CX-5461, also known as pidnarulex, is a strong G4 stabilizer and has received FDA fast-track designation for BRCA1-and BRCA2-mutated cancers. However, quantitative measurements of the unfolding rates of CX-5461-G4 complexes which are important for the regulation function of G4s, remain ***, we employ single-molecule magnetic tweezers to measure the unfolding force distributions of c-MYC G4s in the presence of different concentrations of CX-5461. The unfolding force distributions exhibit three discrete levels of unfolding force peaks, corresponding to three binding modes. In combination with a fluorescent quenching assay and molecular docking to previously reported ligand-c-MYC G4 structure, we assigned the ～69 pN peak corresponding to the 1:1（ligand:G4） complex where CX-5461binds at the G4's 5'-end. The ～84 p N peak is attributed to the 2:1 complex where CX-5461 occupies both the 5' and 3'. Furthermore, using the Bell-Arrhenius model to fit the unfolding force distributions,we determined the zero-force unfolding rates of 1:1, and 2:1 complexes to be（2.4 ± 0.9） × 10-8s-1and（1.4 ± 1.0） × 10-9s-1respectively. These findings provide valuable insights for the development of G4-targeted ligands to combat c-MYC-driven cancers.

背景:巨噬细胞亚型表现出组织异质性,脂肪组织巨噬细胞表型很大程度上受肥胖的影响,肥胖脂肪组织巨噬细胞导致的局部和系统炎症反应被认为是肥胖相关代谢性疾病的关键病理学原因。目的:通过总结脂肪组织巨噬细胞不同亚型炎症特点及其与肥胖相关代谢性疾病的关系,为靶向特定巨噬细胞亚型探索肥胖相关代谢性疾病的防治策略提供参考依据。方法:检索中国知网、PubMed数据库相关文献,中文检索词为“肥胖,脂肪组织,脂肪组织巨噬细胞,巨噬细胞极化,代谢性疾病”;英文检索词为“obesity,adipose tissue,adipose tissue macrophages,macrophage polarization,metabolic diseases”,依据入选标准对检索结果进行录用或排除,最终纳入符合标准的91篇文献进行综述。结果与结论:①巨噬细胞具有组织异质性,正常情况下,脂肪组织巨噬细胞以抗炎M2型常驻巨噬细胞为主,维持组织炎症稳态;发生肥胖时,肥大脂肪细胞周围围绕大量外来浸润性巨噬细胞,并且大部分表现出促炎特点。因此,曾经被认为是促炎M1型的脂肪组织巨噬细胞实际上可能是多种促炎亚型的集合。对各种促炎亚型特点的进一步认识有助于深入了解肥胖脂肪组织炎症紊乱的机制。②肥胖情况下,外来浸润性巨噬细胞围绕在肥大脂肪细胞周围形成冠状结构,目前已发现冠状结构中有6种不同亚型,其中大部分表现出促炎特性,少数亚型具备抗炎特点,因此充分发挥抗炎亚型作用的同时抑制促炎亚型的分化可能是缓解肥胖脂肪组织炎症损害的新靶向。③已有研究发现M3、MMe、CD9^(+)、LAM脂肪组织巨噬细胞亚型参与动脉粥样硬化、糖尿病、胰岛素抵抗、癌症等代谢疾病的发生发展,DARC+和MFehi脂肪组织巨噬细胞亚型在治疗非酒精性脂肪肝、肥胖胰岛素抵抗、铁死亡相关代谢疾病中起到关键作用。上述研究进一步说明肥胖脂肪组织外来浸润巨噬细胞引起的炎症紊乱是肥胖诱发代谢性疾病的重要病理基础,对各种亚型特点的进一步深入研究具有重要的理论和现实意义。

背景:巨噬细胞胞葬作用障碍引起的局部和系统炎症损害与多种肥胖相关代谢性疾病有关,且以胞葬作用为靶向的化合物表现出良好的治疗效果。目的:通过综述肥胖对巨噬细胞胞葬作用各个阶段的影响结果分析肥胖抑制胞葬作用的关键机制,总结以胞葬作用为靶向的化合物治疗代谢性疾病的研究现状,以进一步阐明胞葬作用及其与肥胖相关代谢性疾病的关系,为疾病防治策略提供新思路。方法:以“efferocytosis,metabolism,obesity,obese,atherosclerosis,non-alcoholic steatohepatitis,neurodegeneration,tumor,osteoarthritis,diabetes,compound,medicine,treatment”为英文检索词在PubMed和Web of Science数据库检索英文文献,以“胞葬作用”为中文检索词,在中国知网、万方和维普数据库检索中文文献。经严格筛选最终纳入99篇文献进入综述分析。结果与结论:①参与巨噬细胞胞葬作用“寻我”“食我”过程的因子中含有大量凋亡细胞源性因子,因此“寻我”“食我”过程主要受凋亡细胞调控;参与骨架重组和消化过程的胞葬因子主要来源于巨噬细胞,对巨噬细胞胞葬作用活性具有决定性作用。此结果提示,“寻我”“食我”过程的因子表达水平主要反映细胞凋亡情况,在评价巨噬细胞胞葬作用活性时,选择骨架重组和消化阶段的胞葬因子的表达更具科学性。②肥胖抑制巨噬细胞胞葬作用,但肥胖对多数“寻我”“食我”因子及骨架重组因子具有应激性激活作用,对多数消化因子具有抑制作用。此结果进一步说明,消化阶段对胞葬作用活性的决定性意义,并提示部分研究以“寻我”“食我”胞葬因子表达增加作为胞葬作用增强的依据不可靠;且提示未来在探讨以巨噬细胞胞葬作用为靶向的干预策略时,靶向消化阶段胞葬因子可能更有效。③巨噬细胞胞葬作用激活物对多种代谢性疾病治疗有效,但肿瘤组织巨噬细胞胞葬作用抑制物表现出良好的抗癌效果,说明应根据组织炎症特点合理评价胞葬作用的意义。④胞葬作用是2003年提出的一个较新概念,研究历程较短,胞葬因子复杂,目前关于肥胖对胞葬作用影响的研究仅涉及冰山一角并且大部分处于粗浅水平,对其更深入的机制探讨仍需大量科学实验的进一步验证。

This paper mainly introduces the parallel physics-informed neural networks(PPINNs)method with regularization strategies to solve the data-driven forward-inverse problems of the variable coefficient modified Korteweg-de Vries(VC-MKdV)*** the forward problem of the VC-MKdV equation,the authors use the traditional PINN method to obtain satisfactory data-driven soliton solutions and provide a detailed analysis of the impact of network width and depth on solving accuracy and ***,the author finds that the traditional PINN method outperforms the one with locally adaptive activation functions in solving the data-driven forward problems of the VC-MKdV *** for the data-driven inverse problem of the VC-MKdV equation,the author introduces a parallel neural networks to separately train the solution function and coefficient function,successfully addressing the function discovery problem of the VC-MKdV *** further enhance the network’s generalization ability and noise robustness,the author incorporates two regularization strategies into the *** amount of numerical experimental data in this paper demonstrates that the PPINNs method can effectively address the function discovery problem of the VC-MKdV equation,and the inclusion of appropriate regularization strategies in the PPINNs can improves its performance.

Nanocarbon-based conductive membranes, especially carbon nanotube (CNT)-based membranes, have tremendous potential for wastewater treatment and water purification because of their excellent water permeability and selectivity, as well as their electrochemically enhanced performance (e.g., improved antifouling ability). However, it remains challenging to prepare CNT membranes with high structural stability and high electrical conductivity. In this study, a highly electroconductive and structurally stable polyphenylene/CNT (PP/CNT) composite membrane was prepared by electropolymerizing biphenyl on a CNT hollow fiber membrane. The PP/CNT membrane showed 3.4 and 5.0 times higher electrical conductivity than pure CNT and poly(vinyl alcohol)/CNT (PVA/CNT) membranes, respectively. The structural stability of the membrane was superior to that of the pure CNT membrane and comparable to that of the PVA/CNT membrane. The membrane fouling was significantly alleviated under an electrical assistance of −2.0 V, with a flux loss of only 11.7% after 5 h filtration of humic acid, which is significantly lower than those of PP/CNT membranes without electro-assistance (56.8%) and commercial polyvinylidene fluoride (PVDF) membranes (64.1%). Additionally, the rejection of negatively charged pollutants (humic acid and sodium alginate) was improved by the enhanced electrostatic repulsion. After four consecutive filtration-cleaning cycle tests, the flux recovery rate after backwashing reached 97.2%, which was much higher than those of electricity-free PP/CNT membranes (67.0%) and commercial PVDF membranes (61.1%). This study offers insights into the preparation of stable conductive membranes for membrane fouling control in potential water treatment applications.

*** In recent years,China has carried out an extensive preventative battle against air,water,and soil pollution,and the nation’s environmental quality-as reflected by conventional pollutant indicators—has significantly *** the same time,the issue of emerging contaminants(ECs)is beginning to receive increasing *** generally refer to newly discovered or noticeable pollutants that pose risks to the ecological environment or human *** they have not been included in environmental management,or existing management measures are insufficient to effectively prevent and control their *** ECs of greatest concern generally include persistent organic pollutants(POPs),endocrine-disrupting chemicals(EDCs),pharmaceuticals and personal care products(PPCPs),and *** four categories of ECs are not entirely separate,as they interrelate with each other(Fig.1).Chemical production and product usage are the main sources of *** is the world’s largest producer and consumer of bulk chemicals,and the production value of China’s chemical industry is predicted to reach 50%of the global total by 2030[1].Scientific control of ECs based on their environmental risk assessment is a necessary way to support the prevention and legal governance of ECs.

Efficient Metal Recovery from Industrial Wastewater:Potential Oscillation and Turbulence Mode for Electrochemical System

Efficient metal recovery from industrial wastewater facilitates addressing of the environmental hazards and resource requirements of heavy *** conventional electrodeposition recovery method is hampered by the limitations of interfacial ion transport in charge-transfer reactions,creating challenges for simultaneous rapid and high-quality metal ***,we proposed integrating a transient electric field(TE)and swirling flow(SF)to synchronously enhance bulk mass transfer and promote interfacial ion *** investigated the effects of the operation mode,transient frequency,and flow rate on metal recovery,enabling determination of the optimal operating conditions for rapid and efficient sequential recovery of Cu in TE&SF *** conditions included low and high electric levels of 0 and 4 V,a 50%duty cycle,1 kHz frequency,and 400 L·h^(-1)flow *** kinetic coefficients of TE&SF electrodeposition were 3.5-4.3 and 1.37-1.97 times that of single TE and SF electrodeposition,*** the deposition process under TE and SF conditions confirmed the efficient concurrence of interfacial ion transport and charge transfer under TE and SF synergy,which achieved rapid and highquality metal ***,the combined deposition strategy is considered an effective technique for reducing metal pollution and promoting resource recycling.
