The seabed instability induced by the transient liquefaction when exposed to wave-current may threaten the safety of offshore structures.In this study,the Reynolds-averaged Navier-Stokes(RANS)equations with the k-e turbulence model were used to imitate the fluid dynamics,and Biot's poro-elastic theory was used to simulate the transient seabed response.An in-house solver(porous-fluid-seabed-structure interactions-field operation and manipulation)integrating the flow model and seabed model with the finite volume method was developed.The present model was confirmed with published experimental results and then used to analyze the dynamic process of the fluid-seabed-structure interactions as well as seafloor liquefaction around the jacket foundation under wave-current loading.The simulated results showed that the depth and range for the liquefaction area around the jacket foundation tended to increase at first and then declined as the wave propagated forward in the absence of current.In addition,the results demonstrated that the liquefaction depth under current and wave in the same orientation was greater than that without current.It is worth mentioning that the downstream piles were more prone to liquefaction than the upstream piles when the forward current existed.

Objective: To study the regulatory effect and molecular mechanism of juglone on apoptosis of cervical cancer Hela cells. Methods: Cervical cancer Hela cells were cultured and treated with different dosages of juglone(10, 20, and 40 μmol/L, respectively) and c-Jun N-terminal kinase(JNK) inhibitor SP600125(10, 20, and 40 μmol/L, respectively). Then cellular proliferative activity and the expression of JNK/c-Jun pathway molecule and apoptotic molecule in the cells were detected. Results: After 6, 12, 18 and 24 h of treatment, the value for proliferative activity of cells treated with juglone was significantly lower than that of control group(P<0.05), and the anti-proliferative effect was more significant as the treatment period and juglone dosage increased(P<0.05). The protein expressions of Bax, Cyt C, Fas, Fas L, Caspase-3, p-JNK and p-c-Jun in cells treated with juglone were significantly higher than those of control group(P<0.05), and the protein expressions of Bax, Cyt C, Fas, Fas L, Caspase-3, p-JNK and p-c-Jun increased more remarkably as the juglone dosage increased(P<0.05). In cells treated with 40 μmol/L juglone and SP600125, the protein expressions of Bax, Cyt C, Fas, Fas L and Caspase-3 were significantly lower than those of cells treated with 40 μmol/L juglone(P<0.05), and the protein expressions of Bax, Cyt C, Fas, Fas L and Caspase-3 reduced more remarkably as the SP600125 dosage increased(P<0.05). Conclusion: Juglone can increase the expression of apoptotic molecules in mitochondrial pathway and death receptor pathway by activating JNK/c-Jun pathway, thus inducing apoptosis of cervical cancer cells.

Objective:To study the expression of E6 and E7 mRNA in high-risk human papillomavirus(HPV) HPV-18 and the relationship between the expression of invasive gene and cervical carcinoma.Methods:A total of 119 patients with cervical cancer,cervical erosion and cervical HPV infection who were diagnosed in our hospital were selected and randomly divided into two groups:cervical cancer group(n= 58) and non-cancerous group(n= 61).Another 60 patients with uterine leiomyoma were selected as normal control group.Detection of HPV18 E6,E7 mRNA expression and invasion,migration,proliferation inhibition genes,epithelial mesenchymal transition genes and proliferation related protein content.Results:The relative expression of E6 and E7 HPV-18 in cervical cancer group was significant higher than that in non-cancerous group and control group(mRNA)(P<0.05).The content of TRAF6 and c-FLIP in invasive cervical cancer group was significantly higher than that in non-cancerous group and control group(P<0.05).The mR NA content of CD44v6 and MMP-9 in cervical cancer group was significantly higher than that in non-cancerous group and control group(P<0.05).The content of DEC-1,IKK16,MBP-1 in cervical cancer group was significant lower than that in non-cancerous group and control group(P<0.05).The mR NA content of beta-catenin and Vimentin in cervical cancer group was significantly lower than that in non cancerous group and control group(P<0.05).The proliferation related protein E2F1 of cervical cancer group was significantly lower than that of non-cancerous group and control group,Bmi-1 content was significantly higher than non-cancerous group and control group(P<0.05).Conclusions:The expression of the detection of cervical cancer in high-risk human papilloma virus HPV-18 E6 and E7 mRNA,and the invasion,migration,proliferation inhibition gene,epithelial mesenchymal transition and proliferation related gene protein content,HPV expression rate of mR NA increased with the development of cervical cancer,the expression is also enhanced.The express

Background:China has experienced rapid urbanization in the past 30 years.We aimed to report blood cadmium level(BCL)in the rapidly urbanized Yangtze Plain of China,and explore the association between BCL and 25-hydroxyvitamin D(25(OH)D).Methods:Our data source was the Survey on Prevalence in East China for Metabolic Diseases and Risk Factors(SPECT-China)cross-sectional study(ChiCTR-ECS-14005052,***).We enrolled 3234 subjects from 12 villages in the Yangtze Plain.BCLs were measured by atomic absorption spectrometry.25(OH)D was measured with a chemiluminescence assay.Results:A total of 2560(79.2%)subjects were diagnosed with vitamin D deficiency.The median(interquartile range)BCL was 1.80μg/L(0.60-3.42)for men and 1.40μg/L(0.52-3.10)for women.In women,mean 25(OH)D concentrations were inversely associated with BCL(0.401,95%confidence interval:-0.697 to-0.105 nmol/L lower with each doubling of the BCL)after adjustment for age,educational status,current smoking,body mass index,diabetes,and season.However,there was no significant difference in 25(OH)D across the BCL tertiles for men.Conclusions:BCL in Chinese residents in the Yangtze Plain were much higher than that in developed countries.An inverse association between BCL and 25(OH)D was found in general Chinese women after multivariable adjustment.Future prospective cohort and animal studies are warranted to resolve the direction and temporality of these relationships,and to elucidate the exact mechanisms involved.

The shortage of groundwater resources is a considerable challenge for winter wheat production on the North China Plain.Water-saving technologies and procedures are thus urgently required.To determine the water-saving potential of using micro-sprinkling irrigation(MSI)for winter wheat production,field experiments were conducted from 2012 to 2015.Compared to traditional flooding irrigation(TFI),micro-sprinkling thrice with 90 mm water(MSI1)and micro-sprinkling four times with 120 mm water(MSI2)increased the water use efficiency by 22.5 and 16.2%,respectively,while reducing evapotranspiration by 17.6 and 10.8%.Regardless of the rainfall pattern,MSI(i.e.,MSI1 or MSI2)either stabilized or significantly increased the grain yield,while reducing irrigation water volumes by 20–40%,compared to TFI.Applying the same volumes of irrigation water,MSI(i.e.,MSI3,micro-sprinkling five times with 150 mm water)increased the grain yield and water use efficiency of winter wheat by 4.6 and 11.7%,respectively,compared to TFI.Because MSI could supply irrigation water more frequently in smaller amounts each time,it reduced soil layer compaction,and may have also resulted in a soil water deficit that promoted the spread of roots into the deep soil layer,which is beneficial to photosynthetic production in the critical period.In conclusion,MSI1 or MSI2 either stabilized or significantly increased grain yield while reducing irrigation water volumes by 20–40%compared to TFI,and should provide water-saving technological support in winter wheat production for smallholders on the North China Plain.

Background Dipeptidyl peptidase-lV （DPP-4） inhibitors are now used to improve postprandial glycemic control in type 2 diabetes. However, their effects on hepatic glucose production （HGP） in obesity are not clear. This study was designed to test the hypothesis that gluconeogenesis and HGP can be modulated by DPP-4 inhibitors in obesity. Methods Sprague Dawley male rats were divided into four groups, each on a different diet： general rat chow, n=10 （G）; G＋sitagliptin, n=10; high fat chow （obesity）, n=10 （55% fat calories, HFO）; HFO＋sitagliptin, n=10. After 10 weeks, the rats were fasted overnight and glucose metabolism was determined using 3-3H-glucose and 14C-glycerol as tracers. Results Glycerol rate of appearance （P 〈0.00001）, plasma glycerol （P 〈0.05） and free fatty acid （FFA）（P 〈0.05） concentrations, and HGP （P 〈0.05） were decreased in HFO＋sitagliptin group compared with HFO group, but there was no significant difference between G and G＋sitagliptin groups （P 〉0.05）. Gluconeogenesis in HFO group was five times of that in G rats （P 〈0.01）, but was significantly declined in HFO＋sitagliptin group （P 〈0.0001）. Conclusions Gluconeogenesis and HGP were inhibited by sitagliptin in high fat-induced obese rats due to decreased glycerol availability, which was a result of reduced glycerol release from adipose tissues. The finding suggests that sitagliptin is potentially useful for controlling fasting glucose in obesity, thereby delaying or preventing the development of diabetes.

Relationship between Free Thyroxine and Islet Beta-cell Function in Euthyroid Subjects

Thyroid hormones have a specific effect on glucose-induced insulin secretion from the pancreas.We aimed to investigate the association between euthyroid hormones and islet betacell function in general population and non-treated type 2 diabetes mellitus(T2DM)patients.A total of 5089 euthyroid participants(including 4601 general population and 488 non-treated T2DM patients)were identified from a cross-sectional survey on the prevalence of metabolic diseases and risk factors in East China from February 2014 to June 2016.Anthropometric indices,biochemical parameters,and thyroid hormones were measured.Compared with general population,non-treated T2DM patients exhibited higher total thyroxine(TT4)and free thyroxine(FT4)levels but lower ratio of free triiodothyronine(T3):T4(P<0.01).HOMA-βhad prominently negative correlation with FT4 and positive relationship with free T3:T4 in both groups even after adjusting for age,body mass index(BMI)and smoking.When analyzed by quartiles of FT4 or free T3:T4,there were significantly decreased trend of HOMA-β going with the higher FT4 and lower free T3:T4 in both groups.Linear regression analysis showed that FT4 but not FT3 and free T3:T4 was negatively associated with HOMA-β no matter in general population or T2DM patients,which was independent of age,BMI,smoking,hypertension and lipid profiles.FT4 is independently and negatively associated with islet beta-cell function in euthyroid subjects.Thyroid hormone even in reference range could play an important role in the function of pancreatic islets.

Background Acute pulmonary thromboembolism （APE） causes right ventricular dysfunction （RVD） and cardiac troponin I （cTnl） elevation. Patients with RVD and cTnl elevation have a worse prognosis. Thus, early detection of RVD and cTnl elevation is beneficial for risk stratification. In this study, we assessed 14-day adverse clinical events and combined RVD on transthoracic echocardiography （TTE） with cTnl in risk stratification among a broad spectrum of APE patients. Methods The prospective multi-centre trial included 90 patients with confirmed APE from 12 collaborating hospitals. Acute RVD on TTE was diagnosed in the presence of at least 2 of the following： right ventricular dilatation （without hypertrophy）, loss of inspiratory collapse of inferior vena cava （IVC）, right ventricular （RV） hypokinesis, tricuspid regurgitant jet velocity 〉2.8 m/s. The study patients were divided into two groups according to clinical and echocardiographic findings at presentation： Group Ⅰ： 50 patients with RVD; Group Ⅱ：40 patients without RVD. Results More than half of the patients （50/90, 55.6%） had RVD. Nearly one third （26/90, 28.9%） of patients had elevated cTnl at presentation and only 4.2% on the fourth day after initial therapy. A multiple Logistic regression model implied RVD, right and left ventricular end-diastolic diameter ratio （RVED/LVED）, and cTnl independently predict an adverse 14-day clinical outcome （P〈0.01）. Receiver operating characteristics （ROC） curves revealed that the cut-off values of RVED/LVED and cTnl yielding the highest discriminating power were 0.65 and 0.11 ng/ml, respectively. Furthermore, the incidence of an adverse 14-day clinical event in patients with RVD and elevated cTnl was greater （40.7%） than in patients with elevated cTnl or positive RVD alone （0% and 8.3%, respectively）（P〈0.001）. Conclusions RVD, RVED/LVED, and cTnl are independent predictors of 14-day clinical outcomes. In this study,we investigated the genetics,clinical features,and therapeutic approach of 14 patients with 5a-reductase deficiency in China.Genotyping analysis was performed by direct sequencing of PCR products of the steroid 5α-reductase type 2 gene(SRD5A2).The 5a-reductase activities of three novel mutations were investigated by mutagenesis and an in vitro transfection assay.Most patients presented with a microphallus,variable degrees of hypospadias,and cryptorchidism.Eight of 14 patients(57.1%)were initially reared as females and changed their social gender from female to male after puberty.Nine mutations were identified in the 14 patients.p.G203S,p.Q6X,and p.R227Q were the most prevalent mutations.Three mutations(p.K35N,p.H162P,and p.Y136X)have not been reported previously.The nonsense mutation p.Y136X abolished enzymatic activity,whereas p.K35N and p.H162P retained partial enzymatic activity.Topical administration of dihydrotestosterone during infancy or early childhood combined with hypospadia repair surgery had good therapeutic results.In conclusion,we expand the mutation profile of SRD5A2 in the Chinese population.A rational clinical approach to this disorder requires early and accurate diagnosis,especially genetic diagnosis.
