Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

Aerobic granules cultivated and operated in continuous-flow bioreactor under particle-size selective pressure

A novel method based on the selective pressure of particle size （particle-size cultivation method, PSCM） was developed for the cultivation and operation of aerobic granular sludge in a continuous-flow reactor, and compared with the conventional method based on the selective pressure of settling velocity （settling-velocity cultivation method, SVCM）. Results indicated that aerobic granules could be cultivated in continuous operation mode by this developed method within 14 days. Although in the granulation process, under particle-size selective pressure, mixed liquor suspended solids （MLSS） in the reactor fluctuated greatly and filamentous bacteria dominated the sludge system during the initial operation days, no obvious difference in profile was found between the aerobic granules cultivated by PSCM and SVCM. Moreover, aerobic granules cultivated by PSCM presented larger diameter, lower water content and higher specific rates of nitrification, denitrifieation and phosphorus removal, but lower settling velocity. Under long term operation of more than 30 days, aerobic granules in the continuous-flow reactor could remain stable and obtain good chemical oxygen demand （COD）, NH4^＋-N, total nitrogen （TN） and total phosphorus （TP） removal. The results indicate that PSCM was dependent on the cultivation and maintenance of the stability of aerobic granules in continuous-flow bioreactors.

Multi-model ensemble deep learning method for intelligent fault diagnosis with high-dimensional samples

Deep learning has achieved much success in mechanical intelligent fault diagnosis in recent years. However, many deep learning methods cannot fully extract fault information to recognize mechanical health states when processing high-dimensional samples. Therefore, a multi-model ensemble deep learning method based on deep convolutional neural network (DCNN) is proposed in this study to accomplish fault recognition of high-dimensional samples. First, several 1D DCNN models with different activation functions are trained through dimension reduction learning to obtain different fault features from high-dimensional samples. Second, the obtained features are constructed into 2D images with multiple channels through a conversion method. The integrated 2D feature images can effectively represent the fault characteristic contained in raw high-dimension vibration signals. Lastly, a 2D DCNN model with multi-layer convolution and pooling is used to automatically learn features from the 2D images and identify the fault mode of the mechanical equipment by adopting a softmax classifier. The proposed method, which is validated using the bearing public dataset of Case Western Reserve University, USA and a one-stage reduction gearbox dataset, has high recognition accuracy. Compared with other classical deep learning methods, the proposed fault diagnosis method has considerable improvements.

The stability of underground entry-type excavations will directly affect the working environment and the safety of *** critical span graphs and traditional statistics learning methods can not meet the requirements of high accuracy for stability assessment of entry-type ***,this study proposes a new prediction method based on machine learning to scientifically adjust the critical span ***,the particle swarm optimization(PSO)algorithm is used to optimize the core parameters of the gradient boosting decision tree(GBDT),abbreviated as ***,the classification performance of eight other classifiers including GDBT,k-nearest neighbors(KNN),two kinds of support vector machines(SVM),Gaussian naive Bayes(GNB),logistic regression(LR)and linear discriminant analysis(LDA)are also applied to compare with the proposed *** revealed that compared with the other eight models,the prediction performance of PSO-GBDT is undoubtedly the most reliable,and its classification accuracy is up to ***,this model has great potential to provide a more scientific and accurate choice for the stability prediction of underground *** addition,each classification model is used to predict the stability category of several grid points divided by the critical span graph,and the updated critical span graph of each model is discussed in combination with previous *** results show that the PSO-GBDT model has the advantages of being scientific,accurate and efficient in updating the critical span graph,and its output decision boundary has strict theoretical support,which can help mine operators make favorable economic decisions.

Numerical study of a trapezoidal bypass dual throat nozzle

Bypass Dual Throat Nozzle(BDTN)is a novel type of fluidic thrust vectoring *** improve the infrared stealth performance of BDTN,a nozzle based on BDTN is proposed and numerically *** cross-section along the x-axis of the novel nozzle becomes a trapezoid,which is named“BDTN-TRA.”The main numerical simulation results show that BDTN-TRA can produce a thrust vectoring angle when the upper or lower bypass valve is *** angle difference between the two conditions mentioned above is usually approximately 1°-2°.Even if the two bypasses are closed,BDTN-TRA can produce a small thrust vectoring angle at around 3°-5°.When the sidewall angle increases from 60°to 90°,the thrust coefficient and thrust vectoring angle under each work condition usually decrease.A larger aspect ratio indicates better *** the aspect ratio increases over 7.2,the performance of BDTN-TRA is quite close to that of BDTN with rectangular cross-sections at the same aspect *** features will benefit the control and trimming for future aircraft design,especially for the flying wing layout *** but not least,BDTN-TRA has a more extraordinary mixing performance compared with *** distributions of static temperature and axial velocity along the x-axis of BDTN-TRA with sidewall angle of 60°decrease faster than those of *** the total temperature of the inlet equals 1600 K,the static temperature difference between BDTN-TRA with sidewall angles of 60°and 90°is over 360 K at twice the length of the nozzle downstream of the nozzle exit,which is the reflection for excellent infrared stealth for the fighter.

Robust ensemble of metamodels based on the hybrid error measure

Metamodels have been widely used as an alternative for expensive physical experiments or complex,time-consuming computational simulations to provide a fast but accurate ***,challenge remains in the prior determination of the most suitable metamodel for a particular case because of the lack of information about the actual behavior of a *** addition,existing studies on metamodels have largely restricted on solving deterministic problems(e.g.,data from finite element models),whereas some real-life engineering problems(e.g.,data from physical experiment)are stochastic problems with noisy *** this work,a robust ensemble of metamodels(EMs)is proposed by combining three regression stand-alone metamodels in a weighted sum *** weight factor is adaptively determined according to the hybrid error metric,which combines global and local error measures to improve the accuracy of the ***,three typical individual metamodels that can filter noise are selected to construct the EMs to extend their application in practical engineering *** well-known benchmark problems with different levels of noise and three engineering problems are used to verify the effectiveness of the proposed *** show that the proposed EMs have higher accuracy and robustness than the individual metamodels and other typical EMs in major cases.
