Tissue plasminogen activator is usually used for the treatment of acute ischemic stroke,but the role of endogenous tissue plasminogen activator in traumatic brain injury has been rarely reported.A rat model of traumatic brain injury was established by weight-drop *** tissue plasminogen activator inhibitor neuroserpin(5μL,0.25 mg/mL)was injected into the lateral *** function was assessed by neurological severity *** and axonal injuries were assessed by hematoxylin-eosin staining and Bielschowsky silver *** level of endogenous tissue plasminogen activator was analyzed by western blot *** marker cleaved caspase-3,neuronal marker neurofilament light chain,astrocyte marker glial fibrillary acidic protein and microglial marker Iba-1 were analyzed by immunohistochemical *** cell types were detected by immunofluorescence double *** cells in the damaged cortex were detected by terminal deoxynucleotidyl transferase-mediated digoxigenin-dUTP-biotin nick-end labeling *** neurons in the damaged cortex were detected by Fluoro-Jade B *** of tissue plasminogen activator was increased at 6 hours,and peaked at 3 days after traumatic brain *** apoptosis and axonal injury were detected after traumatic brain ***,neuroserpin enhanced neuronal apoptosis,neuronal injury and axonal injury,and activated microglia and *** further deteriorated neurobehavioral function in rats with traumatic brain *** findings confirm that inhibition of endogenous tissue plasminogen activator aggravates neuronal apoptosis and axonal injury after traumatic brain injury,and activates microglia and *** study was approved by the Biomedical Ethics Committee of Animal Experiments of Shaanxi Province of China in June 2015.

Dear Editor,Polycystic ovary syndrome(PCOS)affects more than 1 in 10 women worldwide.^(1)Despite its high prevalence,PCOS and its accompanying morbidities are likely underdiagnosed.^(2)To induce a PCOS-like phenotype in animals,testosterone and dihydrotestosterone are usually used,indicating that androgen excess is a major contributor to PCOS.^(3)Anti-M€ullerian hormone(AMH)is an upstream regulator of androgen and is well correlated with the androgens.

Implementing high-performance silicon(Si)anode in actual processing and application is highly desirable for next-generation,high-energy Li-ion ***,high content of inactive matrix(including conductive agent and binder)is often indispensable in order to ensure local conductivity and suppress pulverization tendency of Si particles,which thus cause great capacity loss based on the mass of whole ***,we designed an accordion-structured,high-performance electrode with high Si content up to 95%.Si nanoparticles were well anchored into the interlayer spacings of accordion-like graphene arrays,and free-standing electrode was prepared via a simple filtration process without any *** accordion framework ensures strong confinement effect of Si nanoparticles and also provides direct,non-tortuous channels for fast electrochemical reaction *** a consequence,the accordion Si electrodes exhibit ultrahigh,electrode-based capacities up to 3149 mAh g^(-1)(under Si content of 91%),as well as long-term ***,the accordion electrode can bear extreme condition of over-lithiation and maintains stable in full-cell *** design provides a significant stride in high Si content toward realistic,high-performance electrodes.

Effects of Oxalic Acid Addition on the Hydrothermal Synthesis of Kaolinite

Kaolinite was hydrothermally synthesized from alumina gel and silicate by dissolving alumina gel in oxalic acid before it was mixed with silicate, effects of the amount of addition on the species of synthetic products were discussed. The reaction product was characterized by X ray diffraction (XRD) and transmission electron microscopy (TEM). The results show that analcite is the only phase of the reaction solution without oxalic acid, the proportion of kaolinite in product increases with the amount of addition, and kaolinite is the main species when the molar ratio of oxalic acid to alumina reaches 0.6∶1.0. This is because oxalic acid addition is beneficial to the formation of kaolinite through changing the coordination number of aluminium from four to six, while the mixture of alumina gel, before it was dissolved in oxalic acid with silicate interfered with the crystallization of kaolinite.

Ligase-based ultrasensitive detection of DNAzyme cleavage product using molecular beacon

Detection for deoxyribozyme （DNAzyme） cleavage usually needs complex and time-consuming radial labeling, gel electro- phoresis and autoradiography. A new approach was reported for detection DNAzyme cleavage product based on molecular beacon （MB）. Part of the loop of MB was designed to complementary to DNAzyme cleavage product. MB was employed to monitor ligation process of RNA/DNA complex and to convert directly cleavage product information into fluorescence signal. Detection limit of the assay is 0.02 nmol/L. The cleavage product of 8-17 DNAzyme against HCV-RNA was detected perfectly based on this assay. The method is fast, simple and ultrasensitive, which might hold great promise in DNAzyme reaction and DNAzyme gene therapy.

BACKGROUND Early adenocarcinoma mixed with a neuroendocrine carcinoma(NEC)component arising in the gastroesophageal junctional(GEJ)region is rare and even rarer in young ***,we report such a case in a 29-year-old Chinese *** SUMMARY This patient presented to our hospital with a 3-mo history of dysphagia and *** endoscopy revealed an elevated nodule in the distal esophagus 1.6 cm above the GEJ line,without Barrett’s esophagus or involvement of the gastric *** nodule was completely resected by endoscopic submu-cosal dissection(ESD).Pathological examination confirmed diagnosis of intra-mucosal adenocarcinoma mixed with an NEC component,measuring 1.5 ***,both adenocarcinoma and NEC components were positive for P53 with a Ki67 index of 90%;NEC was positive for synaptophysin and ***-generation sequencing of 196 genes demonstrated a novel germline mutation of the ERCC3 gene in the DNA repair pathway and a germline mutation of the RNF43 gene,a common gastric cancer driver gene,in addition to pathogenic somatic mutations in P53 and CHEK2 *** patient was alive without evidence of the disease 36 mo after *** Early adenocarcinoma with an NEC component arising in the distal esophageal side of the GEJ region showed evidence of gastric origin.

Clinical characteristics and prognosis of young patients with colorectal cancer in Eastern China

AIM:To explore the clinical characteristics and prognosis of young patients with colorectal cancer patients in Eastern ***:A total of 1335 patients with colorectal cancer treated from December 1985 to December 2005at the Second Affiliated Hospital of Zhejiang University School of Medicine were studied *** patients were divided into two groups,a younger group(aged≤30 years)and an older group(aged>30 years),and comparison was made in the clinical characteristics and prognosis between the two *** test was used for data analysis of all categorical variables,and overall survival(OS)was calculated by the Kaplan-Meier method.A multivariate analysis was performed using the Cox ***:There were 42(3.1%)and 1293(96.9%)cases in the younger group and older group,*** analysis showed that the 5-and10-year OS in the younger group were 33.9%and26.1%,respectively,and those in the older group were60.1%and 52.2%,*** group had poor survival(χ2=14.146,P=0.000).Multivariate analysis revealed that age was not a dependent factor for prognosis(OR=0.866,95%CI:0.592-1.269,P=0.461).Stratified analysis indicated that in stageⅢandⅣdisease,the 5-and 10-year OS were 24.6%and14.8%in the younger group,and 40.4%and 33.3%in the older group,respectively,with a significant difference between the two groups(χ2=5.101,P=0.024).In the subgroup of radical surgery,the 5-and 10-year OS were 44.3%and 34.2%in the younger group,and69.6%and 60.5%in the older group,with a difference being significant between the two groups(χ2=7.830,P=0.005).CONCLUSION:Compared with older patients,the younger patients have lower survival,especially in the subgroups of stageⅢandⅣdisease and radical surgery.

The effects of heat treatment on tnicrostructure and hardness of laser surface-clad Ni2 !+20%WC+0.5%CeO2 on the heat-resistant cast iron were investigated by means of X-ray diffraction(XRD), transmission electron microscope(TEM) and microhardness test. The experimental results showed that heat-treating at 500’C has no effect on microstructure and hardness of the layers. Although the phase composition of the layers heat-treated at 700°C and 800°C remain unchanged, more Ni3B and N14B3 phases are precipitated on the matrix of the cladding layer, the metastable phase—M7C3 is transformed into steady phase—M23C6, and the precipitated phases coarsened.

Clinical characteristics and distribution of hepatitis B virus genotypes in Guangxi Zhuang population

AIM: To investigate the distribution of HBV genotypes and their YMDD mutations in Guangxi Zhuang population, China, and to study the relationship between HBV genotypes and clinical types of HB, ALT, HBV DNA, HBe system as well as the curative effect of Lamivudine (LAM) on hepatitis ***: A total of 156 cases were randomly chosen as study subjects from 317 patients with chronic hepatitis B (CHB). HBV genotypes were determined by PCR-microcosmic nucleic acid cross-ELISA. YMDD mutations were detected by microcosmic nucleic acid cross-nucleic acid quantitative determination. HBV DNA was detected by fluorescence ratio PCR *** was given to 81 cases and its curative effect was observed by measuring ALT, HBV DNA load, HBeAg, and HBeAg/HBeAb conversion ***: HBV genotypes B, C, D, and non-classified genotypes were found in Guangxi Zhuang *** for 25.6%, 47.4%, 58.3%, and 16.0%,respectively. Seventy-four cases were CD-, CB-, BD-mixed genotypes (47.7%). Forty-six (29.5%) cases had YMDD mutations. Genotype B was mostly found in mild and moderate CHB patients. Genotypes C, D and mixed genotype mostly occurred in severe CHB *** D and CD HBV-infected patients had higher ALT and HBV DNA than patients with other types of HBV infection. There was no significant difference among the genotypes in YMDD mutations, clinical types, ALT and HBV DNA level. Non-classified types geno had a significantly lower positive rate of HBeAg than other genotypes (x2= 12.841, P＜0.05). There was no significant difference in ALT recovery rate, HBV DNA load, HBeAg,and HBeAg/HBeAb conversion rate, 48 wk after LAM treatment between groups of genotypes D, CD, and nonclassified ***: Genotypes B, C, and D, non-classified and mixed genotype of HBV are identified in the Guangxi Zhuang population.
