for subsequent sequencing. Phylogentic analyses basedon nearly full-length 16S ribosomal RNA gene (rDNA) sequences indicated that the actinomycetesisolated were accommodated within genus Rhodococcus of family Nocardiaceae, genus Dietzia of familyDietziaceae, genera Janibacter and Terrabacter of family Instrasporangiaceae and genera Kocuria andA nhrobacter of family Micrococcaceae. One of the strains (P27-24) from the deep-sea sediment atdepth of 3 050 m was found to be identical in 16S rDNA sequence(1474/1474) with theradiation-resistant Kocuria rosea ATCC 187~T isolated from air. More than half of the isolatesshowed the similarities ranging from 99.5% to 99.9% in 16S rDNA sequence to dibenzofran-degrading,butyl 2-ethylhexanoate-hydrolysising and nitrile-metabolizing actinomycetes. All the strainsisolated were psychrotolerant bacteria and grew better on the media prepared with natural seawaterthan on the media prepared with deionized water. Three of them (Dietzia sp. P27-10, Rhodococcus sp.S11-3 and Rhodococcus sp.P11-5) had an obligate growth requirement for salt, confirming that thesestrains are indigenous marine actinomycetes.

Phylogenetic diversity and phenotypic characterization of cultivable bacterioplankton isolated from polar oceans

A set of 27 marine planktonic bacteria isolated from the polar regions was characterized by 16S rDNA sequencing and physiological and biochemical testing. More than half of these bacteria were positive for caseinase, gelatinase and 13-glucosidase, and could utilize glucose, maltose or malic acid as carbon source for cell growth. Twelve isolates expressed nitrate reduction activities. Except for one antarctic isolate BSwlO175 belonging to Actinobacteria phylum, these isolates were classified as γ-Proteobacteria, suggesting that γ-Proteobacteria dominated in cultivable marine bacterioplankton at both poles. Genus Pseudoalteromonas was the predominant group in the Chukchi Sea and the Bering Sea, and genus ShewaneUa dominated in cultivable bacterioplankton in the Prydz Bay. With sequence similarities above 97%, genus Psychrobacter was found at both poles. These 27 isolates were psychrotolerant, and significant 16S rDNA sequence similarities were found not only between arctic and antarctic marine bacteria （〉 99% ）, but also between polar marine bacteria and bacteria from other aquatic environments （ ≥ 98.8% ）, including temperate ocean, deep sea, pond and lake, suggesting that in the polar oceans less temperature-sensitive bacteria may be cosmopolitan and have a bipolar, even global, distribution at the species level.

CuSO4/TiO2 catalysts with high catalytic activity and excellent resistant to SO2 and H2 O,were thought to be promising catalysts used in Selective catalytic reduction of nitrogen oxides by *** performance of catalysts is largely affected by calcination ***,effects of calcination temperature on physicochemical property and catalytic activity of CuSO4/TiO2 catalysts were investigated in *** samples calcined at different temperatures were prepared first and then physicochemical properties of the catalyst were characterized by N2 adsorption-desorption,X-ray diffraction,thermogravimetric analysis,Raman spectra,Fourier-transform infrared spectroscopy,X-ray photoelectron spectroscopy,temperature-pro grammed desorption of NH3,temperature-programmed reduction of H2 and in situ diffuse reflectance infrared Fourier transform *** revealed that high calcination temperature had three main effects on the ***,sintering and anatase transform into rutile with increase of calcination temperature,causing a decrement of specific surface ***,decomposition of CuSO4 under higher calcination temperature,resulting in disappears of Br(?)nsted acid sites(S-OH),which had an adverse effect on surface ***,CuO from the decomposition of CuSO4 changed surface reducibility of the catalyst and favored the process of NH3 oxidation to nitrogen oxides(NOx).Thus,catalytic activity of the catalyst calcined under high temperatures(≥600℃)decreased largely.

Universally improving effect of mixed electron donors on the CO_2 fixing efficiency of non-photosynthetic microbial communities from marine environments

The universality of improved CO2 fixing upon the addition of mixed electron donors（MEDs）composed of Na2 S,NO2-,and S2O32-to non-photosynthetic microbial communities（NPMCs）obtained from 12 locations in four oceans of the world was validated. The CO2 fixing efficiencies of NPMCs were universally enhanced by MED compared with those obtained using H2 alone as electron donor,with average increase of about 276%. An increase in microbial inoculation concentration could increase the net amount of CO2 fixing to853.34 mg/L in the presence of MED. NO2-and S2O32-may play the roles of both electron acceptor and electron donor under aerobic conditions,which may improve the energy utilization efficiency of NPMC and enhance the CO2 fixation efficiency. The sequence determination of 16 S ribosomal deoxyribonucleic acid（rDNA） from 150 bacteria of NPMC showed that more than 50% of the bacteria were symbiotic and there were many heterotrophic bacteria such as Vibrio natriegens. These results indicate that NPMC acts as a symbiotic CO2 fixing system. The interaction between autotrophic and heterotrophic bacteria may be a crucial factor supporting ladder utilization and recycling of energy/carbon source.

Phylogenetic diversity of planktonic bacteria in the Chukchi Borderland region in summer

Planktonic bacteria are abundant in the Chukchi Borderland region. However, little is known about their di- versity and the roles of various bacteria in the ocean. Seawater samples were collected from two stations K2S and K4S where sea ice was melting obviously. The analysis of water samples with fluorescence in situ hybridization （FISH） showed that DMSP-degrading bacteria accounted for 13% of the total bacteria at the station K2S. No aerobic anoxygenic phototrophic （AAP） bacteria were detected in both samples. The bacterial communities were characterized by two 16S rRNA gene clone libraries. Sequences fell into four major lineages of the domain Bacteria, including Proteobacteria （Alpha, Beta and Gamma subclasses）, Bac- teroidetes, Actinobacteria and Firmicutes. No significant difference was found between the two clone li- braries. SAR11 and Rhodobacteraceae clades of Alphaproteobacteria and Pseudoalteromonas of Gammapro- teobacteria constituted three dominant fractions in the clone libraries. A total of 191 heterotrophic bacterial strains were isolated and 76% showed extracellular proteolytic activity. Phylogenetic analysis reveals that the isolates fell into Gammaproteobacteria, Bacteroidetes, Actinobacteria and Firmicutes. The most common genus in both the bacterial isolates and protease-producing bacteria was Pseudoalteromonas. UniFrac data showed suggestive differences in bacterial communities between the Chukchi Borderland and the northern Bering Sea.

A New Filled Function with One Parameter to Solve Global Optimization

In this paper, a new filled function with only one parameter is proposed. The main advantages of the new filled function are that it not only can be analyzed easily, but also can be approximated uniformly by a continuously differentiable function. Thus, a minimizer of the proposed filled function can be obtained easily by using a local optimization algorithm. The obtained minimizer is taken as the initial point to minimize the objective function and a better minimizer will be found. By repeating the above processes, we will find a global minimizer at last. The results of numerical experiments show that the new proposed filled function method is effective.

Phylogenetic relationship and phenotypic comparison of Psychrobacter species isolated from polar oceans

To investigate the phylogenetic relationship and biogeography of bacterioplankton in polar oceans, four Psychrobacter strains, BSw10170, BSw20352, BSw20370, and BSw20461, isolated from seawater of the Beijing Sea, the Chukchi Sea, and the Prydz Bay, were characterized by 16S rDNA sequencing and physiological and biochemical testing. Results demonstrated that close relationships existed between the Arctic and Antarctic strains with sequence similarities higher than 97 %. These four Psychrobacter strains not only showed almost identical phenotypic characteristics among them, but also shared a lot of similarities with those related Psychrobacter species, indicating that psychrotolerance and halotolerance of Psychrobacter strains may be among the reasons for their bipolar, even global distribution in marine environments at the genus level.

Phylogenetic diversity of dinoflagellates in polar regions

Because of the limitations of sampling and seasonal study in polar regions, knowledge of dinoflagellate diversity, distribution and ecology are limited. Dinoflagellates have been incidentally reported from polar regions during some seasons and some populations have been reported as components of microalgae. Surveys of molecular diversity link the genotype of dinoflagellates from polar regions with environmental adaptation. In this study, 37 positive clones of dinoflagellates collected from different sites were used for genotype analysis, providing new insights into the biodiversity and distribution of these species based on 18S rRNA sequencing. Diverse genotypes were recorded for the summer season in Kongsfjorden （high Arctic） whilst a single novel genotype of dinoflagellate was recorded from winter samples from the Antarctic Ocean. Data from ice cores suggests that this single dinoflagellate genotype was adapted to extreme cold and clone library screening found that it was occasionally the only microbial eukaryotic genotype found in winter ice cores. The findings of this study could improve our understanding of the diverse dinoflagellate genotypes occurring in these perennially cold microbial ecosystems.
