Andrographis paniculata contains two main diterpenoid constituents,andrographolide(AGP)and 14-deoxy-11,12-didehydroandrographolide(DDAG),which were found to exhibit antiasthma effects in a mouse asthma ***,due to inadequacies of both compounds in terms of drug-likeness,DDAG analogues were semisynthesised to tackle these *** objective of the study was to investigate the potential of DDAG analogues as new antiasthma *** the analogues,(SRS27)was proven to inhibit cysteinyl leukotrienes(CysLT)and nitric oxide(NO)synthesis in mouse macrophages,like ***,on the other hand,failed to exhibit such ***27 is less cytotoxic than AGP,suggests that a simple chemical modification of DDAG produces a compound with CysLT and NO inhibitory activites similar to AGP while maintaining toxicity profile similar to *** is interesting to note that other analogues such as SRS28,SRS49,SRS76 and SRS83 with chemical modifications on the same carbon numbers 3 and 19 of DDAG were unable to show inhibition of CysLT and NO ***,the potential anti-inflammatory effect of SRS27 was investigated in ovalbumin(OVA)-induced mouse asthma *** compound was administered in a prophylactic manner and showed a substantial decrease in mouse asthma model ***27 at 3mg·kg-1 significantly reduced OVA-induced total cell such as macrophages,eosinophils,lymphocytes and neutrophils,as well as inflammatory cytokines such as IL-4,IL-5,IL-13 and eotaxin in bronchoalveolar lavage BAL *** compound also suppressed serum IgE *** addition,SRS27 suppressed mucus hyper-secretion and expression of inflammatory mediators such as TNF-α,MCP-1,Muc5 ac,RANTES,IL-33 and ***27 is the first known DDAG derivative tested positive in mouse asthma model and as such SRS27 could serve as a prototype prophylactic agent.

The two lim domain-containing proteins from plants （lims） typically exhibit a dual cytoplasmic-nuclear dis-tribution, suggesting that, in addition to their previously described roles in actin cytoskeleton organization, they partici-pate in nuclear processes. Using a south-western blot-based screen aimed at identifying factors that bind to plant histone gene promoters, we isolated a positive clone containing the tobacco lim protein Wlim2 （NtWlim2） cDNA. Using both green fluorescent protein （GFP） fusion-and immunology-based strategies, we provide clear evidence that NtWlim2 local-izes to the actin cytoskeleton, the nucleus, and the nucleolus. Interestingly, the disruption of the actin cytoskeleton by latrunculin B significantly increases NtWlim2 nuclear fraction, pinpointing a possible novel cytoskeletal-nuclear crosstalk. Biochemical and electron microscopy experiments reveal the ability of NtWlim2 to directly bind to actin filaments and to crosslink the latter into thick actin bundles. Electrophoretic mobility shift assays show that NtWlim2 specifically binds to the conserved octameric cis-elements （Oct） of the Arabidopsis histone H4A748 gene promoter and that this binding largely relies on both lim domains. Importantly, reporter-based experiments conducted in Arabidopsis and tobacco proto-plasts confirm the ability of NtWlim2 to bind to and activate the H4A748 gene promoter in live cells. Expression studies indicate the constitutive presence of NtWlim2 mRNA and NtWlim2 protein during tobacco BY-2 cell proliferation and cell cycle progression, suggesting a role of NtWlim2 in the activation of basal histone gene expression. Interestingly, both live cell and in vitro data support NtWlim2 di/oligomerization. We propose that NtWlim2 functions as an actin-stabilizing protein, which, upon cytoskeleton remodeling, shuttles to the nucleus in order to modify gene expression.

AIM:To investigate the effect of electroacupuncture(EA)on the mitochondria-dependent apoptotic signaling pathway in the ciliary muscle of guinea pigs with negative lensinduced myopia(lim).METHODS:Guinea pigs were randomly divided into normal control(NC)group,lim group,lim+SHAM acupoint(lim+SHAM)group,and lim+EA *** in the NC group received no intervention,while those in other three groups were covered with-6.0 diopter(D)lenses on right ***,animals in the lim+EA group received EA at Hegu(LI4)combined with Taiyang(EX-HN5)acupoints,while those in the lim+SHAM group were treated at sham *** treatments for 1,2,and 4wk,morphological changes in ciliary muscles were observed with hematoxylin and eosin(H&E)staining and nick end labeling(TUNEL),and the expression of the mitochondrial apoptotic signaling pathway-related molecules in ciliary muscles was measured by real-time quantitative polymerase chain reaction(qPCR)and Western ***,the adenosine triphosphate(ATP)contents were also determined in ciliary ***:Axial length increased significantly in the lim and lim+SHAM groups and decreased in the lim+EA *** ciliary muscle fibers were broken and destroyed in both lim and lim+SHAM groups,whereas those in the lim+EA group improved *** assay showed the number of apoptotic cells increased in the lim and lim+SHAM groups,whereas reduced in the lim+EA *** contents showed a significant decrease in the lim and lim+SHAM groups,whereas increased after EA *** with the NC group,the dynamin-related protein 1(DRP1),Caspase3,and apoptotic protease activator 1(APAF1)levels were significantly increased in the lim group and decreased in the lim+EA ***:The results provide evidence of EA inhibiting the development of myopia by regulating the mitochondrial apoptotic signaling pathway.

Endoscopic submucosal dissection for early gastric cancer with undifferentiated-type histology:A meta-analysis

AIM: To evaluate the efficacy and safety of endoscopicsubmucosal dissection(ESD) for early gastric cancer(EGC) with undifferentiated-type ***: A systematic literature review was conducted using the core databases. Complete resection,curative resection, en bloc resection, recurrence and adverse event rate were extracted and analyzed. A random effect model was applied. The methodological quality of the enrolled studies was assessed using the Newcastle-Ottawa Scale. Publication bias was evaluated using a funnel plot, the trim and fill method, Egger's test,and a rank correlation ***: Fourteen retrospective studies between2009 and 2014 were identified(972 EGC lesions with undifferentiated-type histology). The total en bloc and complete resection rates were estimated as92.1%(95%CI: 87.4%-95.2%) and 77.5%(95%CI:69.3%-84%), respectively. The total curative resection rate was 61.4%(95%CI: 44.5%-75.9%). The overall recurrence rate was 7.6%(95%CI: 3.4%-16%).limited to histologically diagnosed expanded-criteria lesions, the en bloc and complete resection rates were91.2% and 85.6%, respectively. The curative resection rate was 79.8%.CONCLUSION: In this analysis, ESD is a technically feasible treatment modality for EGC with undifferentiatedtype histology. Long-term studies are needed to confirm these therapeutic outcomes.

利用大白菜细胞核复等位基因型雄性不育两用系‘AB01’的不育株,与多国芸薹属植物基因组计划MBGP的基础材料‘Chiifu’杂交及回交,构建BC1群体。根据SSR检测体系中各主要成份浓度对扩增结果的影响,优化反应体系。选用250对SSR引物,对大白菜核基因雄性不育复等位基因Ms进行SSR+BSA分析,获得了8对多态性引物。在101株BC1个体间对这些引物进一步检测,得到2个与Ms基因连锁的SSR标记cnu_m273和cnu_m295。经连锁分析,二者位于Ms的同一侧,遗传距离分别为4.95cM和7.92cM。

The Basidiomycota constitutes a major phylum of the kingdom Fungi and is second in species numbers to the *** present work provides an overview of all validly published,currently used basidiomycete genera to date in a single *** outline of all genera of Basidiomycota is provided,which includes 1928 currently used genera names,with 1263 synonyms,which are distributed in 241 families,68 orders,18 classes and four *** provide brief notes for each accepted genus including information on classification,number of accepted species,type species,life mode,habitat,distribution,and sequence ***,three phylogenetic analyses with combined LSU,SSU,5.8s,rpb1,rpb2,and ef1 datasets for the subphyla Agaricomycotina,Pucciniomycotina and Ustilaginomycotina are conducted,*** time estimates are provided to the family level with 632 species from 62 orders,168 families and 605 *** study indicates that the divergence times of the subphyla in Basidiomycota are 406-430 Mya,classes are 211-383 Mya,and orders are 99-323 Mya,which are largely consistent with previous *** this study,all phylogenetically supported families were dated,with the families of Agaricomycotina diverging from 27-178 Mya,Pucciniomycotina from 85-222 Mya,and Ustilaginomycotina from 79-177 *** times as additional criterion in ranking provide additional evidence to resolve taxonomic problems in the Basidiomycota taxonomic system,and also provide a better understanding of their phylogeny and evolution.

In HCl-NaOAc of pH 4.30 buffer medium,pioglitazone hydrochloride reacts with eosin Y to form an ion-association complex,and the colour of solutions change obviously at the maximum fading wavelength 516 *** fading colour intensity is also proportional to the concentration of pioglitazone hydrochloride,and a sensitive spectrophotometric method is developed for determination of pioglitazone *** concentration of pioglitazone hydrochloride in the range of 7.05×10-7 mol/L～2.48×10-5 mol/L obeys Beer’ law at the maximum fading wavelength 516 *** apparent molar absorptivity ε516 is 3.65×104 L·mol-1·cm-1,and the detection limit is 3.16×10-7 mol/*** method has high sensitivity and fairly good selectivity,and was applied to the determination of pioglitazone hydrochloride in medicine samples and human serum and human urine with satisfactory results.
