Hypomethylation of glycine dehydrogenase promoter in peripheral blood mononuclear cells is a new diagnostic marker of hepatitis B virus-associated hepatocellular carcinoma

Background: Glycine dehydrogenase(GLDC) plays an important role in the initiation and proliferation of several human cancers. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnostic value for hepatitis B virus-associated hepatocellular carcinoma(HBV-HCC). Methods: We enrolled 197 patients, 111 with HBV-HCC, 51 with chronic hepatitis B(CHB), and 35 healthy controls(HCs). The methylation status of GLDC promoter in peripheral mononuclear cells(PBMCs) was identified by methylation specific polymerase chain reaction(MSP). The mRNA expression was examined using real-time quantitative polymerase chain reaction(q PCR). Results: The methylation frequency of the GLDC promoter was significantly lower in HBV-HCC patients(27.0%) compared to that in CHB patients(68.6%) and HCs(74.3%)( P < 0.001). The methylated group had lower alanine aminotransferase level( P = 0.035) and lower rates of tumor node metastasis(TNM) Ⅲ/Ⅳ( P = 0.043) and T3/T4( P = 0.026). TNM stage was identified to be an independent factor for GLDC promoter methylation. GLDC mRNA levels in CHB patients and HCs were significantly lower than those in HBV-HCC patients( P = 0.022 and P < 0.001, respectively). GLDC mRNA levels were significantly higher in HBV-HCC patients with unmethylated GLDC promoters than those with methylated GLDC promoters( P = 0.003). The diagnostic accuracy of alpha-fetoprotein(AFP) combined with GLDC promoter methylation for HBV-HCC was improved compared with that of AFP alone(AUC: 0.782 vs. 0.630, P < 0.001). In addition, GLDC promoter methylation was an independent predictor for overall survival of HBV-HCC patients( P = 0.038). Conclusions: The methylation frequency of GLDC promoter was lower in PBMCs from HBV-HCC patients than that from patients with CHB and HCs. The combination of AFP and GLDC promoter hypomethylation significantly improved the diagnostic accuracy of HBV-HCC.

Cationic engineered nanodiamonds for efficient antibacterial surface with strong wear resistance

The spread of diseases caused by bacterial adhesion and immobilization in public places constitutes a serious threat to public *** of bacteria spread by the construction of an antibacterial surface takes precedence over post-infection ***,we demonstrate an effective antibacterial surface with strong wear resistance by constructing cationic engineered nanodiamonds(C-NDs).The C-NDs with positive surface potentials interact effectively with bacteria through electrostatic interactions,where the C-NDs act on the phospholipid bilayer and lead to bacterial membrane collapse and rupture through hydrogen bonding and residual surface oxygen-containing reactive *** this case,bactericidal rate of 99.99%and bacterial biofilm inhibition rate of more than 80%can be achieved with the C-NDs concentration of 1 mg/*** addition,the C-NDs show outstanding antibacterial stability,retaining over 87%of the antibacterial effect after stimulation by adverse environments of heat,acid,and external ***,an antibacterial surface with high wear resistance obtained by integrating C-NDs with commercial plastics has been *** antibacterial surface with a mass fraction of 1 wt.%C-NDs improved abrasion resistance by 3981 times,with 99%killing of adherent *** work provides an effective strategy for highly efficient antibacterial wear-resistant surface,showing great practical applications in public health environments.

Sea-level change in coastal areas of China:Status in 2021

Nomogram prediction of vessels encapsulating tumor clusters in small hepatocellular carcinoma≤3 cm based on enhanced magnetic resonance imaging

Study of deep transportation and plugging performance of deformable gel particles in porous media

Deformable gel particles(DGPs) possess the capability of deep profile control and flooding. However, the deep migration behavior and plugging mechanism along their path remain unclear. Breakage, an inevitable phenomenon during particle migration, significantly impacts the deep plugging effect. Due to the complexity of the process, few studies have been conducted on this subject. In this paper, we conducted DGP flow experiments using a physical model of a multi-point sandpack under various injection rates and particle sizes. Particle size and concentration tests were performed at each measurement point to investigate the transportation behavior of particles in the deep part of the reservoir. The residual resistance coefficient and concentration changes along the porous media were combined to analyze the plugging performance of DGPs. Furthermore, the particle breakage along their path was revealed by analyzing the changes in particle size along the way. A mathematical model of breakage and concentration changes along the path was established. The results showed that the passage after breakage is a significant migration behavior of particles in porous media. The particles were reduced to less than half of their initial size at the front of the porous media. Breakage is an essential reason for the continuous decreases in particle concentration, size, and residual resistance coefficient. However, the particles can remain in porous media after breakage and play a significant role in deep plugging. Higher injection rates or larger particle sizes resulted in faster breakage along the injection direction, higher degrees of breakage, and faster decreases in residual resistance coefficient along the path. These conditions also led to a weaker deep plugging ability. Smaller particles were more evenly retained along the path, but more particles flowed out of the porous media, resulting in a poor deep plugging effect. Tuning heterogeneous microstructures to enhance mechanical properties of nano-TiN particle reinforced Haynes 230 composites by laser powder bed fusion

Laser powder bed fusion(LPBF) is considered to be one of the most promising additive manufacturing technologies for producing components with geometries and high geometrical precision that are unattainable by traditional technologies. The superalloy exhibits exceptional mechanical and high-temperature performances,rendering it a prime candidate for advanced aero-engine applications. Despite the high demand for LPBF-manufactured superalloys, the superalloy is one of the materials manufactured difficultly by LPBF due to their large laser absorptivity fluctuation, poor molten pool stability and sharp temperature gradient. Hence, superalloys are characterized by severe pores, undesirable coarse columnar grains and poor mechanical properties. In this work, the effect of nano-Ti N particles on defects, molten pool characteristics and microstructure and performance of the composites were investigated. The 4.5 wt% Ti N/Haynes230 samples exhibited exceptional nanohardness and elastic modulus with maximum values reaching 5.53 GPa and 240.03 GPa, respectively. These superior mechanical properties were attributed to the combined effects of spatter and gas pore inhibition, grain refinement and duplex nanophases strengthening. Moreover, the stability of molten pool was enhanced, and spatter was effectively suppressed by adding nano-Ti N particles, while grain refinement and columnar to equiaxed transitions were promoted. Furthermore, the matrix exhibited a high dislocation density due to a significant hindrance of dislocation movement caused by massive nano-phases(e.g., Ti N and M23C6), resulting in the formation of extensive dislocation tangles and *** work offers novel insights into the role of nanoparticles reinforced superalloy composites by LPBF.

Sodium butyrate protects against focal cerebral ischemic injury through regulation of the nuclear receptor Nur77

Focal cerebral ischemia is a major cardiovascular disease that seriously threatens human health. Sodium butyrate has been identified as a neuromodulator in cerebral ischemia but the underlying mechanisms remain unclear. The present study aimed to clarify the neuronal anti-inflammatory mechanisms of sodium butyrate against focal cerebral ischemia. A focal cerebral ischemic rat model was established by administering sodium butyrate prior to middle cerebral artery occlusion (MCAO). The neuroprotective effects of sodium butyrate were evaluated 24 h after MCAO， using neurological score， infarction volume， and inflammatory biomarkers as the criteria. The expressions of nuclear receptor subfamily 4 group A1 (Nur77)， nod-like receptor protein 3 (NLRP3)， caspase-1 and interleukin-1β (IL-1β) in brain tissues were also analyzed. To explore the role of sodium butyrate in Nur77/NLRP3-mediated inflammatory response， oxygen and glucose deprivation (OGD)-stimulated HT22 cells were treated with sodium butyrate and siNur77. Sodium butyrate treatment significantly attenuated neurological impairment and neuroinflammation induced by cerebral ischemia. Sodium butyrate-mediated dysregulation of Nur77 expression and production of NLRP3-related inflammatory factors were observed in cerebral ischemia in vivo and in vitro. In addition， Nur77 was identified as a negative regulator of NLRP3 in HT22 cells under OGD. The results indicate that the anti-inflammatory effect of sodium butyrate was achieved by upregulating Nur77 expression， which subsequently suppressed neuronal NLRP3-mediated inflammatory response. Overall， sodium butyrate clearly has a neuroprotective effect against focal cerebral ischemia by modulating Nur77/NLRP3-mediated inflammatory response.
