Crosstalk between RnA m^(6)A modification and epigenetic factors in plant gene regulation

n^(6)-methyladenosine(m^(6)A)is the most abundant modification observed in eukaryotic *** in transcriptome-wide m^(6)A mapping and sequencing technologies have enabled the identification of several conserved motifs in plants,including the RRACH(R=A/G and H=A/C/U)and UGUAW(W=U or A)***,the mechanisms underlying deposition of m^(6)A marks at specific positions in the conserved motifs of individual transcripts remain to be *** from plant and animal studies suggests that m^(6)A writer or eraser components are recruited to specific genomic loci through interactions with particular transcription factors,5-methylcytosine DnA methylation marks,and histone *** addition,recent studies in animal cells have shown that microRnAs play a role in depositing m^(6)A marks at specific sites in transcripts through a base-pairing mechanism.m^(6)A also affects the biogenesis and function of chromatin-associated regulatory RnAs and long noncoding *** we have less of an understanding of the link between m^(6)A modification and epigenetic factors in plants than in animals,recent progress in identifying the proteins that interact with m^(6)A writer or eraser components has provided insights into the crosstalk between m^(6)A modification and epigenetic factors,which plays a crucial role in transcript-specific methylation and regulation of m^(6)A in plants.

Iterative physical optics method based on efficient occlusion judgment with bounding volume hierarchy technology

This paper builds a binary tree for the target based on the bounding volume hierarchy technology,thereby achieving strict acceleration of the shadow judgment process and reducing the computational complexity from the original O(n^(3))to O(n^(2)logn).numerical results show that the proposed method is more efficient than the traditional *** is verified in multiple examples that the proposed method can complete the convergence of the ***,the proposed method avoids the error of judging the lit-shadow relationship based on the normal vector,which is beneficial to current iteration and *** with the brute force method,the current method can improve the simulation efficiency by 2 orders of *** proposed method is more suitable for scattering problems in electrically large cavities and complex scenarios.

Transforming nanoscale VO_(2)(B) into a scalable sodium-ion electrode

The intermittent nature of renewable energies requires highly reliable grid-level energy storage approaches.A critical consideration in developing this technology is the areal capacity which determines battery performance and influences the cost of battery *** related importance is finding new ways of developing scalable *** recent years,threedimensional(3D)printing of conductive scaffolds has emerged as an alternative to overcome the scalability limitations of commercial tape cast *** research carried out in the current study demonstrates a successful scalability pathway for nanoscale VO_(2)(B),a desirable cathode for sodium-ion batteries which has a nano-flower morphology with a crystallite sizeng VO_(2)(B)onto a graphene aerogel scaffold,we were able to achieve mass loading of over 100 mg·cm^(-2) and still possess an areal capacity of 10 mAh·cm^(-2) at a current density of 5 mA·cm^(-2).Moreover,after 1000 cycles,these electrodes retained 75% to 80% of their initial *** at high loading levels,the electrodeposited VO_(2)(B)exhibits pseudocapacitive material signatures such as a box-like voltammetry response,linear galvanostatic response,and no phase change upon *** scalability of the VO_(2)(B)electrode is demonstrated in a series of experiments which show the areal capacity to scale upon increase in both mass loading and electrode thickness,with only small changes in specific *** study establishes that nanoscale materials can be scaled up to achieve thick electrodes without compromising their electrochemical properties.

Enhancing the activity and succinyl-CoA specificity of 3-ketoacyl-CoA thiolase Tfu_0875 through rational binding pocket engineering

The 3-ketoacyl-CoA thiolase is the rate-limiting enzyme for linear dicarboxylic acids ***,the promiscuous substrate specificity and suboptimal catalytic performance have restricted its *** we present both biochemical and structural analyses of a high-efficiency 3-ketoacyl-CoA thiolase Tfu_***,Tfu_0875 displayed heightened activity and substrate specificity for succinyl-CoA,a key precursor in adipic acid *** enhance its performance,a deep learning approach(DLKcat)was employed to identify effective mutants,and a computational strategy,known as the greedy accumulated strategy for protein engineering(GRAPE),was used to accumulate these effective *** the mutants,Tfu_0875n249W/L163H/E217L exhibited the highest specific activity(320%of wild-type Tfu_0875),the greatest catalytic efficiency(kcat/KM=1.00 min-1mM-1),the highest succinyl-CoA specificity(KM=0.59 mM,28.1%of Tfu_0875)and dramatically reduced substrate binding energy(-30.25 kcal mol^(-1)v.s.-15.94 kcal mol^(-1)).A structural comparison between Tfu_0875n249W/L163H/E217L and the wild type Tfu_0875 revealed that the increased interaction between the enzyme and succinyl-CoA was the primary reason for the enhanced enzyme *** interaction facilitated rapid substrate anchoring and ***,a reduced binding pocket volume improved substrate specificity by enhancing the complementarity between the binding pocket and the substrate in stereo ***,our rationally designed mutant,Tfu_0875n249W/L163H/E217L,increased the adipic acid titer by 1.35-fold compared to the wild type Tfu_0875 in shake *** demonstrated enzymatic methods provide a promising enzyme variant for the adipic acid *** above effective substrate binding pocket engineering strategy can be beneficial for the production of other industrially competitive biobased chemicals when be applied to other thiolases.

Microplastics may reduce the fish bioaccumulation of organic pollutants

Microplastics have frequently been regarded as a threat to aquatic organisms due to their potential to enhance the accessibility of organic pollutants. However, studies of microplastics as “carriers” in the process of bioaccumulation have only been conducted on a few organic contaminants. In this study, we employed machine learning algorithms to predict the sorption capacity of 83 exogenous organic pollutants(in CCL4) on microplastics, the prediction model exhibits exceptional predictive performance,achieving R2values of 0.8651 and 0.6962, RMSE values of 0.4625 and 0.1841 for the sorption coefficients Kfand n, respectively,and quantitatively estimated the contribution of microplastics to the bioaccumulation levels of organic pollutants in Gadus morhua. By considering both natural and microplastic-ingesting pathways, we demonstrated that the ingestion of microplastics containing organic pollutants facilitates the entry of organic pollutants into Gadus morhua. However, due to their robust sorption capacity, microplastics transport the organic pollutants accumulated through the natural pathway out of the fish, thereby preventing bioaccumulation. These findings elucidate the “cleaning” mechanism of microplastics concerning various organic pollutants in aquatic organisms, providing valuable insights for accurately assessing the risk of microplastics in aquatic environments.

Accelerating the Screening of Modified MA_(2)Z_(4)Catalysts for Hydrogen Evolution Reaction by Deep Learning-Based Local Geometric Analysis

Machine learning(ML)integrated with density functional theory(DFT)calculations have recently been used to accelerate the design and discovery of single-atom catalysts(SACs)by establishing deep structure–activity *** traditional ML models are always difficult to identify the structural differences among the single-atom systems with different modification methods,leading to the limitation of the potential application *** to the structural properties of several typical two-dimensional MA_(2)Z_(4)-based single-atom systems(bare MA_(2)Z_(4)and metal single-atom doped/supported MA_(2)Z_(4)),an improved crystal graph convolutional neural network(CGCnn)classification model was employed,instead of the traditional machine learning regression model,to address the challenge of incompatibility in the studied *** CGCnn model was optimized using crystal graph representation in which the geometric configuration was divided into active layer,surface layer,and bulk layer(ASB-GCnn).Through ML and DFT calculations,five potential single-atom hydrogen evolution reaction(HER)catalysts were screened from chemical space of 600 MA_(2)Z_(4)-based materials,especially V_(1)/HfSn_(2)n_(4)(S)with high stability and activity(Δ_(GH*)is 0.06 eV).Further projected density of states(pDOS)analysis in combination with the wave function analysis of the SAC-H bond revealed that the SAC-dz^(2)orbital coincided with the H-s orbital around the energy level of−2.50 eV,and orbital analysis confirmed the formation ofσ*** study provides an efficient multistep screening design framework of metal single-atom catalyst for HER systems with similar two-dimensional supports but different geometric configurations.

In water, n,n′-diarylpyridinium thiazolo[5,4-d]thiazole guests(G1–G3) were encapsulated by cucurbit[8]uril(Q[8]) to construct Q[8]_(3)/G_(2) complexes, in which the encapsulated G molecules exist as an exact face-to-face dimer. These complexation behaviors have been indicated by proton nuclear magnetic resonance(~1H nMR) spectroscopy and isothermal titration calorimetry(ITC),and have been confirmed by X-ray single crystal structural analysis of Q[8]_(3)/G2_(2). While free guests G1–G3 have no ability to sensitize singlet oxygen, they all become effective singlet oxygen sensitizers when complexed with Q[8]. By irradiation of white light, Q[8]_(3)/G_(2) complexes induced selective oxidation of aryl sulfide to aryl sulfoxide in good yields and selective oxidation of dimethyl sulfide(DMS) to dimethyl sulfoxide(DMSO) in excellent yields without over-oxidation to dimethyl sulfone(DMSO_(2))even for elongated irradiation. High-yield photo-oxidation of DMS to DMSO_(2) was only achieved by irradiation of blue light in the presence of Q[8]_(3)/G1_(2), which was also effective in photobleaching of Rhodamine B in water. The higher photo-oxidation power of Q[8]_(3)/G1_(2) has been ascribed to an additional generation of reactive oxygen species besides singlet oxygen.

Experimental and first-principles calculation of Tin growth mechanism on CeAlO_(3) surface in steel

Regular Tin is harmful to the toughness of steel,and its shape and size need to be *** the behavior of Tin precipitation on CeAlO_(3) surfaces is critical for controlling the morphology and formation process of CeAlO_(3)–Tin composite inclusions in the *** results showed that Tin had a square morphology on the CeAlO3 surface,and electron backscatter diffraction phase identification results revealed the orientation relationship between CeAlO_(3) and Tin as follows:(001)_(CeAlO_(3))//(110)_(Tin),(100)_(CeAlO_(3))/(001)_(Tin),and[010]_(CeAlO_(3))/[110]_(Tin).The CeAlO3 crystal structure was studied using the first-principles calculation method,and the adsorption and growth process of Tin on the CeAlO_(3) surface were investigated from the atomic *** calculation results indicate that there is no metallic bonding present in the CeAlO_(3) *** the low-index crystal planes of CeAlO_(3),the(110)planes terminated with O and CeAlO have the highest and lowest surface energies,respectively,with values of 0.373 and 0.051 eV/Å^(2).On the high surface energy plane of CeAlO_(3),the Tin atomic permutation structure is consistent with the arrangement of Ti and n atoms in Tin(100)or(110).For the low surface energy plane of CeAlO_(3),the Ti and n atoms are arranged in a ring-like structure.
