Adsorption,in vitro digestion and human gut microbiota regulation characteristics of three Poria cocos polysaccharides

Poria cocos(PC)is a famous traditional Chinese medicine(TCM)and a widely used healthcare ingredient,which has antiobesity,enhancing immunity and improving sleep effects.Traditionally,only water-soluble poria polysaccharide(WSP)is extracted and applied for clinical application,while insoluble polysaccharide(alkali-soluble poria polysaccharide,ASP)is discarded as herb residue.However,the whole PC has also been historically utilized as functional herbal food.Considering the beneficial role of dietary fiber and the traditional use of PC,ASP may also contribute substantially to the therapy function of PC.Compared to WSP,little attention has been paid to ASP and ASP modified product carboxymethyl poria polysaccharide(CMP)which has been used as an antitumor adjuvant drug.In this study,the oil,cholesterol,metal ions and polyphenols adsorption ability,in vitro simulated digestive and the gut microbiota fermentation characteristics of WSP,ASP and CMP were studied to evaluate the functional values of three P.cocos polysaccharides(PCPs).The results showed that all three PCPs had good adsorption capacity on cholesterol,polyphenols and metal ions(Cd^(2+)/Zn^(2+)/Mg^(2+)),among which ASP showed the highest capacity than WSP and CMP.The adsorption capacity of all three PCPs on heavy metal ions(Cd^(2+)/Zn^(2+))was stronger than that of non-heavy metal ions(Mg^(2+));The in vitro digestibility of all three PCPs was very low,but WSP was slightly higher than ASP and CMP;Moreover,the indigestible residue of all three PCPs could improve the richness and diversity of gut microbiota,among which ASP had the greatest influence.In general,ASP and CMP could significantly promote the proliferation of some probiotics and inhibit the growth of some harmful bacteria.The gut microbiota diversity of CMP was reduced,but the richness of probiotics,especially Parabacteroides distasonis was significantly enhanced compared with the ASP group,and the growth of harmful bacteria Klebsiella pneumoniae was inhibited after CMP treatment.The short-chain fatty

Nasopharyngeal carcinoma(NPC),a highly invasive and metastatic cancer,is widely prevalent in South China,Southeast Asia,and North Africa.NPC patients with stages I and II are often asymptomatic.Therefore,NPC patients are usually diagnosed with advanced-stage(stages III and IV)at their first visit.Detection of NPC at early stage is the key to improve the survival of patients.Epstein-Barr virus(EBV)plays an essential role in NPC occurrence in endemic areas.EBV-based biomarkers,including EBV antibody in serum and EBV DNA in plasma,have been specifically proposed for NPC early detection.Herein,we summarize recent advances in biomarkers for the early detection of NPC.

Effect of early stepwise cardiopulmonary rehabilitation on function and quality of life in sepsis patients

BACKGROUND Sepsis,as a non-limiting host infection disease,can be accompanied by serious complications such as organ failure,which seriously threatens patient quality of life.AIM To investigate the effect of early stepwise cardiopulmonary rehabilitation on cardiopulmonary function and quality of life in patients evacuated from mechanical ventilation with sepsis.METHODS A total of 80 patients with sepsis who were hospitalized in our hospital from January 2021 to January 2022 were selected and divided into the observation group(n=40)and the control group(n=40)according to the random number table method.The observation group was treated with early stepwise cardiopulmonary rehabilitation,and the control group was treated with a conventional treatment regimen.Cardiac function indexes(central venous pressure,cardiac troponin I,B-type brain natriuretic peptide),lung function indicators(diaphragmatic mobility,changes in central venous oxygen saturation,oxygenation index),and quality of life(Quality of Life Evaluation Scale)were compared between the two groups after treatment.RESULTS After treatment,the central venous pressure,diaphragm mobility,central venous oxygen saturation,oxygenation index,and Quality of Life Evaluation Scale scores in the observation group were higher than those in the control group,and the differences were statistically significant(P<0.05).The observation group was less than that of the control group for other parameters,and the differences were statistically significant(P<0.05).CONCLUSION Early stepwise cardiopulmonary rehabilitation can effectively enhance cardiac and pulmonary function and improve the quality of life in patients evacuated from mechanical ventilation with sepsis.

The endoplasmic reticulum(ER)is connected to mitochondria through mitochondria-associated ER membranes(MAMs).MAMs provide a framework for crosstalk between the ER and mitochondria,playing a crucial role in regulating cellular calcium balance,lipid metabolism,and cell death.Dysregulation of MAMs is involved in the development of chronic liver disease(CLD).In CLD,changes in MAMs structure and function occur due to factors such as cellular stress,inflammation,and oxidative stress,leading to abnormal interactions between mitochondria and the ER,resulting in liver cell injury,fibrosis,and impaired liver function.Traditional Chinese medicine has shown some research progress in regulating MAMs signaling and treating CLD.This paper reviews the literature on the association between mitochondria and the ER,as well as the intervention of traditional Chinese medicine in regulating CLD.

BACKGROUND The study on predicting the differentiation grade of colorectal cancer(CRC)based on magnetic resonance imaging(MRI)has not been reported yet.Developing a non-invasive model to predict the differentiation grade of CRC is of great value.AIM To develop and validate machine learning-based models for predicting the differ-entiation grade of CRC based on T2-weighted images(T2WI).METHODS We retrospectively collected the preoperative imaging and clinical data of 315 patients with CRC who underwent surgery from March 2018 to July 2023.Patients were randomly assigned to a training cohort(n=220)or a validation cohort(n=95)at a 7:3 ratio.Lesions were delineated layer by layer on high-resolution T2WI.Least absolute shrinkage and selection operator regression was applied to screen for radiomic features.Radiomics and clinical models were constructed using the multilayer perceptron(MLP)algorithm.These radiomic features and clinically relevant variables(selected based on a significance level of P<0.05 in the training set)were used to construct radiomics-clinical models.The performance of the three models(clinical,radiomic,and radiomic-clinical model)were evaluated using the area under the curve(AUC),calibration curve and decision curve analysis(DCA).RESULTS After feature selection,eight radiomic features were retained from the initial 1781 features to construct the radiomic model.Eight different classifiers,including logistic regression,support vector machine,k-nearest neighbours,random forest,extreme trees,extreme gradient boosting,light gradient boosting machine,and MLP,were used to construct the model,with MLP demonstrating the best diagnostic performance.The AUC of the radiomic-clinical model was 0.862(95%CI:0.796-0.927)in the training cohort and 0.761(95%CI:0.635-0.887)in the validation cohort.The AUC for the radiomic model was 0.796(95%CI:0.723-0.869)in the training cohort and 0.735(95%CI:0.604-0.866)in the validation cohort.The clinical model achieved an AUC of 0.751(95%CI:0.661-0.842)in the training cohort and

土地利用/覆被变化会增加区域碳储量或引发碳损耗,进而影响全球气候.研究湾区土地利用与碳储量变化的影响机制,对区域生态系统保护及社会经济可持续发展具有重要意义.基于InVEST模型和PLUS模型,分析了粤港澳大湾区土地利用与碳储量的动态变化特征及影响机制,进一步模拟预测了2030年3种发展情景(自然趋势情景、建设用地优先情景和生态优先情景)下土地利用类型变化对碳储量的影响.结果表明:(1)2000~2020年粤港澳大湾区的耕地(减少5.38%)和建设用地(增加8.68%)变化最显著,其余用地类型的变化较小.林地作为大湾区的主要用地类型,同样也是重要的碳库,空间上集中分布于东、北、西三面的台地和丘陵地区,碳储存低值区集中于中部平原,以建设用地为主.(2)受不同用地类型相互转化的影响,粤港澳大湾区的碳损失了20.12×10^(6) Mg.耕地和林地面积的减少以及建设用地面积的增加是碳损耗的主要原因.(3)模拟研究发现生态保护措施将有效提高区域碳储量.具体来说,当耕地、林地和草地向建设用地的转化减少20%,湿地和水体向建设用地的转化减少30%时(生态优先情景),区域碳储量可增加2.58×10^(6) Mg.研究结果可以为土地利用和城市发展规划的制定提供理论依据,促进粤港澳大湾区“碳达峰碳中和”目标的实现.

Assessment and improvement of Noah-MP for simulating water and heat exchange over alpine grassland in growing season

Alpine grassland is the main ecosystem of the Tibetan Plateau(TP),thus accurate simulation of water and heat exchange in the grassland will significantly enhance the understanding of the land-atmosphere interaction process on the TP.In this study,we assessed and improved the ensemble numerical simulations of the community Noah land surface model with multiparameterization options(Noah-MP)by using observations collected from four alpine grassland observation sites.The four observation sites belong to the upper Heihe River Basin Integrated Observatory Network located in the northeastern part of the TP.First,an ensemble of 1008 numerical simulation experiments,based on multiparameterization options of seven physical processes/variables in the Noah-MP,was carried out for the vegetation growing season.The Taylor skill score was then used to assess the model performance and select the optimal combination of parameterization options for a more exact simulation of the water and heat exchange in alpine grassland.The accuracy of Noah-MP simulation was further improved by introducing new parameterizations of thermal roughness length,soil hydraulic properties,and vertical root distribution.It was found that:(1)Simulation of water and heat exchange over alpine grassland in the growing season was mainly affected by the parameterizations of dynamic vegetation,canopy stomatal resistance,runoff and groundwater dynamics,and surface exchange coefficient for heat transfer.Selection of different parameterization options for these four physical processes/variables led to large differences in the simulation of water and heat fluxes.(2)The optimal combination of parameterization options selected in the current Noah-MP framework suffered from significant overestimation of sensible heat flux(H)and underestimation of soil moisture(θ)at all observation sites.(3)The overestimation of H was significantly improved by introducing a new parameterization of thermal roughness length.Furthermore,the underestimation ofθwas resolved by introducing a

Constructing a functional hybrid coating appears to be a promising strategy for addressing the poor corrosion resistance and insufficient endothelialization of Mg-based stents.Nevertheless,the steps for preparing composite coatings are usually complicated and time-consuming.Herein,a novel composite coating,composed of bioactive magnesium thioctic acid(MTA)layer formed by deposition and corrosion-resistant magnesium hydroxide(Mg(OH)_(2))layer grown in situ,is simply fabricated on ZE21B alloys via one-step electrodeposition.Scanning electron microscopy(SEM)shows that the electrodeposited coating has a compact and uniform structure.And the high adhesion of the MTA/Mg(OH)_(2)hybrid coating is also confirmed by the micro-scratch test.Electrochemical test,scanning kelvin probe(SKP),and hydrogen evolution measurement indicate that the hybrid coating effectively reduces the degradation rate of Mg substrates.Haemocompatibility experiment and cell culture trial detect that the composite coating is of fine biocompatibility.Finally,the preparation mechanism of MTA/Mg(OH)_(2)hybrid coatings is discussed and proposed.This coating shows a great potential application for cardiovascular stents.

TANK-binding kinase 1(TBK1)is an essential protein kinase for activation of interferon regulatory factor 3(IRF3)and induction of the type I interferons(IFN-I).Although the biochemical regulation of TBK1 activation has been studied,little is known about how enterovirus 71(EV71)employs the deubiquitinases(DUBs)to regulate TBK1 activation for viral immune evasion.Here,we found that EV71 infection upregulated the expression of ubiquitinspecific protease 24(USP24).Further studies revealed that USP24 physically interacted with TBK1,and can reduce K63-linked polyubiquitination of TBK1.Knockdown of USP24 upregulated TBK1 K63-linked polyubiquitination,promoted the phosphorylation and nuclear translocation of IRF3,and in turn improved IFN-I production during EV71 infection.As a consequence,USP24 knockdown dramatically inhibited EV71 infection.This study revealed USP24 as a novel regulator of TBK1 activation,which promotes the understanding of immune evasion mechanisms of EV71 and could provide a potential strategy for treatment of EV71 infection.

Effects of Rectangular Wing Vortex Generators on the Thermal-Hydraulic Performance of Louvered Fin and Flat Tube Heat Exchanger

In this paper,a novel composite heat transfer enhancement technique comprised of louvered fins(LFs)and rectangular wing vortex generators(RWVGs)is proposed to improve the LF side thermal-hydraulic performance of louvered fin and flat tube heat exchangers(LFHEs).After validation of the LF side pressure dropΔP and heat transfer coefficient hLFof the baseline by experiments,the numerical method is applied to investigate the influential mechanisms of the RWVG parameters(the number N(7 to 15),attack angleβ(30°to 90°),height H_(VG)(0.8 mm to 2 mm)and width W_(VG)(0.8 mm to 1.2 mm))on the performance of the LFHE in the velocity range of 3 m/s to 10 m/s.Results show that thermal-hydraulic performance of the LFHE is significantly impacted by the RWVGs,and according to the performance evaluation criteria(PEC),the LFHE achieves its optimal thermal-hydraulic performance when N=7,β=45°,H_(VG)=1.8 mm and W_(VG)=1 mm.Compared to the baseline,the maximum,minimum and average increments of PEC for the optimal case are 13.85%,4.67%and 8.39%,respectively.
