Monitoring the Cultivated Slope Land in the Three Gorges Reservoir Area Based on Remote Sensing and GIS

By means of combining auto-extraction with manual interpretation, the current distribution information about cultivated land is obtained. The distribution information of 1992 is extracted from the dynamic polygons of 2002. The monitoring mini-system of the cultivated slope land is established. In the system, detailed surveys, focused on the resources of cultivated slope land, are carried out. The results indicate that the area of the cultivated slope land is very large. Meanwhile, there are lots of cultivated steep slopes with gradient above 3.°. The areas of steep land cultivated had been slowly reduced from 1992 to 2002. At the same time, the pressures of returning farm land to forestry are great in all counties. The conflicts between population growth, insufficient grain supply and stagnant economic development sharpen increasingly. It is inevitable to improve the agricultural structure.

The paper makes detailed analysis on the changing processes of the main types of land use in Three Gorges Reservoir Area through the techniques of RS （Remote Sense） and GIS （Geography Information System）. Based on the theory of landscape ecology, many types of landscape pattern indexes are calculated and made correlation analysis. The results indicate that： in 1995 the ecological environment of Three Gorges Reservoir Area was the most vulnerable in the recent 50 years, but resumed in 2002; the landscape pattern diversity index, fractal dimension index and fragmentation index has close correlation with the area of land use/cover change （LUCC）. The research also releases that the increase of the cultivated land, grassland and construction land deteriorates the landscape ecological pattern, while the increase of forest land area will help restore the landscape ecological pattern to its original state. The rapid change of grassland is an important index of the change of landscape pattern, because it has the most effect on change of landscape pattern index.

Cloning and characterization of interferon stimulated genes Viperin and ISG15,and their promoters from snakehead Channa argus

By suppression subtractive hybridization, rapid amplification of cDNA ends and gene walking methods, interferon stimulated genes (ISGs), Viperin and ISG15, and their promoters have been cloned and characterized from snakehead Channa argus. The Viperin cDNA was found to be 1474 nt and contain an open reading frame (ORF) of 1059 nt that translates into a putative peptide of 3.2 amino acid (aa). The putative peptide of Viperin shows high identity to that in teleosts and mammals except for the N-terminal 70 aa. The ISG15 cDNA was found to be 758 nt and contain an ORF of 468 nt that translates into a putative peptide of 155 aa. The putative peptide of ISG15 is composed of two tandem repeats of ubiquitin-like (UBL) domains, and a canonical conjugation motif (LRGG) at C-terminal. Viperin and ISG15 promoter regions were characterized by the presence of interferon stimulating response elements (ISRE) and γ-IFN activation sites (GAS). ISRE is a feature of IFN-induced gene promoter and partially overlaps interferon regulatory factor (IRF) 1 and IRF2 recognition sites. GAS is responsible for the γ-IFN mediated transcription. One conserved site for NF-κB was found in the promoter region of Viperin. This is the first report of conservative binding motif for NF-κB in accordance with the consensus sequence (GGGRNNYYCC) among teleost ISG promoters. Moreover, there were also TATA, CAAT and Sp1 transcription factor sites in Viperin and ISG15 promoters. In 5′ untranslated region (UTR), snakehead ISG15 gene contains a single intron, which differs from Viperin gene. The transcripts of Vipeirn and ISG15 mRNA were mainly expressed in head kidney, posterior kidney, spleen and gill. The expression levels in liver were found to increase obviously in response to induction by IFN-inducer poly I∶C.

Time of impact of fragment P2 of comet SL 9 on Jupiter

FROM 1994-07-16 to 1994-07-22, 21 fragments of comet SL 9 impacted against Jupiter in succession. During the impact period the impact of fragments D, E, K, N, P2 and S was observed in china. We used a 1.56-m telescope with the CCD camera of Shanghai Observatory to monitor Jupiter and its Galilean Satellites, obtaining 1000 high-quality images at B, V, I wave bands. Our data on optical wave bands are rather complete and valuable in the world. Specially, we got some images of Jupiter, Io and Europa in the impact time of fragment P2 impacting against Jupiter. This will help us understand the precise time, position, depth and energy of the P2’s collision. In this note, we report the impact time of fragment P2 of comet SL 9 and Jupiter. According to the characteristics of water and erosion environments of different natural zones on the Loess Plateau, this paper studies changes of vegetation types, distribution boundaries of forest and grass as well as restoration capacity of vegetation in different natural zones in the middle Yellow River. The annual precipitation of 53. mm is the critical annual pre-cipitation for forest and grass distribution in the middle Yellow River. Among the zonal and azonal environmental factors affecting watershed sediment yield, the intermediate diameter D50 (mm) of suspended load and forest coverage (V, %) play the leading role. Of them the effect weight of forest coverage (V, %) on catchments sediment yield is only 3.4% less than the role of the intermediate diameter (D50, mm), they are almost the same. To effectively control soil erosion in semiarid, especially in hilly-gullied areas and make sediment transport modulus reduce to less than 6000 t/km2, it is rather difficult by merely relying on natural restoration of forest. In the process of cultivated land conversion into forest land and grassland, measures suiting local conditions should be adopted in tree species selection and artificial afforestation (grass planting) based on management with biological measures for slopeland and engineering measures for hilly-gullied areas, so that watershed forest coverage in key counties can reach at least over 3.%.Compared with the standard period of precipitation prior to the 1960s, with the decrease of an-nual precipitation at various periods, plant productivities decline to different degrees under natural conditions. The main reason accountable for the low survival rate of new seedlings and grass over years is due to precipitation decrease. In light with regression models of annual pre-cipitation and natural vegetation productivities, it is possible to obtain estimated values of wa-tershed natural vegetation productivity and eco-water consumption needed for the restoration to

Endosymbiotic dinoflagellates in the genus Symbiodinium are among the most abundant and important group of photosynthetic protists found in coral reef *** order to further characterize this diversity and compare with other regions of the Pacific,samples from 44 species of scleractinian corals representing 20 genera and 9 families,were collected from tropical reefs in southern Hainan Island,*** gradient gel electrophoresis fingerprinting of the ribosomal internal transcribed spacer 2 identified 11 genetically distinct Symbiodinium types that have been reported *** majority of reef-building coral species(88.6%)harbored only one subcladal type of symbiont,dominated by host-generalist C1 and C3.and was influenced little by the host’s apparent mode of symbiont *** species harbored more than one clade of Symbiodinium(clades C,D)*** geographically isolated from the rest of the Pacific,the symbiont diversity in southern Hainan Island was relatively low and similar to both the Great Barrier Reef and Hawaii symbiont assemblages(dominated by clade C Symbiodinium).These results indicate that a specialist symbiont is not a prerequisite for existence in remote and isolated areas,but additional work in other geographic regions is necessary to test this idea.

Characterization of a sodium-regulated glutaminase from cyanobacterium Synechocystis sp. PCC 6803

Glutaminase is widely distributed among microorganisms and mammals with important functions. Lit-tle is known regarding the biochemical properties and functions of the deamidating enzyme glutami-nase in cyanobacteria. In this study a putative glutaminase encoded by gene slr2079 in Synechocystis sp. PCC 6803.was investigated. The slr2079 was expressed as histidine-tagged fusion protein in Es-cherichia coli. The purified protein possessed glutaminase activity, validating the functional assign-ment of the genomic annotation. The apparent Km value of the recombinant protein for glutamine was 26.6 ± 0.9 mmol/L, which was comparable to that for some of other microbial glutaminases. Analysis of the purified protein revealed a two-fold increase in catalytic activity in the presence of 1 mol/L Na+. Moreover, the Km value was decreased to 12.2 ± 1.9 mmol/L in the presence of Na+. These data demon-strate that the recombinant protein Slr2079 is a glutaminase which is regulated by Na+ through in-creasing its affinity for substrate glutamine. The slr2079 gene was successfully disrupted in Synecho-cystis by targeted mutagenesis and the △slr2079 mutant strain was analyzed. No differences in cell growth and oxygen evolution rate were observed between △slr2079 and the wild type under standard growth conditions, demonstrating slr2079 is not essential in Synechocystis. Under high salt stress condition, however, △slr2079 cells grew 1.25-fold faster than wild-type cells. Moreover, the photosyn-thetic oxygen evolution rate of △slr2079 cells was higher than that of the wild-type. To further charac-terize this phenotype, a number of salt stress-related genes were analyzed by semi-quantitative RT-PCR. Expression of gdhB and prc was enhanced and expression of desD and guaA was repressed in △slr2079 compared to the wild type.
